1. About MaestroNano
MaestroNano, a micro‐volume spectrophotometer, is suitable for research use only.
The main function of MaestroNano is the concentration measurement from two
micro liter nucleic acid or protein solution. Nucleic acid means double‐stranded
nucleic acid (dsDNA), single‐stranded nucleic acid (ssDNA), and ribonucleic acid
(RNA).
MaestroNano has an embedded system, and there are ONLY three buttons as user
interface. After dispensing sample on the surface of fiber terminals, users may use
the buttons to select sample type and begin to measure. The measurement results
can be obtained from liquid crystal display (LCD), and thermal printer.
MaestroNano is a specialized instrument for bio‐chemical research only, especially
for micro‐volume solution concentration measurement. It is designed by Maestrogen
and realizes a user‐friendly interface, accurate measurement, competitive price, and
stylish industrial design. To satisfy most people’s requirements for concentration
measurement, MaestroNano defines a most reliable measurement range of
concentration of 2~2000 ng/μL, and successfully resists the interferences of ambient
light for a more accurate measurement. Also, MaestroNano does not intend to
change light pathlength during measurement to reduce frequency of calibration.
Instead, MaestroNano develops and offers an in‐house mathematical algorithm to
help users acquire a more reliable, repeatable, and reproducible bio‐chemical
solution concentrations.
Please handle optical fiber with care. When unpacking MaestroNano, please remove
the top filler material with care, not to damage or pull/drag the optical fibers. When
packing MaestroNano if needed, please cover the instrument with the same bubble
bag as we provided and place into the lower filler material for firm fixation. Place the
top filler material on MaestroNano as the way you opened the box for the first time,
letting the optical fiber fixed by bubble bag and top filler material. When operating
MaestroNano, it is suggested not to touch optical fiber for any purposes. In our
suggested operation flow, there is no need to handle with optical fiber segments,
only handling with metal pads that fixed with optical fibers. For suggestions to our
local service centers, please check with your customers when delivered if the
functions of MaestroNano are no problem. All the instructions abovementioned are
wholeheartedly cared due to we always hope to notify our customers with general
notices and avoid unintentional damage.

WARRANTY
The MaestroNano is warranted against defects in materials and workmanship for 1
year. If any defects occur in the instrument or accessories during this warranty period,
Maestrogen will repair or replace the defective parts at its discretion without charge.
The following defects, however, are specifically excluded:
1. Defects caused by improper operation.
2. Repair or modification done by anyone other than Maestrogen or an authorized
agent.
3. Damage caused by substituting alternative parts.
4. Use of fittings or spare parts supplied by anyone other than Maestrogen.
5. Damage caused by accident or misuse.
6. Damage caused by disaster.
7. Corrosion caused by improper solvent or sample.
For any inquiry or request for repair service, contact your local Maestrogen office.
Inform Maestrogen of the model and serial number of your instrument.

standards and EMC regulations. Certified products are safe to use when operated in

instrument for purposes other than those for which it is intended, or by
modifications of the instrument not performed by Maestrogen or an authorized

IMPORTANT NOTICE
Please, read the installation instruction carefully before using the spectrophotometer.
This instrument is intended for clinical and research laboratory use with nucleic
acid/protein solution. Before each instrument delivered to customers, Maestrogen
performs an in‐house algorithm calibration by a non‐adjustable optical pathlength of
instrument between upper and lower fibers which are respectively inserted in the
upper‐portion and the base‐portion metal plates. In order to maintain the accuracy
of measurement, it is suggested to prevent the upper‐portion metal plate from
damage of any external force, and avoid impact, collision, vibration, or slight shaking
when the system is under measurement. It is also suggested that MaestroNano is
operated at a stable bench in room temperature (about 25℃) without strong wild
blowing, wetted and dusted bench surface, and fire or smoke.

Software Instruction
1. ”Main Menu” Page
The “Main Menu” page includes five items. User can select sample type in item 1,
run measurement in item 2, and print measurement result in item 3.

Maestrogen suggests user setting local time in item 4 when use the instrument for
the first time.

2. ”Select Sample” Page
Before starting measurement, user should select
sample solution type first in “Select Sample”
page. There are four sample types in
MaestroNano, and each type has its specialized
algorithm (more detail in appendices).

3. ”Start Measurement” Page
There are three sections in “Start Measurement” page. User can select to measure
blank, to measure sample solution, to switch
highlight bar to “Data_01” section, and to save
(or without save) & exit to “Main Menu” page.
The “Description” section displays the
description of “Function” section items. User can
not switch highlight bar to “Description” section.

After blank measurement, the “Data_01” section shows “Return” and “* blank”
items. It is the same as sample solution measurement, and the “* sample_n” item
shows in “Data_01” section. For each data sheet, the capacity for sample saving is

thirty. MaestroNano allows totally to save ten data sheets. It is easy for user to
re‐measure the blank, switch the highlight bar to “Blank” in “Function” section, and
then press “Enter” key.

When the highlight bar is switched to “Data_01” section, the lower part of screen
shows “Output” information. Move the highlight bar to select sample_1, and the
measurement result of sample_1 will be shown in the “Output” section. Also, user
can press “Enter” key to delete sample_1.

When measurement is completed, select “Save & Exit” item to execute “Data_01”
saving and exit to “Main Menu“ page.

4. ”Show & Print Result” Page
MaestroNano has a thermal printer, and the
width of thermal paper is 5 cm. Select the third
item of “Main Menu” page, and the display
shows “Data Menu” page. This page lists all
saved data sheet; the maximal capacity for
saving is ten sheets. The latest saved sheet is
always listed on top. Fig. 2‐14 “Data Menu” page
Switch the highlight bar to data sheet, and press
“Enter” key to printer option page.
There are three sections in the printer option page; the operation interface is similar
to ”Start Measurement” page. User can switch highlight bar between “Function”
section and “Data_01” section.
The function of the first item is to select all data (not including printing). Press one
time to select all data in the “Data_01” section (Fig. 2‐16), and press again to cancel
all selects (Fig 2‐15). After selecting printing data, user can move highlight bar to
“Print” and press “Enter” button to print selected data.

If there is any error occurred with printer, the display shows error code.
The troubleshooting suggestions are listed in appendices. If this error can not be
eliminated, please contact with your local customer service.

5. ”Time Setting” Page
Maestrogen suggests that user should set local time in item 4 when using this
instrument for the first time. The time setting description is as blow.
Upper row:
HH Hours, MM Minute, SS Second.
Lower row:
YY Year, MM Month, DD Date.
User can press “Up” or “Down” button to
change number, and press “Enter” to switch

6. Shutdown
The power switch button works at any page except “measurement” and “printer”
pages. To avoid accidentally pressing the button to shutdown the system,
MaestroNano shows the double confirmation message when user presses the power
switch button.

3. Installation & Operation Flow
INSTALLATION
Carefully unpack the MaestroNano as follow:
1. First remove the filler material at the top.
2. Remove the instrument from the two bubble material shells and place it on a
stable, horizontal surface.
3. Remove the protection packaging materials.
Stand alone installation
1. The instrument must be placed on a bench leaving at least 10 cm of space all
around in order to avoid any crash that may reduce the measurement accuracy.
2. Connect the instrument to the power using the annexed cable. The plug must
have a ground connection. The adaptor supplies 24 V DC voltage.
3. Make sure the instrument is not connected with PC while users want to start up.
GENERAL PRECAUTIONS
∙ Prevent the upper‐portion and the base‐portion metal from damage of any external
force.
∙ Do not measure any solution when the drawer is open.
∙ Do not splash liquids directly on the instrument.
∙ Switch off the instrument immediately after its use.
∙ The power switch button of the MaestroNano is no segment type. The user can
press first time to start the instrument, and press again to select shutdown. If user
presses the power switch button non‐intentionally when the instrument is being
operated, the system will display a shutdown confirmation message.
GENERAL OPERATION
Make sure the instrument is NOT connected to PC with USB port before starting up.
And pug power line before using MaestroNano to measure nucleic acids or proteins
solution.

Measurement Flow
Step 1: Press the power switch button one time
to start MaestroNano, and the display
shows “Main Menu” page when the
instrument is ready.
Step 2: Select the sample type in the “Select
Sample” page. User can press the “UP” /
“DOWN” button to change selection,
and the “ENTER” button to select
sample.
Step 3: After selecting the sample mode, the display shows “Main Menu” page.
Select the “Starting Measure” item. Switch the highlight bar to “Blank”.
Step 4: Open the arm of the upper‐potion metal, and then dispense the blank
solution on the surface of the base‐portion fiber terminal.

Step 5: Close the arm, and make sure that the solution is drawn into a column
between the surfaces of upper and lower fiber terminal.

Step 6: Press the “Enter” button to start measurement. The default setting of this
system is measuring blank solution first. Otherwise, sample solution with
nucleic acids or proteins cannot be measured. The display shows “blank:
measurement completed” after measurement.

Step 7: After blank solution measurement, clean the surface of upper and lower fiber
terminal with a soft clean cloth, and dispense the sample solution on the
surface of the base‐portion fiber terminal. Switch the highlight bar to select
“Sample” in “Function” section, and press “Enter” button.
Step 8: After dispensing sample solution, press “Enter” button to measure. User can
real‐time check the measurement result in the “Data” and “output” section.
Also, user can delete any unreasonable data in the “Data” section if needed.
Step 9: Clean the surface of upper and lower fiber terminal with a soft clean cloth.
Data Output
Method 1 – LCD:
After measuring, the LCD will display the latest results (Fig 2‐11) before user press
any key.
Method 2 ‐ Printer:
Please refer to the section of software instruction, or Fig 2‐15 and Fig 2‐16.
Method 3 ‐ USB connection (A male to A male cable):
User can access the measurement results from MaestroNano connected to PC
via USB transfer. The LCD will show the following information while
MaestroNano is connected to PC via USB connection.
After data access, user can remove the connection like removing USB flash
memory stick on PC. Step 1 is removing from software, and the step 2 is
removing from hardware.

4. Maintenance
CLEANING
Before cleaning the housing of MaestroNano with a soft clean cloth, make sure the
instrument is in the power‐off state and the power cord is disconnected. Maestrogen
suggests that the instrument should be cleaned by industrial alcohol (< 80% vol.) as
cleaning liquid. The instrument is designed for only one optical pathlength with
in‐house algorithm. Therefore, it is important to keep the surface of pin clean.

PARTS REPLACEMENT
If MaestroNano is occurred with the problems as below, please contact your local
customer service to exchange the problematic parts.
1. MaestroNano shows the wrong time (even after time setting). It indicates that the
battery may be dead. User can replace the battery on the side of printer. The
battery model is CR2032 (3V).
2. If the LED on the adaptor can not light up, please contact your supplier to replace
the adaptor.
3. The printer dose not work after trying all the troubleshooting suggestions. Please
replace printer.
4. After starting MaestroNano, the liquid crystal display (LCD) dose not show any
message, but the printer power led (Green) is light up. Please replace LCD.
5. When MaestroNano is measuring, a visible light can be seen from the base‐portion
fiber terminal, and a low frequency voice can be heard. If the measurement result
is unreasonable, user can check these two items to confirm if the light source is
dead or not.

REPLACING THE PRINTER PAPER
The printer uses the thermal paper, and its size is width 57mm and phi 15mm. Paper
replacing flow is described as follows:

Open the printer drawer, and press the hook on the printer.

Take out the paper roll and remove the roller.

Feed the paper roll into the slot (Fig. 4‐3), and then keep the edge of the paper in the
center between printer head and roller (Fig. 4‐4).

Push the printer cover to assemble with the printer head.

Finally, close the printer drawer. The paper roll replacement is completed.
RETURN FOR UPGRADE
MaestroNano is designed to allow users measuring the sample solution
concentration without a computer. If there is any updated in‐house algorithm for
faster measurement, Maestrogen will announce updating schedule at official
website.

5. Appendices
CALCULATION THEORY
DNA/RNA
The algorithm of MaestroNano is based on Beer‐Lambert Law. Theoretically, the
change of optical intensity (I) is adopted to calculate the optical density (O.D.) of the
sample solution having a certain optical path length, i.e., the absorbance (A) of light
at a specified wavelength that has passed through the sample solution having the
optical path length, which may be expressed as:
O.D. = A ≣ ‐Log(T) = ‐Log(I / I0)
where A is the absorbance of light having passed through the sample solution, T is
the transmittance, I and I0 denote the intensity of light having passed through the
sample solution and a reference/blank solution, respectively. Since the absorbance of
light having passed through the solution in a quartz tube of 10‐mm in width is
conventionally used as a reference, the absorbance converted from the optical
intensity as measured is normalized using the Beer‐Lambert Law based on the
10‐mm standard optical path to obtain the absorbance of light having passed through
the sample solution having a 10‐mm optical path.
According to the Beer–Lambert Law, the dependence of the solution concentration (c)
upon the optical path length (P) and the absorbance (A) can be expressed as:
c = (A × e) / P
where e is the wavelength‐dependent extinction coefficient. For example, the e value
is 50ng‐cm/μL for double‐stranded deoxyribonucleic acids (DNA), 33ng‐cm/μL for
single‐stranded deoxyribonucleic acids, and 40ng‐cm/μL for ribonucleic nucleic acids
(RNA). The nucleic acid concentration of the sample solution may be calculated
according to the absorbance of the standard optical path length (10mm) and the e
value. MaestroNano adopts single optical path length with in‐house developed
algorithms. After measurement, MaestroNano calculates the concentration from the
acquired results and embedded algorithms. The embedded algorithms of
MaestroNano are verified and calibrated to implant into the integrated circuits for
each instrument.
Theoretically, absorbance is proportional to the optical path length. Therefore, users
can fit the unknown concentration solution via the in‐house algorithms which is
generated by each type solution.

Protein
MaestroNano only displays the absorbance at 280nm, 260nm, and the ratio of
A280nm/A260nm after the protein sample measured. The absorbance value is
normalized to a 10 mm path length.
TROUBLESHOOTING
If you encounter the following problems, please refer to the suggestion as follows.
For further instructions, please contact with your local customer services.
1. Instrument can not be started up.
Checking item:
(a)Check the instrument is using the correct power socket or not.
(b)Check if the instrument disconnects with PC or not. It should be disconnected.
(c)The LED on the adaptor will be lighting, if the adaptor is working.
2. The measurement result shows “Error: please try again”.
Checking item:
(a)Make sure that the droplet is contacted with upper and lower fiber terminal.
(b)When MaestroNano is in the measurement state, please check if the table is
stable and not shaking.
(c)Confirm the light source remains to be lighting when the instrument is in the
measurement progresses.
(d)Check if the fiber is broken. If the fiber occurs bending, it needs 3~4 weeks for
returning to Maestrogen to replace a new fiber set

3. Printer Error.
The troubleshooting suggestions are listed as follow:
If this error can not be eliminated, please contact with your local customer service.
4. Battery is out of power.
When the battery power is low or out of power, MaestroNano will show low power
message when the instrument is started up. After replacing new battery (Model:
CR2032, 3V), it is suggested to set time first to avoid incorrect time record with
sample measurement.

WARNING:Be careful to handle the outside fiber
Due to the intrinsic property, the outside fiber of MaestroNano is very
weak. Please be careful to handle the outside fiber and follow the notice
shown as below.

DATA access from MaestroNano (USB)
If users want to access measurement results from MaestroNano, the USB
transfer is one solution in addition to thermal printer.
1. Instrument can only be started up without connecting to PC.
2. After measuring and “saving”, do not shutdown MaestroNano, and
connect the instrument to PC via USB transfer. (A male to A male
cable)
3. There is a flash memory appeared in PC (“My computer” in Windows
series). At the same time, MaestroNano can not be operated, and
there is a sentence “USB connection: data access” shown on the LCD
screen of MaestroNano.
4. Copy the data from MaestroNano, and the data in this flash memory
is “Read Only”.
5. Remove the flash memory from software operating system, and
remove the hardware afterward.
6. User can operate MaestroNano to shutdown or measure.
