Effect of sugar in

Aspergillus nigar
EXP aim : To determine the affect of different type of sugar in
Aspergillus nigar growth
Introduction
in this experiment we get Aspergillus niger and we cultivate it
in mendal medium which offer good condition to it
Then supplement the medium with different types of sugar in
different flask to see the affects of these sugar in growth of
Aspergillus nigar
These sugars are (xylose – glucose – maltose – fructose) .
Material
• Shaking incubator
• Flask with cotton plug
• Hemocytometer
• Distiller
• Analytical parlance
Procedure
1. we get asparagillus nigar and calculate its concentration using
hemocytometer
2. Prepare mendal medium
3. Inoculmn 1*10^7 spore / ml in mendal medium in flask with cotton plug
4. Add maltose to the medium
5. Incubate the flask in shaking incubator
Discussion
Aspergillus niger typically thrives in aerobic conditions with a pH
range of 2 to 7.5. The optimal temperature for growth is around 30-
37°C. It's known for its ability to grow on a variety of substrates,
especially those rich in carbon sources, such as sugars So We
prepared mandel's medium is culture medium commonly is a
complex of nitrogenous sources that include cellulase secretion
that used to growth Aspergillus Niger
The composition of the medium is
●1.4g (NH4)2SO4 used as pH regulation/differential medium/osmotic regulation
●2g KH2po4 as ph regulation/enhancer
●0.3g CaCl2.2H2O as regulation of membrane permeability/signal transduction
●0.3g MgSo4.7H2O as osmotic balance/buffering capacity ●0.005g FeSo4.7H2O as
regulation of gene expression
●0.0016g MnSo4.7H2O as metabolic process /regulation of gene expression ●0.0014g
ZnSo4.7H2O as essential nutrients/ co_factor /regulation role
●0.002g CoCl2.7H2O as vitamin B12 synthesis/regulation role
●0.75g Peptone as nutrient source/enhanced growth and biomass products
●0.3g urea as nitrogen source/differential medium
●1g Tween80 as emulsifier/ enhancer / oxygen transfer/surfactants stabilizer/solubilizer and
disperant

Cotton plug is used for air exchange the incubator is shaking to be


sure the liquid is homogeneous and the the humidity is perfect for
fungus growth
all flask have the same components the only difference of them is
the type of sugar we add to see which sugars have the greatest
affect of fungus growth so after on week of incubation we will get
sample of each flask and calculate its concentration
