Van den Bergh reaction
 Van den Bergh reaction is a chemical reaction used to measure bilirubin levels
in blood

 It determines the amount of conjugated bilirubin in the blood and is highly


useful in understanding the nature of jaundice.

 Van den bergh reagent is mixture of equal volumes of sulfanilic acid (in dilute
HCL) and sodium nitrite.

 Priniciple: Diazotised sulfanilic acid reacts with bilirubin to form purple


colored azobilirubin
 Direct Positive: Conjugated bilirubin produces color as it is soluble in reagent.

 Indirect Positive: Unconjugated bilirubin do not produce color directly as it is


insoluble but it gives color when mixed with methanol.

 If both conjugated and unconjugated bilirubin are present the reaction is termed a
biphasic reaction.
