Exploration Homework: Week 8 This weeks Exploration Homework is designed to help you clarify your knowledge of the mitotic checkpoint signal and the APC. Use the discussion from class, the SciLit reading from Week 7 and any additional sources to answer the following questions. Be sure to read the complete question and answer all parts. Answers should be in full, coherent sentences 1. In your own words, describe what the anaphase-promoting complex does in the cell and how and when it is activated. Use three sentences or less. Key Points: The anaphase-promoting complex is a complex of proteins that attaches ubiquitin to proteins (mainly securing & cyclins) to destroy them. The APC is activated in metaphase when Cdc20 binds to it. This will occur once all the kinetochores are attached & the mitotic checkpoint signal is turned off. 2. In your own words, describe what the metaphase (mitotic) checkpoint signal actually is, when and how its produced and how the signal affects chromosomes? Why is the signal important? Use one or two sentences for each answer. Key points: WHAT IS THE SIGNAL: The signal is a mechanism used for regulation during mitosis to prevent the cell from dividing if there are any unattached chromosomes. WHEN IS IT PRODUCED: The signal is turned on if there are any unattached chromosomes along the metaphase plate. HOW IS IT PRODUCED: The signal is produced by using APC/Cdc20 inhibitors to inhibit APC & keep the cell in metaphase so that it doesnt divide. HOW IT ALTERS MITOSIS: If the signal is turned on, then the cell will stay in metaphase until the chromosomes are all attached before moving on to anaphase. WHY IS IT IMPORTANT:
If there wasnt a signal to prevent unattached chromosomes from dividing, then
aneuploidy can occur which may lead to tumorigenesis.
