Determination of Acidogenic
Bacteria Isolate which is able to Produce the Highest Titration Acidity Total

SUMMARY

Tofu wastewater is able to cause a serious contamination problem


because ot the high organic pollutant content. One of the wastewater treatment
can be applied in tofu industry is an anaerobical biology process. This process
has four main stages: hydrolysis, acidogenesis, acetogenesis, and
methanogenesis. Each step involves different bacterial groups. Acidogenesis
stage plays an important role in anaerobical decomposition of tofu wastewater,
because asidogenesis product of acetic acid is a primary substrate for
methanogenic bacteria to produce methane. Acidogenesis stage is performed by
various groups of bacteria, the majority are an obligate aerobic bacteria and the
others are facultative anaerobic bacteria. However, it has not been yet known a
dominant acidogenic bacteria produced the highest organic acid.
This research had an objective to find an acidogenic bacteria to produce
the highest organic acid from asidogenesis stage of tofu wastewater
decomposition. Implementation of the study included sampling, making
anaerobical system by in vitro, isolation and purification acidogenic bacteria,
isolate selection by TSIA, making inoculum, test of acidogenic capability, data
analysis, morphological (colony and cell) and biochemical identification,
conclusion and suggestion. The experimental design used was Randomized
Block Design arranged in a factorial. Treatments given were inoculum
percentage (A): A1=5 %, A2=10 %, and A3=15% and incubation time (B): B1=24
hours, B2=48 hours, and B3=72 hours. Each treatment combination was
repeated three times. Parameters measured were production of titration acidity
total, pH, degree of sugar reduction, and acidogenic bacteria total.
One of pure isolate coded 1031 was known its capability of glucose
fermentation as a resulted of TSIA selection. Identification of colony morphology
of selected isolate were bone-white, round shape, raised surface (elevation), flat
edge and cell morphology were Gram-negative bacillus-shaped. Based on the
biochemical identification showed selected isolate was Hafnia alvei biogp 1 with
a probability value of 94.53%. Result showed inoculum percentage and
incubation time has significant effect in production of titration acidity total. The
best treatment combination was A3B1 (inoculum percentage of 15 % and
incubation time in 24 hours). A3B1 treatment produced titration acidity total 2,70
%, pH 6,33, degree of sugar reduction 0,07 %, and acidogenic bacteria total
1,1x108 CFU/ml. Further study by giving various sugar as substrate needed in
order to produce organic acid optimally.
