:Mechanism of Action

 It is an antihistamine drug that causes allergy symptoms such as sneezing,


runny nose, itching and watery eyes. Diphenhydramine inhibits the binding of
histamine to H1 receptors, which reduces symptoms.

 Reduces histamine-related vasodilation and increases capillary permeability.

 It is well absorbed after oral administration and is widely distributed


throughout the body, including the central nervous system.

 It is metabolized in the liver and largely excreted in the urine as metabolites


:Medicinal uses

 This medicine is used by mouth: To treat and relieve symptoms of allergic


reactions caused by histamine release, including nasal allergies and colds,
such as itching, rash, runny nose, itchy throat, watery eyes, and sneezing. It
is also used to treat vomiting, nausea, and dizziness caused by motion
sickness. To aid relaxation and sleep, this medicine is used topically: to
relieve pain and itching associated with insect bites, minor cuts and sunburn,
or due to rashes caused by poison ivy, poison oak and poison sumac.
:Contraindications and side effects

 Diphenhydramine is contraindicated in people who are allergic to diphenhydramine, or any of


the substances used in the compound. And people who suffer from breathing problems such as
asthma attacks
:Pharmacological forms

 Diphenhydramine is available in the following dosage forms:

 Capsules: 25 mg, 50 mg.


 Tablets: 12.5 mg, 25 mg, 50 mg.
 Chewable tablets: 12.5 mg.
 Solution for intravenous or intramuscular injection: 50 mg/ml.
 Syrup, oral solution elixir: 12.5 mg/5 ml.
 Cream, gel 50 mg.
