Factors Affecting Enzyme Activity

Introduction
This experiment was conducted about the enzyme urease that
break down the hydrolysis of urea to carbon dioxide and ammonia
that has a high pH which can be used to be pH indicator that was
red cabbage extract. The experiment was trying to find how pH
influence the color, the effect of urease from soya beans on urea,
and effect of pH on urease.
The expectation of this experiment was if the substance was
acid, the color will change to be red tone and if the substance was
base, the color will change to be blue tone. The color will be purple,
if the substance was neutral. And also the pH number will follow
the thing that we add. The color in tube that add soya beans will
change faster. Lastly, the proper pH will react well on urease.

Procedure 1



Why is it important to have the three reference tubes?
- It is important to have three reference tubes because it will test
that inhibitor work properly and how the color change when react with
acid, base, and neutral.
Can you predict what color of the solution will be if the urease enzyme
convert urea to ammonia.
- The color will change to be blue because ammonia is base.
Procedure 2




What are the purposes of procedure 2?
- The purposes of the procedure is to test that enzyme work
properly or not. The results can tell us that the tube that add enzyme,
the color will change faster and the tube that add water should not
change.
Why do we need to add an enzyme into a test tube without urea even
though we can predict that no enzyme activity will occur?
- We need to add enzyme into a test tube because to test that
enzyme does not be base or acid.

How does pH affect the urease activity? How do you know?
- If the proper pH affect the urease activity, it will react well
and can see the changes but if it is the unsuitable pH, it will
change the shape and cant react

From your results, what pH would you use and avoid if you
have to conduct an experiment using urease enzyme and why?
- The pH that we would use if we have to conduct an
experiment using urease enzyme was base and avoid acid
because urease would react well when the pH was base.
Because basic pH will also change the color of the indicator to
blue/green which is very similar when ammonia has been
produced by the urease enzyme, how can we make sure that the
color change is actually due to the urease activity? Is it possible
to test this?
- We can make sure that the color change is actually due to
urease activity by adding acid into indicator and then adding
urease to see that it will return to be neutral or not.

Conclusion
The things that our group have learned from this experiment
was how the inhibitor works, enzyme is neutral that if we add it
into the tube, the color will change faster, the ammonia is base,
urease will react well if pH is base.
The prediction and the result from this experiment are also
the same that the color changes and pH number same as our
expectation. For example, in procedure 1, the tube that is added
critic acid that is acid, the color change to be red. The tube that is
added sodium bicarbonate that is base, the color change to be
blue. The tube that is added water, the color is not change. In
procedure 3, the result of color and pH number follow the thing
that we add.

For the error in this experiment, the pH number of the test
tube 4 in the procedure 3 after adding 2ml of soya beans is less
than the test tube 3 that adding less sodium bicarbonate or
baking soda. The pH number of the test tube 4 is 7.3 but the test
tube 3 is 7.9. The ways that we can improve in the next time are
carefully adding the solution and also using the device that is
more accurate.
