TITLE: MEDIA AND INOCULUM PREPARATION

OBJECTIVE

1) To prepare inoculum using the yeast, Saccharomyces cerevisiae


2) To prepare media using the YPD Broth
3) To analyse fermentation data

INTRODUCTION

The yeast Saccharomyces cerevisiae is a single-celled microorganism that is capable of rapid division on
defined medium. In this experiment, Saccharomyces cerevisiae was the inoculum that was used.
Inoculum is a small amount of substance containing organisms from a pure culture which is used to start
a new culture or to infect an experimental animal. Generally, there are two types of media which are
cell culture and microbiological culture. Microbiological culture is generally used for growing
microorganisms such as bacteria or yeast.

PROCEDURE

MEDIA PREPARATION

5 g of YPD powder was weighed and inserted into the Schott Bottle

The powder was diluted with distilled water

5 g of sucrose was added into the solution

Distilled water was added until it reached 500 ml


INOCULUM PREPARATION

5 g of Saccharomyces cerevisiae was weighed and added into the media prepared

CONCLUSION
As a conclusion, we were able to prepare media and inoculum for the cultivation of yeast. General
media such as the YPD broth is suitable for the growth of Saccharomyces cerevisiae. Based on the result,
we were able to see that there was an increase in the growth of the yeast, Saccharomyces cerevisiae.
The irregular reading of the glucose level could indicate that there were other factors that contributes to
disturbs the reading.
