constants.

Distribution of tissues to the Compartments
Classification of a particular tissue, for example brain, into central or
peripheral compartment depends upon the physicochemical properties of
the drug.
A highly lipophilic drug can cross the BBB, and brain would be included into
the central compartment.
In contrast, a polar drug can’t penetrate the BBB and brain in this case will
be a part of peripheral compartment despite the fact that it is a highly
perfused organ.

Three Compartment Open Model
► The three-compartment model is an extension of the two-compartment model,
with an additional deep tissue compartment.
► A drug that demonstrates the necessity of a three-compartment open model is
distributed most rapidly to a highly perfused central compartment, less rapidly to
the second or tissue compartment, and very slowly to the third or deep tissue
compartment, containing such poorly perfused tissue as bone and fat.
► The deep tissue compartment may also represent tightly bound drug in the tissues.
