1 Purpose
This describes the processes used to manage chromatographic hardware on the Empower
Chromatography Data System(CDS).

2 Scope
This document covers the use of Empower CDS at any laboratory where the corporate
Empower implementation has been adopted.
The covers the addition, modification and removal of LAC/E acquisition servers and
chromatographic hardware in the Empower CDS.
2.1 Definitions
Term/Acronym Definition
CDS Chromatography Data System. A computer application which acquires,
processes and reports data from chromatographic systems. Example:
Empower, supplied by Waters.
LAC/E Laboratory Acquisition and Control/Environment. The Waters acquisition
server. A computer which is connected to the network and acquires data
from chromatographic systems. The LAC/E may also control the system.
In the event of network failure, the LAC/E will buffer any data currently
being acquired.
Node An item on a network. For Empower a LAC/E is a node.
Sat/In Satellite Interface device. An analogue to digital converter which acquires
data from chromatographic systems. The Sat/In communicates with the
instrument via contact closure event signals. The Sat/In is connected
between the chromatographic system and the LAC/E.
Instrument Chromatographic hardware unit connected to Empower as a single entity.
An instrument may consist of a complete chromatographic system
(e.g.an Agilent 1100), a separate module (e.g. a detector) or a Sat/In.
System A collection of one or more instruments or modules which constitute a
complete chromatograph.
LAN Local Area Network
MAC Media Access Control
NIC Network Interface Card
Ethernet LAN technology used to connect computers to each other. LAC/E servers
are connected to the GSK LAN via a NIC. Ethernet connections may also
be used to connect instruments to a LAC/E.
IP Address Internet Protocol address. A unique number which identifies each
computer in a network. IP addresses may be static (never change) or
dynamic (assigned each time a computer is connected to the network).
DHCP Dynamic Host Configuration Protocol. A protocol which automatically
assigns IP addresses to computers using a DHCP server.
Crossover Cable An interface cable used to connect one computer directly to another or an
instrument directly to an instrument network card.
Network Switch A device that connects multiple devices (i.e. computers) to a LAN.
busLac/E Instrument interface used to connect to multiple Waters systems.
(Contains an IEEE488 connection - also known as GPIB or HPIB).
RJ45, DB9 Connectors used to terminate connections between computers.

Peak simulator A device which generates an analogue output similar to the output of a
chromatograph’s detector. May also be used to check analogue to digital
converters.

Avocent Hub A device which converts the serial connection to Ethernet Connection.

3 Instructions
3.1 Process Flow
A process flow diagram for the connection of new acquisition servers and chromatographic
hardware can be found in the below diagram.

3.2 Manufacturers and Models of Chromatography systems


Empower can connect and control different makes of Chromatography system.
Waters: W2690/5 Alliance Separation Module, 2487 UV detector, 2996 PDA detector, e2690/5
Alliance Separation Module, 2489 UV detector, 2998 PDA detector, 2475 FLR detector, Acquity
UPLC
Agilent: A1100 HPLC, A1200 HPLC, A1200RRLC, A1220 HPLC, A1260 UPLC, A1290 UPLC,
A 6890/6890+/6890N GC, A7890A/B GC, 7694 HS, G1888 HS, 7697 HS.
Shimadzu: Shimadzu Prominence, LC-2010A/C, LC-2010AHT/CHT, GC-2010, GC-2010 Plus,
GC-2014.
PerkinElmer: Clarus GC 400, 480, 500, 580, 600, 680

Other makes are Beckman Coulter, CTC or DaVinci, DANI Instruments, Distek, Erweka,
Hanson, Hitachi, Sotax, Varian

3.3 General
Instruments are connected to Empower via Three kinds of interface devices.
- busLac/E,
- Ethernet and
- Serial – can be done using RS-232 cable or Avocent hub or Edge Port Cable or USB.
A LAC/E acquisition server is a dedicated PC, running Empower software and instrument
drivers which contains interface cards. The LAC/E, which connects to the Corporate network,
transfers CDS data to and from the CDS application server. The LAC/E contains a hard drive so
that if the connection to the server is lost, any sample set which has already started will continue
to acquire data. The data will be buffered in LAC/E until the server connection is restored.
In general Waters chromatographs should be connected to the IEEE488 connection on a bus
LAC/E card. Agilent 1100 and 1200 systems controlled by Empower are connected via the LAN
card. Agilent 6890 systems are connected via the Equinox card. All other systems which are not
controlled by Empower are connected via the LAN card using satellite analogue to digital
interfaces (Sat/Ins).
There are several connection combinations available. The configurations described in this
document are recommended, although it is possible to use other, fully documented, controlled
configurations where required. All systems which cannot be controlled by Empower should be
connected to Sat/In devices and controlled outside Empower. It is acceptable to use the
configurations described in combination where a chromatograph is a combination of different
modules.
All Empower transactions described in this GSOP are accessed from the configuration manager
which can be accessed by selecting the configure system button from the main Empower pro
interface or by selecting Manage>Configuration in the Quickstart interface.
Whenever prompted by Empower the user must add an audit comment detailing the reason for
the change. The comment may be selected from the menu provided or entered as free text.

3.4 Configure of New LAC/E in Empower


Request Local IT to Provide the LAC/E with required interface card, then locate it to a
convenient location at site, connect to the site LAN and power up.
In configuration manager select File > New > Empower Node. Enter the name of the LAC/E and
the local time zone in the resulting dialogue box. Empower will connect to the LAC/E.
The new LAC/E will appear in the acquisition server list.
Select the new LAC/E from the acquisition server list and select File > Properties.
The resulting dialogue has the following tabs:
- General – enter a comment giving the location of the LAC/E
- Instruments – if instruments are already connected to the LAC/E they will appear here.
- If instruments are missing from the list, ensure they are powered up and select ‘Scan
Instruments’.
- Serial Ports – Gives information on instruments connected to the serial ports on the
LAC/E.
- Configure DHCP – Gives information on devices connected to the instrument LAN card.
- Access – controls user access to instruments on the LAC/E. Check the Share
instruments box and Owner and Groups option. Select the user groups who require
access. Do not check the password required box.
3.5 Prerequisite - For Systems Connected by Ethernet
3.5.1 IP address Configuration
- For every LAC/E with at least one system connected by Ethernet, beforehand, the LAC/E
must be configured.
- In the Configuration Manager window, in the Nodes list, select the LAC/E, right-click -
Properties
- Go to Configure DHCP tab, then click on Configure DHCP. Enter a comment in the audit trail
window, and click OK.
- In the Waters DHCP Server Configuration window, select Server - Configuration Wizard

- Select the Instrument LAN (Secondary NIC card used to control the systems), And, click on
Next. (In the below example, two additional Ethernet cards are available). Select the

Custom, Set IP address as 192.168.0.1 and, click on Next

- Click on Finish. The lace will reboot. Check beforehand that no other system connected to
the lace is currently used.

3.5.2 IP Address description

3.5.3 Connection of several systems by Ethernet


3.5.3.1 One Ethernet card is Available for each system
- Each Ethernet card must have an IP address with a specific range. As in example above where two
Ethernet cards are available: LAN 1 in range 0 and LAN 2 in range 2.
- IP address of the system connected to LAN 1 Ethernet card must have the same IP range as LAN 1
card.
- IP address of the system connected to LAN 2 Ethernet card must have the same IP range as LAN 2
card.

3.5.3.2 Only one Ethernet card is available for each LAC/E


- Each system is connected to Ethernet card via a network switch.
- In this case, as same as previously, systems and Ethernet card connected have the same IP range.

3.6 Connect Waters System (IEEE488)


Requirements:
- Waters LAC/E configured on Empower with busLac/E interface card installed.
- IEEE488 cables.

- Ensure all the modules are powered OFF.
- Arrange the LAC/E and the HPLC hardware in a convenient location. Use an IEEE488
connector to connect the first module to the LAC/E box. Connect each further module in
a ‘daisy chain’ using the stacking connectors.
- Each module connected to a given LAC/E must have a unique IEEE488 address (Refer
to the instrument manuals to set these addresses).
- Power ON All modules.
- Empower will automatically detect the instruments. Go to the LAC/E properties, and to
the Instruments tab and click Scan instruments.
- The Empower instrument address is in the format W<Model>@ n where <Model> is the
model number (i.e. 2690/5, 2487, 2996) and n is the IEEE488 address.

3.7 Connect Waters System (Ethernet)


3.7.1 Waters E2695 Alliance with 2998 PDA or 2489 UV or 2475 FLR detector
Requirements:
- Waters LAC/E configured for Empower with Secondary NIC card installed.
- 3 Straight cables and an Ethernet switch (Note: do not use a hub).

- Ensure all the modules are powered OFF.


- Using 3 straight cables, connect lace - switch and switch – instruments (e2695 and
2489).
- Power ON All modules.
- Empower will automatically detect the instruments. Go to the LAC/E properties, and to
the Instruments tab and click Scan instruments.
- The Empower instrument address is in the format W<Model># Serial Number where
<Model> is the model number (i.e. e2690/5, 2489, 2998, 2475) and Serial number is the
Module serial number.
Note: Only 2475 detector should have 2 wire cable connected from Alliance 2695 for
triggering the data acquisition.
3.7.2 Waters Acquity UPLC
Requirements:
- Waters LAC/E configured for Empower with Secondary NIC card installed.
- A crossover cable (if connecting a single instrument to a LAC/E NIC).
- A minimum of 3 network cables and an Ethernet switch (if connecting multiple instruments to
the LAC/E instrument NIC) (Note: do not use a hub).

- Ensure all the Acquity UPLC modules are powered OFF.


- The HT column heater, column manager, or column heater/cooler incorporates an
internal Ethernet switch. that accommodates the PC (workstation) and up to six
ACQUITY UPLC modules. Connect the shielded Ethernet cables from each module to
the rear panel of the HT column heater, column manager, or column heater/cooler. The
HT column heater, column manager, or column heater/cooler is connected internally to
the Ethernet switch.
- Connect the HT column heater, column manager, or column heater/cooler to the LAC/E
instrument NIC using a crossover cable, if connecting one system.
- Using 3 straight cables, Connect LAC/E - switch and switch - HT column heater, column
manager, or column heater/cooler.
- Power ON All modules.
- Empower will automatically detect the instruments. Go to the LAC/E properties, and to
the Instruments tab and click Scan instruments.
- The Empower instrument address is in the format ACQ-MOD#Serial Number where
<Mod> is the 3 letters of Module (i.e.QSM, FTN, CM, PDA, TUV) and Serial number is
the Module serial number.

3.8. Connect Waters system (IEEE & Ethernet)


3.8.1 Waters 2690/5 Alliance with 2998 or 2489 or 2475
Requirements:
- Waters LAC/E configured for Empower with busLac/E interface card and Secondary NIC
card installed.
- A crossover cable.
- IEEE488 cable.

- Ensure all Modules are powered OFF.
- Use an IEEE488 connector to connect 2690 module  BusLAC/E card on LAC/E box.
- Use a cross cable to connect Secondary NIC of LAC/E 2489/2998/2475 detector.
- 2690/5 module connected to a given LAC/E must have a unique IEEE488 address.
Refer to the instrument manuals to set this address.
- Power ON All modules.
- Empower will automatically detect the instruments. Go to the LAC/E properties, and to
the Instruments tab and click Scan instruments.
- The Empower instrument address is in the format below format
W<Model>@ n where <Model> is the model number (i.e. 2690/5) and n is the IEEE488
address
W<Model># Serial Number where <Model> is the model number (i.e. 2489, 2998, 2475)
and Serial number is the Module serial number.

Note: Only 2475 detector should have 2 wire cable connected from Alliance 2695 for
triggering the data acquisition.

3.9 Connect Agilent system (Ethernet)


3.9.1 Agilent HPLC (1100/1200) UPLC (1260/1290)
Requirements:
- Waters LAC/E configured for Empower with Secondary NIC card installed.
- A crossover cable (if connecting a single instrument to a LAC/E Secondary NIC).
- A minimum of 3 network cables and an Ethernet switch (if connecting multiple instruments to
the LAC/E Secondary NIC) (Note: do not use a hub).

- Ensure all the modules are powered OFF.


- Connect each Module with other Module using CAN cables (Module1 – Module2,
Module 2 – Module 3 and so on).
- Connect Detector to the LAC/E instrument NIC using a crossover cable, if connecting
one system.
- Using 3 straight cables, Connect LAC/E - switch and switch – Detector of Instrument1
and switch – Detector of Instrument2.
- Make sure each instrument has IP address assigned (192.168.0.2 for Instrument1 &
192.168.0.3 for Instrument2). Refer to the instrument manuals to set this address.
- Power ON All modules.
- Go to the LAC/E properties, and to the Configure DHCP tab and click Configure DHCP,
add, Enter the IP address & MAC address (MAC can found on the back side of the
instrument) of Instrument 1, Select Instrument Type as Agilent LC and Type Serial
Number (Can give like HPLC1 or Detector Serial Number) and Click ok.
- Go to Instruments tab and Click Scan Instruments. This should show the instruments.

3.9.2 Agilent 7890A/B GC with G1888 Headspace


Requirements:
- Waters LAC/E configured for Empower with Secondary NIC card installed.
- A minimum of 3 network cables and an Ethernet switch (Note: do not use a hub).

- Ensure all the modules are powered OFF.
- Using 3 straight cables, Connect LAC/E - switch and switch – GC and switch –
Headspace.
- Make sure each instrument has IP address assigned (192.168.0.2 for GC & 192.168.0.3
for Headspace). Refer to the instrument manuals to set this address.
- Power ON All modules.
- Go to the LAC/E properties, and to the Configure DHCP tab and click Configure DHCP,
add, Enter the IP address & MAC address (MAC can found on the back side of the
instrument) of GC, Select Instrument Type as A7890 and Type Serial Number (Can give
like HPLC1 or GC Serial Number) and Click ok.
- Follow the above step again for configuring G1888. The only difference is Instrument
Type and Serial Number.
- Go to Instruments tab and Click Scan Instruments. This should show the instruments.

3.9.3 Agilent 7890A/B GC with 7697 Headspace


Requirements:
- Waters LAC/E configured for Empower with Secondary NIC installed.
- A minimum of 3 network cables and an Ethernet switch (Note: do not use a hub).

- Ensure all the modules are powered OFF.


- Using 3 straight cables, Connect LAC/E - switch and switch – GC and switch –
Headspace.
- Make sure each instrument has IP address assigned (192.168.0.2 for GC & 192.168.0.3
for Headspace). Refer to the instrument manuals to set this address.
- Power ON All modules.
- Go to the LAC/E properties, and to the Configure DHCP tab and click Configure DHCP,
add, Enter the IP address & MAC address (MAC can found on the back side of the
instrument) of GC, Select Instrument Type as A7890 and Type Serial Number (Can give
like HPLC1 or GC Serial Number) and Click ok.
- Follow the above step again for configuring 7697. The only difference is Instrument Type
and Serial Number.
- Go to Instruments tab and Click Scan Instruments. This should show the instruments.

3.10 Connect Agilent system (Serial)


3.10.1 Agilent 6890/6890+/6890N with 7694 headspace
Requirements:
- Waters LAC/E configured for Empower with Secondary NIC installed.
- A Crossover cable.
- Avocent Hub
- 2 DB9-RJ45 Adaptors & 2 Serial cables

- Ensure all the modules are powered OFF.
- Using a Cross over cable, Connect Secondary NIC of LAC/E – LAN on Avocent hub.
- Connect DB9-RJ45 Adaptor to Modem/RS-232 port on GC. Then Connect the Serial
cable to other end of DB9-RJ45 Adaptor to Port 1 on Avocent Hub.
- Connect DB9-RJ45 Adaptor to RS-232 port on Headspace. Then Connect the Serial
cable to other end of DB9-RJ45 Adaptor to Port 2 on Avocent Hub.
- Make sure below settings are configured on GC & Headspace (Refer to instrument
manuals).

- Make sure Avocent hub has IP address assigned (192.168.0.100) (Refer to Avocent hub
manual).
- Power ON All modules.
- Go to the LAC/E properties, and to the Serial ports tab, Select the 6890 on port 1 of 8
port serial device section and select HS7694 on port 1 of 8 port serial device section.
- Go to Instruments tab and Click Scan Instruments. This should show the instruments.

3.11 Configure system in Empower


Ensure all instruments are connected and powered up.
From configuration manager select File > New > Chromatographic System. Then select create a
new system. After making a selection Empower will prompt to select the appropriate LAC/E.
A dialogue is displayed showing all the instruments connected to the LAC/E.
Select the required instruments from the available instruments list and drag to the new system
list. Once all the required instruments have been selected click ‘Next’.
An access control dialogue is displayed.
- Check the Share instruments box and Owner and Groups option
- Select the user groups who require access
- Do not check the password required box.
- Next
The system name dialogue is displayed.
- Enter the system name in the format XXX<Site convention> where XXX is the 3 letter
site IT code and <Site convention> identifies the chromatographic system.
Note: The site naming convention must ensure that instrument names are unique.
- Enter a system comment which includes a brief description of the system and its
location.
- Check the ‘on line’ box
- ‘Finish’
The new system will appear in the System list in configuration manager.
