Phytochemical Test for Tannins

Use
• Tannin is one of the major active ingredients found in plant
based medicines; it serves as caustics for cationic dyes
(tannin dyes) used in the dyestuff industry, and also in the
production of inks (iron gallate ink), textile dyes, antioxidants
in beverages, and coagulant in rubber production, as well as
possessing antiviral, antibacterial, and antitumor activity.
Tannin has been reported to selectively inhibit HIV
replication. Other uses of tannin are for wine, fruit juice, and
beer clarification in food industries.
Test for Tannins

Brief procedure
(ex. 2mL aq. extract + 2mL 1% HCL (boiled))

10 mL sample extract + evaporate over a steam bath + 20 mL


of distilled water + 5 drops of 10% sodium chloride solution.
A. Gelatin test
Brief procedure
(ex. 2mL aq. extract + 2mL 1% HCL (boiled))

1 portion of the filtrate of the sample extract + 3


drops 1% gelatin solution + 3 drops 10% NaCl
B. Ferric chloride test
Brief procedure
(ex. 2mL aq. extract + 2mL 1% HCL (boiled))

1 portion of the filtrate of the sample extract + 3


drops ferric chloride + tannic acid solution
