Treatment Drugs

1. Reversible anti-cholinesterases (Mild and Moderate forms of AD


Ex. Tacrine
-Orally active
-provides improvement in memory cognition and general well being
-It also facililitates the release of Ach fro nerve endings
Adverse effects
hepatotoxicity,- use was restricted

2. Newer Reversible antcholinesterase inhibitors


Ex:Donepezil, (5mg once /day)
rivastigmine, (1.5mg orally BD)
galantamine (4mg orally BD)

Adverse effects
Peripheral cholinergic side effects (Diarrhoea, nausea and vomiting)
3. Structural analouges of Ach
Ex: Acetyl –L-carnitine
4. Non –competitive antagonist of NMDA receptors
Ex Mimantine

 Some research studies showed that Rosiglitazone, statins, Ibuprofen shows beneficial effect in
 Vit A, C antioxidants, zinc, selenium biflavanoids significantly reduces the progression of AD.
Addiction
Intense craving to procure the drug by any means
Development of tolerance(Need more dos go get the same rewarding experience)
Lifethreatening withdrawl effects

Habituation
No intense craving for seeking
No development of tolerance
No distressful withdrawl effect
1. Short term substitution of the abused drugs
2. Aversion therapy
3. Use of proper antagonists to prevent relapse
