I/ INTRODUCTION

Brief introduction about the experiment


Counter-current immunoelectrophoresis depends on movement of antigen towards the anode
and of antibody towards the cathode through the agar under electric field. The migration of
antigen and antibody is greatly facilitated under electric field, and create the line of precipitation.
Rocket electrophoresis is the test in which having the appearance of the precipitin bands in the
shape of rocket appearance at the end of the reaction. In this method, antibody is incorporated
in the gel and antigen is placed in wells cut in the gel.electric current is then passed through the
gel, which facilitates the migration of antigen into the agar.
Objectives of this labwork experiment
- qualify the antigen being tested
- estimation of antigen concentration
Other techniques used for the same reason.
Precipitation in solution: ring test and flocculation test
Precipitation in agar: radial immunodiffusion and double diffusion
1/Compare your experiment with other groups’ experiments, give comments.
The results of our group is different with other. There is only one line of precipitate between
control antibody with specific antigen while in group 1, there are 2 lines between the control and
sample antibody with same antigen. This is because in my group, the antibody sample is not
specific for the antigen as two of antigen in our experiment is different with each other.
