Collected bacteria samples (B and C)


Introduction

Bacteria can be found everywhere. It is typically live in water, soil, organic matter or the
bodies of plants and animals. Bacteria also can be found in human maybe in mouth, skin, urine
and other parts of the human body. We can collect the bacteria by using the different materials
such as Petri dish, agar, cotton buds and alcohol lamp. This material will serve for the preparing
of bacteria to grow.

Discussion

Use a sterilized inoculating loop or sterile swabs to collect bacteria from different locations
and then streak each Petri dish with your sample. This involves a bit more technique than the
previous experiment but offers a wider choice of bacteria sampling locations.

Materials

1. Prepared petri dishes containing agar medium and nutrients.


2. Bacteria collected
3. Wax pencil for labeling dishes.
4. Masking tape.
5. Sterile swabs or inoculating loop.
6. Alcohol burner (source of flame to sterilize inoculating loop.)
7. Bleach

Procedures

1. Prepare petri dishes


2. When ready to use, let dishes come to room temperature before taking samples.
3. Collect bacteria from each location using one swab or resterilized inoculating loop) for
each new spot.
4. Inoculate each dish by streaking a pattern gently across the entire agar surface without
tearing into it.
5. Replace cover on dish, tape closed, and label each dish so you know the source of
bacteria. Store upside down.
6. Let grow in undisturbed warm location, ideally in an environment around 100° F (37°C)
– not in sunlight or on a heating register.
7. You should see growth within a couple of days. The dishes will start to smell which
means the bacteria are growing.
8. Make observation and keep records of what you see growing in each dish. Can you make
any conclusions about what locations had the most bacteria?
9. Before disposing of dishes in the trash the bacteria should be destroyed. Pour a small
amount of household bleach over the colonies while holding dish over sink. Caution – do
not allow bleach to touch your skin, eyes or clothes. It will burn!
Observations

First Day (September 03, 2016)

At 12:30 in the afternoon. I put


my urine in the petri dish that have
agar by using cotton buds and gently
strain it to the agar. Nothing
happened the first day.

Second Day (September 04, 2016)

There are a lot of dots and the


urine that I strain in agar is very
visible.

Third day (September 05, 2016)

It is almost the same on the


second day of my observation.

Fourth Day (September 06, 2016)

The dots become double and the


urine that I strain become more
visible.
Fifth Day (September 07, 2016)

On the petri dish that label letter


A the dots disappear. And in letter B
and C it becomes moist.

Sixth Day (September 08, 2016)

When the dots disappear on letter


A it become worm and in letter B
and C become a big Circle that looks
like a mold.

Seventh Day (September 09, 2016)

Its just the same on the 6th day of


my observation.

Conclusion

Based on my observation I conclude that bacteria can grow in agar and I prove that they
can divide themselves.

BACTERIAL GROWTH (A)


Introduction

Bacteria are microorganism that can grow everywhere. It can be found anywhere and also in our
human body. By means of different materials we can grow the bacteria. Agar is really a good
substance that can be used to bacteria for them to grow. It is an excellent medium for supplying
bacteria with nutrients.

Caution

Most bacteria collected in the environment will not be harmful. However, once they multiply
into millions of colonies in a petri dish they become more of a hazard. Be sure to protect open
cuts with rubber gloves and never ingest or breathe in growing bacteria. Keep growing petri
dishes taped closed until your experiment is done. Then you should safely destroy the fuzzy
bacteria colonies using bleach.

DIRECT CONTACT

Discussion

In this type of experiment, bacteria are transferred directly to the prepared petri dish via direct
contact. You can test the effectiveness of different soaps by treating different petri dishes with
“dirty” hands before washing and “clean” hands after washing. Or, you can press a variety of
common objects like coins, combs, etc. on different plates and compare the bacteria growth that
results.

Materials

Prepared agar in a petri dish, bacteria on hands, coins, money bills, etcs.., marker, masking tape.

Procedures

1. Prepared petri dishes should be refrigerated until used and always stored upside down (i.e
media in upper dish , cover on bottom.) this keeps condensation which forms in the lid
from dropping onto and disrupting the bacteria growing surface.
2. When ready to use, let dishes come to room temperature before taking samples (about
one hour.)
3. Without tearing the agar surface, inoculate the dish by gently pressing finger nails, coin
etc onto agar surface. (direct contact of lips or tongue is NOT a good idea.)
4. Replace cover on dish, tape closed, and label each dish so you know the source of the
bacteria. Store upside down.
5. Let grow in undisturbed warm location, ideally in an environment around 100°F (37°C0
– not in sunlight or on a heating register.

the bacteria are growing.

Observation

Based on my observation from day 1 up to day 7 the bacteria I get from my tongue and
put on the agar by means of using cotton buds. On the 1 st day it moist but on the 2nd day up to the
4th day there’s a lot of dots appeared on the agar which is very visible. But on the 5 th day all of
the dots disappeared and become a web like in the spider and there’s a worm that appeared in the
agar as you can in the picture.

Conclusion

I conclude that by means of petri dish, agar and different materials that bacteria can be
grow instantly because agar is very excellent medium for supplying bacteria with nutrients. And
also the environment has a large contribution for the bacteria to grow. Most especially during the
times that you expose it to the sun.

MICROBIAL GROWTH CONTROL (C)

Introduction

Bacteria can grow easily most especially when you put in petri dish that have agar. But also
bacteria can be treated or grow rapidly by means of herbal medicine or different substances like
alcohol, betadine, or even medicine like penicillin and others. In this experiment that conducted
it will see how the bacteria reacted to the substance that was drop in the area that have bacteria.

Materials

Petri dish, Alcohol lamp, Urine and Oregano

Observation

Based on my observation, when the oregano was drop on the area of the bacteria the
oregano was spread all over the whole area of petri dish and the agar absorbs it.

Conclusion

Based on my observation the herbal medicine which is the oregano is cannot treat the
bacteria but it can cause the bacteria to grow rapidly.
