This ISE User Guide (Supplement) offers detail descriptions, limited troubleshooting to common
problems, and maintenance procedures to be used in conjunction with the conventional user guide.
Use also the conventional user guide.

Whom and What This Manual is for
This manual is intended for laboratory personnel, and explains troubleshooting techniques and
maintenance procedures on a day-to-day basis for the ISE unit of the AU480.

Objective readers
This manual is intended for laboratory personnel who have completed the training course offered by
Beckman Coulter, or personnel who have been trained by those who have taken Beckman Coulter
training course.

In order to use the optional unit ISE safely and under optimal conditions, the aim of this manual is to

ISE MAINTENANCE FLOW
In order to maintain the original performance of the AU480 ISE unit
and to keep it operating in good condition users should perform
periodic maintenance on the analyzer.
This part of “ISE MAINTENANCE FLOW” describes the daily,

5 DAILY MAINTENANCE

5.1 Check the Buffer Syringe for Leaks


The buffer syringe dispenses a precise small amount of serum or reagent. If liquid leaks from
the syringe, the correct amount of buffer solution is not dispensed and may cause the syringe to
malfunction. Before starting daily analysis, be sure to check the buffer solution and sample syringes
for leaks.

Procedure for check the Buffer Syringe for Leaks


Materials:

Dry, clean cloth or paper towel

The R syringe is used for the ISE buffer solution.


If the syringe needs to be replaced because of leaks check, prepare a new R
TIP syringe.

1. Open the right front door of the analyzer.


2. Verify that the system in either Warm up or Standby mode and the power to the system is off.

• If protective gloves, etc., get wet, thoroughly rinse them immediately with water.

• Verify that strong alkali such as AU wash solution alkali does not come in contact
CAUTION with the syringe cases. If the alkali wash solution comes in contact with the
syringe case cracks may occur. If there is contact with the syringe case rinse
thoroughly with water.

3. Using a dry cloth, wipe the bottom of the syringe case, the joint of the case head and syringe
case, and the vicinity of fixing screws to check for leaks.
If any of the liquid has leaked, wipe it off with a dry clean cloth.
4. Visually check the case head for cracks.
If any cracks are found on the case head, replace it with a new one. After replacing the case head
advance to step 9.
5. Check to see if the case head and the syringe are loose.

• If the case head and syringe case are loose, tighten them by turning the syringe case
clockwise toward the case head.

• If any leak was found, allow approximately five minutes after tightening the case head and
syringe case. Then, recheck the bottom of the syringe case for leaks.

6. Check to see if the two fixing screws on the case head, fixing nut, and piston fixing screw are
loose.

• If loosen, firmly tighten the fixing screws, fixing nut, and piston fixing screw.

• If a leak was found, allow approximately five minutes after tightening the case head and
syringe case. Then, recheck the bottom of the syringe case, etc. for leaks.
7. Visually check to see if any liquid is leaking inside the syringe case.
If any leak was found, go to section “11.6 Replace the Buffer Syringe” in this chapter.
8. From the AU480 “Home” window select Menu List>Maintenance>User Maintenance>ISE
Maintenance>Maintenance to display the “ISE Maintenance: Maintenance tab” window.

9. Select I n s p e c t I S E b u f f e r S y r i n g e f o r L e a k from the list of maintenance


items.
10. Select U p d a t e .
A confirmation window appears.
11. Select O K .
12. Close the right front door of the analyzer.

• Reagent may drip from the case head during syringe replacement. Wipe off the
reagent with a paper towel.
CAUTION • If leaks still occur even after tightening the loose parts, replace the syringe.

5.2 Auto-wash the Sample Pot and Electrode Line

When handling wash solution, wear protective gloves to avoid contact with the wash
solution.
If hands or clothing come into contact with the wash solution, wash them
CAUTION immediately with water. Should wash solution get splashed accidentally in the eyes
or mouth, rinse it immediately with water and consult a physician.

If the sample pot and electrode line remain unwashed, contamination between samples will occur,
which may result in a calibration or data error. After completing daily analysis, be sure to auto-wash
the sample pot and electrode line. It takes approximately 4 minutes from start to auto-wash the sample
pot and electrode line.
If performing an analysis after auto-wash be sure to execute calibration before starting the analysis.

Procedure for Auto-wash the Sample Pot and Electrode Line


Materials:

• ISE Cleaning Solution : AUH1019

• Sample cup : MU8532

1. Verify that the system is in Warm up or Standby mode.


2. Fill the sample cup with approximately 1 mL of ISE Cleaning Solution.
3. Open the sub cover of the STAT table and press the TA B L E R OTAT I O N / D I A G button.
Rotate the STAT table until the “CLEAN” position marked on the STAT table is visible.
4. Set the sample cup filled with the ISE Cleaning Solution in the “CLEAN” position on the STAT
table.

Avoid spilling wash solution when handling the reagent bottle. If the wash solution is
spilled, wipe it off immediately with a cloth or paper towel soaked in water.

6 WEEKLY MAINTENANCE

6.1 Check the Selectivity of the Na and K Electrodes


The Na electrode and K electrode are ion-selective electrodes. If the selectivity of electrodes
deteriorates, the ISE unit is affected by ions other than those to be measured, affecting analysis
results.
To check the electrodes for deterioration, check the selectivity of the Na and K electrodes every week
after terminating analysis.

Do not leave the bottle of selectivity check liquid open. Evaporation may effect the
concentration of the selectivity check liquid or it may crystallize.
CAUTION

Procedure for Check the Selectivity


Materials:

• Selectivity check liquid : AUH1018

• Sample cup : MU8532

1. Verify that the system is in the Warm up or Standby mode.


2. Pour the Na and K electrode check liquid in separate sample cups.
3. Open the STAT table cover (small).
4. Press the TA B L E R OTAT I O N / D I A G button. Rotate the STAT table until the U-H and
U-L positions are visible on the STAT table.
5. Place the filled sample cups on the STAT table.

• Place the Na-electrode sample cup in the U-H (SEL-Na) position.

• Place the K-electrode sample cup in the U-L (SEL-K) position.


6. Close the STAT table cover (small).

The selectivity check procedure will start and display the check results.
11. Check the selectivity data.
For abnormal data, the background of the numeric value is displayed in yellow.
The system will judge a value more than 160 mmol/L for Na-electrode and a value more than 6.0
mmol/L for K-electrode as abnormal data.
If the check results are abnormal, replace the relevant electrode.
For details on replacing the electrode, refer to section “8.1 Replace the Na, K, or Cl Electrode” in
this chapter.

12. Remove previously placed Na and K sample cups from STAT table.
13. The maintenance date will be automatically updated in the list.

6.2 Enhanced Cleaning of Electrode Line


If the sample pot and electrode line remain unwashed, contamination between samples will occur,
resulting in a calibration or data error. After the Na and K selectivity check, perform the enhanced
cleaning of the electrode line procedure.
Refer to Nos. (1) to (11) in the “4 TUBING BLOCK DIAGRAM FOR ISE MAINTENANCE” in this
chapter.

Procedure for Enhanced Cleaning of the Electrode Line (Auto)


Materials:

• ISE Cleaning Solution : AUH1019

• Sample cup : MU8532

When handling wash solution, wear protective gloves to avoid contact with the wash
solution.
If hands or clothing come into contact with the wash solution, wash them
CAUTION immediately with water. Should wash solution get splashed accidentally in the eyes
or mouth, rinse it immediately with water and consult a physician.

2. Fill the sample cup with 1.5 mL of ISE Cleaning Solution.
3. Set the sample cup filled with the wash solution in the “CLEAN” position on the STAT table.

Maintenance>Maintenance to display the “ISE Maintenance: Maintenance tab” window.

7 MAINTENANCE REQUIRED EVERY TWO


WEEKS OR 3,000 SAMPLES

7.1 Manually Wash the Mix Bar and Liquid Level Sensors
If the mix bar and liquid level sensors are not washed over an extended period of time, they become
contaminated with serum, urine or a fur, etc.
Wash the mix bar and liquid level sensors with alcohol every two weeks or every 3,000 samples,
whichever comes first.

10. Loosen the knob clamping the mixing unit. Gently lift the mixing unit to dismount it.

Be careful not to bend or break the liquid level sensors when washing them.
CAUTION

11. Wipe the two liquid level sensors and mix bar with a cloth dampened with ethyl alcohol (ethanol).

14. Reconnect the connectors of the liquid level sensor (714) and mixing motor (706) to the mixing
unit.

The MID solution is supplied.
16. Close the ISE cover.
17. Close the main cover.

Do not change the orientation and position of two nozzles attached to the mixing unit
and do not use force pulling on the connecting tubes of the mixing unit.

7.2 Manually Wash the Sample Pot and the Sample Pot Tubes
The sample pot tubes are the three tubes that run between the sample pot, electrode unit, and T-
connector. When the system processes samples that contain large amounts of fiblin and protein debris
may build up in the tubes over time.

• Deionized water or distilled water

• Sonicator filled with deionized water

1. Verify that the system is in Warm up or Standby mode.

mount plate.
9. Loosen the sample pot clamp knob and lift the sample pot.
10. Loosen the tube connected to the bottom of the sample pot, and then dismount the sample pot.
Remove the sample pot and tubes on the electrode unit side (location (4) in the tubing block
diagram) from the electrode unit.

11. Remove the tube attached to the bottom of the sample pot.
12. Remove the tube from the electrode unit and the bypass tube.
13. Remove three tubes from the T-connector.
14. Supply washing water or the sample diluent of approximately 2% concentration of alkali wash
solution to the sonicator filled with deionized water.

To prevent the REF electrode block from deforming due to the ultrasonic cleaning,
follow these precautions.
• Do not perform ultrasonic cleaning for more than ten minutes.
CAUTION
• Use cleaning liquid at room temperature.

• Use the sonicator rated at 600W or less.


If the output of the sonicator is uncertain, contact the manufacturer of the
sonicator.

15. Wash the T-connector and the two tubes together with the sample pot in the sonicator filled with
deionized water for approximately 10 minutes.
16. Rinse the sample pot, T-connector, and the three tubes with deionized water or distilled water.
17. Wipe the sample pot, T-connector, and the three tubes with a wiper cloth.
18. Connect the three tubes to the T-connector.

Make sure to push the tubes all the way on the T connector to avoid leaks.
CAUTION

19. Connect the other end of the tube to the original position of the electrode unit and the bypass
tube.
20. Attach the tube to the bottom of the sample pot.
Tighten the tube knob firmly.

Tighten the knob firmly when attaching the tube to the bottom of the sample pot.
CAUTION

21. Remount the sample pot in place.


Insert the sample pot fixing knob into the notch on a side of the sample pot, and then match the
hole on the other side of the pot with the positioning pin.
22. Tighten the sample pot fixing knob to secure the sample pot.
23. Gently remount the mixing unit in place, then secure the unit by tightening the knob.

When remounting the mixing unit be careful not to get the tube caught in the
analyzer’s peripheral part.

12. Connect the blue cord, yellow cord, and red cord to the Cl electrode, Na electrode, and K
electrode, respectively.
13. Mount the three new electrodes on the electrode block.
Mount the electrodes in order of Cl, Na, and K from the sample pot side to the front side.
14. Move the lock lever forward to clamp the electrodes.
15. Press the TA B L E R OTAT I O N / D I A G button.
The MID solution is supplied to the tube.
The two roller pumps turn on and move the solution in each roller tube.
Press and hold down the TA B L E R OTAT I O N / D I A G button until the roller pumps are
activated five times. Visually check the tube to see if the solution passing through the electrodes
includes air bubbles. Repeat this operation until there are no air bubbles in the tube.
16. Select U p d a t e .
A confirmation window appears.
17. Enter the serial number of each replacement electrode, and then select O K .
The “Electrode Serial No. Setup” window is closed.
18. Close the ISE cover.
19. Close the main cover.
20. Allows at least 5 minutes after closing the main cover, and then perform calibration analysis.

To obtain the best possible analysis data, perform two successive calibration
measurements to confirm the electrode stability.
If the difference in factor value between the first and second calibrations is within the
CAUTION following values, the electrodes are stable. The MID solution concentrations in the
following table have been set from “Specific Test Parameters: ISE” menus.

MONTHS OR 20,000 SAMPLES
When performing maintenance required every three months or every 20,000 samples, replace
the roller tubes for MID solution dispense, mixture aspiration, and individual tubes (tubes between
the sample pot and electrode unit, drain tube on the REF electrode block side, pinch valve tubing,
and drain tube). This section describes a series of procedure in which maintenance is performed
concurrently for all tubes.
When performing maintenance for each tube separately, observe the procedure of a relevant
maintenance item described in section 9.1 or later.

9.1 Replace the Roller Tubes for MID Solution Dispense and
Mixture Aspiration
The roller tubes will deteriorate due to friction with each roller pump and vibrations. If used for an
extended period of time these tubes will break. Replace the roller tubes with new ones every three
months or every 20,000 samples.

9.2 Replace the Sample Pot Tubes


If the system analyzes samples that contain large amounts of fibrin and protein such as a dialysis
sample, fibrin, protein, etc. will accumulate near the T-connector between the sample pot and
electrode unit, which may result in a calibration or data error.
Since the accumulated contaminants may not be removed with the washing procedure performed
every two weeks or every 3000 samples, replace the tubes and T-connector with new ones every
three months or 20,000 samples.
For details on how to clean sample pot, tubes, and T-connector, refer to section “7.2 Manually Wash
the Sample Pot and the Sample Pot Tubes” in this chapter.

Performing the Maintenance Procedure


1. Disconnect the connectors of the liquid level sensor (714) and mixing motor (706) from the mixing
unit.
2. Loosen the knob clamping the mixing unit. Gently lift the mixing unit to dismount it and place it on
mount plate.
3. Loosen the sample pot clamp knob and lift the sample pot.
4. Loosen the tube connected to the bottom of the sample pot, and then dismount the sample pot.
Remove the sample pot and tubes on the electrode unit side (location (4) in the tubing block
diagram) from the electrode unit.
5. Connect the new set of tubes in place in order of the electrode unit side and waste liquid side (B
in the tubing block diagram).
Attach the tube to the bottom of the sample pot.

Make sure to push the tubes all the way on the T connector to avoid leaks. Tighten
the knob firmly when attaching the tube to the bottom of the sample pot.
CAUTION

6. Remount the sample pot in place.


Insert the sample pot clamp knob into the notch on a side of the sample pot, and then match the
hole on the other side of the pot with the positioning pin.
7. Tighten the sample pot clamp knob to clamp the sample pot.
8. Gently remount the mixing unit in place, and then clamp the unit by tightening the knob.

When remounting the mixing unit in place, be careful not to splash the buffer solution
and MID solution.
CAUTION

9. Reconnect the connectors of the liquid level sensor (714) and mixing motor (706) of the mixing
unit in place.

Tubing
The REF block-side drain tube is subject to accumulation of contaminants and may adversely affect
analysis data due to flow-rate decrease and noise interference if it is used over an extended period of
time. Replace the drain tube with a new one every three months or every 20,000 samples.
The pinch valve tubing is provided with pinch valves.
If the pinch valve tubing is used for an extended period of time, the section pinched by the valve
will deteriorate. Replace the pinch valve tubing with a new one every three months or every 20,000
samples. Concurrently replace the REF block-side drain tube and pinch valve tubing.

Performing the Maintenance Procedure


1. Move the lock lever toward the back to release the electrodes from being clamped.
2. Gently lift up the block on which the REF electrode is mounted.
3. Pull out the REF electrode cord.
4. While holding the REF electrode block by hand, pull the tube set 2 out of the REF electrode
block.
5. Attach the tube side of a new tube set 2 to the REF electrode block (6).
6. Reconnect the REF electrode cord in place.
7. Set the REF electrode block in the original position.
8. Move the lock lever to the clamp position.
9. Disconnect the three ends of the pinch valve tubing.
Disconnect the tube ends at positions A, B and C in the tubing block diagram.
10. Remove the pinch valve tubing from the pinch valve groove.
Remove the tube in the top groove of the pinch valve directly upward. Remove the tube in the
bottom groove of the pinch valve upward while pressing the top end of the pinch valve with finger.

Do not mount each branch tube in a different groove on the pinch valve. If a tube is
mounted backwards, liquid will not be supplied and drained correctly.
Mount a shorter tube in the bottom groove of the pinch valve (between A and C in
CAUTION the tubing block diagram). Mount longer tube in the top groove of the pinch valve
(between B and C in the tubing block diagram).

11. Mount a new section of pinch valve tubing by connecting the branches at positions A, B, and C in
the tubing block diagram.
Connect the longer Y-branched tube at position B.
12. Insert the two branch tubes into the grooves on the pinch valve, so that each tube passes through
the middle of the groove.
Mount the tube in the bottom groove of the pinch valve while pressing the top end of the pinch
valve with finger, in the same way the tube was removed. Mount the tube in the top groove of the
pinch valve directly.

Performing the Maintenance Procedure


1. Remove the drain tube end from the tube joint (No.3) of the roller pump for mixture aspiration.
The drain tube end on the roller pump side in block G in the tubing block diagram is
disconnected.
2. Remove the drain tube end from the hook on the drain tank, and then dismount the whole drain
tube.
The drain tube in block D in the tubing block diagram is dismounted.

When handling the solution hypochlorite solution, wear protective gloves to avoid
coming into contact with the sodium hypochlorite solution.
If hands or clothing come into contact with the wash solution, wash them
CAUTION immediately with water. Should wash solution get splashed accidentally in the eyes
or mouth, rinse it immediately with water and consult a physician.

3. Prepare approximately 50 mL of the diluted sodium hypochlorite solution.


4. Pour the diluted sodium hypochlorite solution into the drain tank directly from its top (position H in
the tubing block diagram).
Pour 50 mL as a guide of the solution amount.
5. Allow approximately 10 minutes after pouring the sodium hypochlorite solution, and then pour a
sufficient amount of water into the drain tank from its top (position H in the tubing block diagram)
to rinse out the sodium hypochlorite solution.
6. Connect a new drain tube to the mixture aspiration roller tube and the drain tank side, and then
remount it in place.

When connecting the tubing, make sure to push the tube all the way to avoid leaking
joints.
CAUTION

Verify that the solution aspired reliably from the sample pot into the electrode unit
and drained from the drain tube, and verify that no air bubbles are present in the
drain tank.

hand, otherwise the cord could break.
CAUTION

12. Loosen the set screw, then gently remove the REF electrode along with the set screw.
13. Remove the packing of the REF electrode.

Attaching the REF electrode


1. Verify that no air bubbles are in the electrode bar on a new REF electrode.
If air bubbles are found on the electrode bar of the new REF electrode, remove the bubbles by
pointing the electrode bar down and tapping it with a finger.
2. Insert a new packing into the REF electrode insertion hole.
At this time verify that the packing is not cracked or broken. If so, replace the packing.
3. Insert the set screw into the REF electrode insertion hole while turning it.
Insert the screw all the way until torque changes.
4. Insert the REF electrode into the REF electrode insertion hole (through the set screw).
Insert the REF electrode so that the electrode tip can be properly inserted into the packing.

The REF electrode tip can be easily inserted into the packing if it is dampened with
deionized water.
TIP

5. After inserting the REF electrode tighten the coupling screw by rotating it 1/4 to 1/2 turn, then
secure the REF electrode.
6. Connect the REF electrode cord correctly.
7. Remount the block in place.
8. Clamp the lock lever.
9. Select M I D / R E F P r i m e .
10. Select O K .
11. Press the TA B L E R OTAT I O N / D I A G button.
The MID solution will be injected into the tube.
The two roller pumps are activated and solution flows into the individual roller tubes.
Press the TA B L E R OTAT I O N / D I A G button until the roller pumps are activated five
times or so. Visually check for air bubbles in the tubes.
Press the TA B L E R OTAT I O N / D I A G button again until no air bubbles are found in the
tube.
If air bubbles will not purge, push in the cap screw.

11.1 Replace the Sample Pot


Replace the sample pot if accumulated contaminants cannot be removed during the washing
procedure performed every two weeks or every 3000 samples or if any breakage or a large flaw is
found in the pot.

Performing the Maintenance Procedure


1. Disconnect the connectors of the liquid level sensor (714) and mixing motor (706) from the mixing
unit.
2. Loosen the knob clamping the mixing unit. Gently lift the mixing unit to dismount it and place it on
mount plate.
3. Loosen the sample pot clamp knob to dismount the sample pot.
4. Loosen the knob on the tube attached to the bottom of the sample pot to remove the tube from
the pot.
5. Attach the tube to the bottom of a new sample pot.

Tighten the knob firmly when attaching the tube to the bottom of the sample pot.
TIP

6. Remount the sample pot in place.


7. Insert the sample pot clamp knob into the notch on a side of the sample pot, and then match the
hole on the other side of the pot with the positioning pin.
8. Tighten the sample pot clamp knob to clamp the sample pot.
9. Gently remount the mixing unit in place, and then clamp the unit by tightening the knob.

When remounting the mixing unit in place, avoid splashing the buffer solution and
MID solution.
CAUTION

10. Reconnect the connectors of the liquid level sensor (714) and mixing motor (706) of the mixing
unit in place.
11. Press the TA B L E R OTAT I O N / D I A G button two or three times.
Confirm that liquid is properly aspired from the sample pot to the electrode unit.
If liquid remains in the sample pot check, the tube connections.

Performing the Maintenance Procedure


1. Move the lock lever to release the electrodes from being clamped.
2. Remove the three electrodes from the electrode unit.
3. Pull the cords connected to the Cl electrode, Na electrode, and K electrode out of each electrode.
4. Remove the tube connecting to the sample pot from the electrode block.

Four O-rings are attached to each side of three electrodes and the metal part
(location E in the tubing block diagram) that comes in contact with the Cl electrode.
CAUTION Be careful not to loosen the O-rings when removing the electrodes.

5. Pass the mandolin strings through the flow path hole on the side of the electrode block.
If contamination disturbed, it may get stuck in the mixture flow path in the electrode block. Bind
the mandolin strings up to the maximum thickness that can pass through the flow path.
6. Remove the stuck contamination by turning the mandolin strings while holding both ends by
hand.
When contaminants stick on the mandolin strings, wipe them with a dry, clean cloth or paper
towel several times.
7. Connect the blue cord, yellow cord, and red cord to the Cl electrode, Na electrode, and K
electrode, respectively.
8. Mount the three electrodes on the electrode block.
Attach the electrodes in order of Cl, Na, and K from the sample pot side.
9. Move the lock lever to clamp the electrodes.
10. Mount the tube connecting from the sample pot to the electrode block.
11. Press the TA B L E R OTAT I O N / D I A G button.
The two roller pumps are activated and solution flows into individual roller tubes.
Press the TA B L E R OTAT I O N / D I A G button until the roller pumps are activated five

Performing the Maintenance Procedure


1. Move the lock lever to release the electrodes from being clamped.
2. Remove the K electrode from the electrode stack.
3. Remove the electrode electrical connection.
4. Remove the O-ring from the electrode.
5. Wash the O-ring and O-ring inserted groove on the electrode side using the nozzle-type wash
bottle filled with deionized water.
At this time there is no problem if deionized water comes in the electrode flow path during
washing.
6. Wipe the side face (location F in the tubing block diagram) of the REF electrode block in contact
with the K electrode using a clean cloth dampened with deionized water.
7. Sufficiently wipe the K electrode, O-ring, and REF electrode side faces that have been washed
with deionized water using a dry, clean cloth.
8. Connect the electrode wire to the electrode.
9. Attach the K electrode to the electrode stack.
Mount the electrodes in order of Cl, Na, and K from the sample pot side toward the lock lever
side.

Three O-rings are attached to one side of three electrodes. Be careful not to lose the
O-rings when mounting the electrodes.
CAUTION

10. Move the lock lever to clamp the electrodes.


11. Confirm that D ra i n - F l o w c e l l has been selected on the “ISE Maintenance: Maintenance
tab” window.
12. Press the TA B L E R OTAT I O N / D I A G button.
The two roller pumps are activated and solution flows into the individual roller tubes.

Performing the Maintenance Procedure


1. Move the lock lever to release the electrodes from being clamped.
2. Gently lift up the block on which the REF electrode is mounted.
3. Pull the cord connecting to the REF electrode out of the electrode.
4. Loosen the cap screw on the REF electrode and gently remove the electrode along with the cap
screw.
Remove the REF electrode packing in the block.
5. While holding the REF electrode block by hand, pull the waste liquid drain tube out of the REF
electrode block.
6. Remove the REF solution tube (location G in the tubing block diagram) connected to the lower
side of the REF electrode block.
Dismount the REF electrode block.
7. Attach the REF solution tube (location G in the tubing block diagram) to a new REF electrode
block.
8. Insert the REF electrode packing into the block.
If the packing is cracked or broken. Replace the packing (part No.MU9900).
9. Insert the cap screw into the REF electrode insertion hole while turning it with fingers. Insert the
screw all the way until torque changes.
10. Verify that there are no air bubbles in the electrode block of the REF electrode.
If any air bubble is found, remove the air bubble by tapping the electrode block with fingers with
pointing it downward.
11. Insert the REF electrode into the REF electrode block.
At this time make sure that the REF electrode tip is correctly inserted into the packing.

The REF electrode tip can easily be inserted into the packing if it is damped with
deionized water.
TIP

12. After inserting the REF electrode tighten the cap screw by rotating it 1/4 to 1/2 turn, then secure
the REF electrode.
13. Connect the roller tube for waste liquid with the roller pump.
14. Reconnect REF electrode cord in place.
15. Remount the REF electrode block in place.
16. Move the lock lever to the clamp position.
17. Confirm that D ra i n - F l o w c e l l has been selected from the “Single Operation” buttons on
the “ISE Maintenance: Maintenance tab” window.
18. Press the TA B L E R OTAT I O N / D I A G button.
The two roller pumps are activated and solution flows into the individual roller tubes.

To prevent the REF electrode block from becoming deformed due to ultrasonic
cleaning, follow these precautions. If the REF electrode block has been deformed or
cracked, replace it.
CAUTION
• Do not perform ultrasonic cleaning for more than ten minutes.

• Use a cleaning liquid at room temperature.

• Use a sonicator rated at 600W or less.


If the output of the sonicator is uncertain, contact the manufacturer of the
sonicator.

Procedure for wash the REF electrode block:


Perform ultrasonic cleaning for the electrode block dismounted in step 6 of this
maintenance procedure for 10 minutes using a sonicator which is filled with a 2%
TIP
sample diluent of AU alkaline wash solution.
If no sonicator is available, soak it in the 2% sample diluent of AU alkaline wash
solution for more than 30 minutes. At this time make sure that the diluted wash
solution flows into the flow path in the electrode block.
After cleaning the electrode block, thoroughly rinse the wash solution from the block
under running water, wipe it with a clean cloth, and then implement the maintenance
procedure from step 7 to step 21.

Refer to section “10.1 Replace the REF Electrode and Packing” in section
“10. MAINTENANCE REQUIRED EVERY TWO YEARS OR 150,000 SAMPLES” in this chapter.

11.5 Replace Reagents


The ISE reagent on board stability is specified below for each reagent. Replace expired reagent
bottles with new ones. When the quantity of reagent becomes insufficient, an alarm message will
appear. After the message displays the system is capable of analyzing approximately 180 samples.
Replace the reagent before the bottle empties.

On Board Stability of Reagents for the ISE Unit


The on board stability for the buffer solution, MID solution, and REF solution used for the ISE unit are
specified. If an expired reagent is used, data will be incorrect. Replace the corresponding reagent
before the on board stability expires.
On Board Stability: Check the package insert for on board stability claims.

On board stability Remark


Buffer solution 2 months after breaking seal New solution can be added only once.
MID solution 1 month after breaking seal No solution can be added.

Procedure for Replace ISE Reagents


Materials:

• Buffer solution tank : AUH1011

• MID solution tank : AUH1012

• REF solution tank : AUH1013

• Never replace reagents during analysis. A data error will result.

• Reference solution is highly concentrated. Do not contaminate other bottles with


CAUTION Reference solution.

1. Open the right front door of the analyzer.


2. Pull out the reagent bottle to be replaced.
3. Loosen the cap of the reagent bottle, then remove the aspiration tube.

• When the aspiration tube is removed from the tank, the solution in the tube may
drip in the system. If this occurs wipe it off with a dry cloth or paper towel wearing
protective gloves.
CAUTION
• Do not handle the aspiration tube with bare hands. Reagents will become
contaminated, resulting in defective data.

4. Place the aspiration tube in a new bottle, tighten the cap, and place it on the instrument.
Dispose of the old solution as liquid waste is handled.

• Push the reagent tank all the way until it stops. If the reagent tank is positioned forward,
the liquid level sensor will not detect the reagent level. This may display an alarm message
indicating there is insufficient reagent.
CAUTION
• If the roller tube for the MID solution is removed from the roller pump, the solution in the
tubing flows backwards into each reagent tank. Do not remove the roller tube for the MID
solution from the roller pump when replacing either of the reagent tanks.

5. Push the reagent bottle to the back of its position.

If the O-ring remains in the case head, carefully remove the O-ring with tweezers. Be careful not
to scratch the case head.

• Set the R syringe in place. The S syringe and R syringe are identified with the piston shaft
diameter.

D = 5mm: R syringe
D = 2mm: S syringe

• Do not pull out the piston from a new syringe. Once the piston has been pulled out, the
syringe cannot maintain the dispensing accuracy even if the piston is inserted in the
original position.

8. Insert the new R syringe into the case head.


9. Mount the syringe case on the case head.
10. Insert the syringe into the mounting grooves by holding the case head. Tighten the lock nut while
holding the case head by hand.

11.7 Procedure for Enhanced Cleaning of the Electrode Line


(Manual)
To help eliminate sample buildup and residue from high volume sample testing, this ISE cleaning may
be necessary on an as-needed basis. This method should be used when the ISE calibration slopes
are affected, or if a build-up/residue is present upon inspection of the sample pot or T-tubing. This is
the ONLY ISE cleaning procedure recommended by Beckman Coulter.

Materials:

• ISE Cleaning Solution : AUH1019

• Pipette

When handling wash solution, wear protective gloves to avoid contact with the wash
solution.
If hands or clothing come into contact with the wash solution, wash them
CAUTION immediately with water. Should wash solution get splashed accidentally in the eyes
or mouth, rinse it immediately with water and consult a physician.

Clean the Electrodes


1. Verify that the system is in Warm up or Standby mode.
2. Open the main cover.
3. Open the ISE cover.
4. From the AU480 “Home” window select Menu List>Maintenance>User Maintenance>ISE
Maintenance>Maintenance to display the “ISE Maintenance: Maintenance tab” window.

Materials:

1. Open the right front door of the analyzer.


2. Verify that the system is in Warm up or Standby mode.
3. Loosen the fixing screws of the two tubes connected to the case head and remove the tubes.

4. Loosen the piston fixing screw located at the bottom of the syringe.
5. Loosen the lock nut located at the top of the syringe.
6. Pull the syringe case to the front to remove the syringe from the mounting grooves.

To avoid damaging the syringe case, do not apply excessive force to the fixing
screws when removing them.
CAUTION

7. While holding the case head and syringe case, turn the syringe case counterclockwise to remove
it.

Avoid getting AU Alkali wash solutions on the syringe case and syringe head. The
strong alkali adhering to the syringe case and case head will cause cracks on them.
If the strong alkali has adhered to, remove the syringe case and case head and
CAUTION wash them with water.

12 RESETTING THE SYSTEM WHEN SWITCHED


TO THE STOP MODE WHILE PERFORMING
MAINTENANCE
After performing maintenance in Stop mode, reset the system by performing the following:
1. Determine and correct the cause of the error.
2. Confirm that “Analyzer Maintenance: Maintenance tab” window is displayed from the AU480
“Home” window in following order.
3. Select U p d a t e .
4. Select C a n c e l .
5. Select the S t o p / S t a n d b y button. After initialization the analyzer goes into Warm up or
Standby mode.
