Chapter 1 Cautions
For ensure safety in operating the instrument, please abide by following rules.
1.Power Voltage: AC 220V-240V/50Hz
Do not use the power requirements which specification beyond above range,
otherwise it may damage instrument or result in fire and getting an electric shock. 。
It is recommended to use external electronic stabilizer in the unstable electricity
supply area, to avoid drastic fluctuation of voltage which may affect reliability of
measuring date and damage the high voltage module and photomultiplier tube.
2.Do not share same socket with other unrelated devices!
It is recommended that the power supply of the laboratory be stable. If it is not, the
manufacturer highly recommends using electronic stabilizer.
3.Make sure that the instrument is not subjected to direct sunlight, other
radioactive light, strong magnetic field, strong electric field and other
radioactive substances!
It must be increasing measuring precision and prolonging the lifespan of instrument
if operating it in the lightless laboratory.
4.Do not block ventilating unit, otherwise it becomes damp or overheated easily
and leads to faults!
Do not place the instrument close to heat source such as heating stove, radiator; do
not put any containers filled with water or small metal articles on or near the
instrument. Please pay close attention to keep corrosive liquid and solid substances
away from the instrument.
5. Insure that the door is completely closed while measuring
If the lid of door does not close completely, the measuring instrument in the host
computer will terminate the operation, and a notice will be out by the software. Now,
you should check that whether the lid is closed well and clean the sundries around
the lid up. If the malfunction still can not be obviated, it manifest that the cover is
broken or the hermetic part is damaged, please contact with our after-sales service
technicians.
6.It is forbidden to drop any foreign matters into instrument on purpose!
Once the foreign matter is found in instrument, please pull out the plug from socket
immediately, then one can take it out of instrument, or contact with our after-sales
service technicians if necessary.
7.Avoid collision with other objects!
Do not damage the cover surface and monitor of instrument.
8.Please pull out plug and switch off all electric units relative to the instrument
on condition of thunderstorm weather so as to avoid damaging on instrument
struck by thunderbolt!
9.Do not disassemble, repair or refit the instrument!
Do not disassemble body shell or open its covers casually because of high voltage.
The instrument is of the function of device spoilage by itself when open its cover,
therefore user has to shoulder the responsibility if he disassemble the instrument

unauthorized.
10.Keep the surface of instrument cleanness!
Please pull out the plug from socket before clean the instrument.
Do not clean the cover and monitor with volatile solvents or diluent,vacuum cleaner,
polish and abrasive paper, etc..
User may use soft dry cloth to clean the cover and monitor of the instrument.
Do not clean the surface of instrument with liquid or foggy detergents.
It is forbidden to clean the instrument with detergent directly even the cover is pretty
dirty. User should wring out wet soft cloth to clean the cover first, then wipe it down
with dry soft cloth.
11.Emergency handling!
If smoke is rising from the instrument or strange smell is giving off, user must pull
out the plug immediately and ask for our technician’s help.
If it occurs death or other abnormal phenomena, please cut off power at once. If the
instrument can not works well after reset it, please refers to “Chapter 7 maintenance
& Abnormal handling”, or ask for our technician’s help if the problem is still exist.
If it occurs collision incident or cover is damaged seriously, please pull out plug
immediately and contact with us.

Cautions and Warnings contain important information for the safe operation and use of
this product. DO NOT use this product without first completely reading and
understanding these instructions. If you are unable to understand the Dangers,
Warnings, Cautions or Instructions, contact the manufacturer or an authorized service
dealer before attempting to use this equipment - otherwise, injury or damage may
occur.
When using electrical products, basic safety precautions should always be followed,
including the following.

1) Only trained and qualified medical personnel should operate the device
2) The equipment is rated to operate at an altitude less than 2000 m.
3) Equipment not suitable for use in the presence of flammable mixtures, explosion
gas.
4) Do not use flammable solvent with fire point less than 900C.
5) Replace with same type and rating fuse, fuse specification as below:
F1, F2: AC 250V, F 1A L.
6) WARNING: No use-serviceable parts inside, before servicing to authorized
representative or manufacturer!
Never operate this product if
a) the power cord or the plug are damaged.
b) the device is not working properly.
c) the device has been dropped or damaged.
d) the device fell into water.
e) if the device sounds aloud or sharply suddenly, and if the output air is too hot or
smells bad.

f) the parts/components damaged.
In such cases return the device to the manufacturer or an authorised service dealer
for inspection and repair. Don’t open the device and change any part yourself.
7) If the supply cord is damaged, it must be replaced by a special cord or assembly
available
from the manufacturer or its service agent.
8) The installation category(overvoltage category)for this instrument is Ⅱ,and it is
classified as portable equipment. The instrument has a pollution degree rating of 2
and may
be installed in an environment that has non-conductive pollutants only.
9) Class I appliance, the equipment shall be connected to a MAINS socket outlet
with a protective earthing connection. Do not use an adapter to a two-terminal outlet
in order to avoid electric shock!
A three-pin plug is used in this instrument. A grounding electrical outlet must be
used to satisfy the safety standard.
10) Do not place the equipment on floor, put it on desk.
11) Do not operate this device in a stationary recreational vehicle, boat, or motor
home or similar places. Keep the device away from children.
12) Never insert hands or fingers into the contact fluid tube, container while the
device is operating.
13) Do not use accessories not recommended by the manufacturer.
14) Place the device on a flat and solid surface.
15) WARNING: To avoid electrical shock, always unplug the device power cord
before cleaning the device.
16) Minimize contact with chemicals. Wear appropriate personal protective
equipment when handling chemicals (e.g., safety glasses, gloves, or protective
clothing)
17) Minimize the inhalation of chemicals. Do not leave chemical containers open.
Use only with adequate ventilation
After emptying the waste container, seal it with the cap provided.
Dispose of the contents of the waste tray and waste bottle in accordance with good
laboratory practices and local, state/provincial, or national environmental and health
regulations.
18) Some of the chemicals used with this instrument may be listed as hazardous by
their manufacturer. When hazards exist, warnings are prominently displayed on the
labels of all
chemicals.
19) Samples containing material of human origin must be treated as potentially
infectious.
The used lancet and test strip are biohazard materials which should be disposed of
properly to avoid injury or contamination.
If a sample is spilled on the device, wipe up immediately and apply disinfectant.
Handle waste properly, according to legislation on water pollution, and on the
treatment of drainage and waste matter.

2.1 Overview

Firstly, Thanks for your selection of our state-of-the-art equipment:


QQ-96(MLD-2000A) Chemiluminescence Immunoassay Analyzer. For operator’s
safety and to make the most of performance of instrument and prolong its lifespan,
please read the User’s Manual and other relative materials carefully before operating
it. Please contact with us at any time once the problem is occurred during operation.

2.2 Working conditions:


Ambient temperature: 10~30℃ ;
Relative humidity: ≤85%;
Atmospheric pressure: 86~106kPa;
Keep the instrument away from strong magnetic field, strong electric field, heat
source, water source and corrosive liquid.

2.3 Handling of waste:


Residual solution from washer and used reactive plate need careful and skilful
handling after testing so as not to harm human beings or pollute environment.

2.5 Intended use and suitable range for reagent:


Based on single photon counting principle,
QQ-96(MLD-2000A)Chemiluminescence Immunoassay Analyzer is the fruit of
advanced CLIA technology, which can performs the analysis of chemiluminescence
immunoassay, cell, microorganism luminescence, and enzyme activeness testing of
ATP, NAD (P) H etc. so as to support clinical diagnosis. Within these ranges,
microplate as the solid phase or reactive system and, reagents of unnecessary
original sampling chemiluminescence and biological luminescence are suitable.

2.6 Principle
Chemiluminescence Immunoassay analysis consists of immune response system and
chemiluminescence system. With the same principle of ELISA, Chemiluminescence
response system always adopts DAS-ELISA, competitive and indirect method as
reactive model, see Figure 2-6-1, 2-6-2 and 2-6-3:

The principle of chemiluminescence lies in enzyme during immune response acts on
luminescent substrate, which occurs chemical reaction and releases a great quantity
of energy, then produce singlet state medium that emit photon at the same time when
it back to the stable triplet state. Capitalized on luminescent signal device one can
measure optical quantum yield, which forms direct proportion with quantity of
awaited substance in sample. Therefore it can establish standard curve and calculate
out the content of awaited substance in sample.
We adopts the method of catalysis Luminol by HRP, see Figure 2-6.4

Currently, compare luminescent substrate developed by Chinese manufacturers with


similar products produced by foreign manufacturers, its main performance has
reached the same level of advanced products and cost decrease dramatically. In this
luminescent substrate system we select special complex potentiator, which enjoys
fast luminescence, high intensity and long luminescence period that can last from 30
to 60 minutes, so it totally meet the needs of clinical measuring. Test result of its
luminescence period is showed at Figure 2-6-5.

Additionally, it is of precise three-dimension drive unit, sensitive photon counting
system, complete mathematic model database, which used to complete positioning,
testing and data analysis of all wells of ELISA plate so as to ensure stability and
accuracy of measuring results.

Chapter 3 Structure Features and Working Principle

3.1 Structure Features

The outlook of QQ-96(MLD-2000A) was carefully designed by the esthetics expert,


according with the principle of anthropological engineering science. Highly integrate
chip and function unit inside the crust.

There are 2 keys and 3 indicators on the front board.
POWER key :turn on or off power supply.
PAUSE key: pause the working.
POWER indicator: Turn on the POWER key, the POWER indicator will light up.
GATE indicator: when the door is opened through fair and foul, the GATE
indicator will light up and the instrument will can not work or stop working.
WORK indicator: the WORK indicator will light up when the instrument is
working.

3.2 Working Principle
Measuring photons part of QQ-96(MLD-2000A) is single photon register.
The core part is a special photomultiplier tube which is a super high vacuum
column terrarium.
The phonic side (window) which covered with special rare metals could produce
photoelectric effect was called light negative polar. Inside the photomultiplier tube,
there are many of electrode which was arranged by special manner, we call it
accelerating polar or beat polar. The other side of photomultiplier tube was positive
pole. The poles we stated above all have high current voltage. When photon hit the
negative pole, dissociate electron (photoelectron) have faint energy was bright
because of photoelectric effect. High current voltage compels photoelectron
leaving negative pole then hit the first acceleration polar again, photoelectron that
get adding power provided by the high current voltage became new electron with
more power and larger magnitude. time and time again ,under the reaction of
acceleration pole ,positive pole give birth to a electric pulse signal have more
power compared to the first sample electron on the surface of positive pole .the
amplified signal was counted by the frequency machine after wiping off yawp. The
counting result was quantity of photon impulse which is RLU ( Relative
Luminescence Unit)。Generally speaking, one RLU equal to ten photon units.

3.3 Operation
1. First confirm the instrument have connected with electrical supply reliably.
2. Press the POWER key. POWER indicator will light.

3. Turn on the computer and monitor. Run our special software for CLIA.
4. Open the door and the GATE indicator will light.
5. Put the 96holes plate awaiting on the board inside the instrument.
6. Close the door and the GATE indicator will quench.
7. Through the special software for CLIA control the instrument to work. When the
instrument is working the WORK indicator will light. After finishing work, the
WORK indicator will quench.
8. Analyze the date from the instrument through the special software.

Chapter 4 Installation

4.1 Opening the package and checking-up


Ripping the seal upwards with the paper knife, taking out the instrument
carefully, and examining whether the adjunct and collocate are completely according
to the checking and accepting list. At last, delivering it to the installing engineer after
the signature of the principal of the section office.

4.2 Installation brief introduction


The System composing of the mainframe of the CLIA 、 computer
mainframe、monitor and printer.
The gradient should less than 45 degrees when transporting or transiting the
instrument, and trying your best to avoid violent shaking and moisture atmosphere.
The instrument should been installed on a actinic stabilization table which is been
ventilated and keep away from the point-blank light.
The electrical source which the instrument used must be the standard power of
220-240V/50HZ which has earthing line, and the earthing line must contact with the
earth very well, and the voltage fluctuate range should be less than 10%,if you
can’t meet the requirement you must install the manostat.
4.3 Installation procedures
Put the instrument on the steady working platform, tear off the seal ,open the
door , unlade the crew which can fixup the salver with screw driver ,take out the clip
and close the door .
Connect the computer to the analyzer and make sure the electric source line in
its position. Finally, linking electrical source line and printing cable line of the
printer (not belong to the basic equipment of QQ-96(MLD-2000A)).the printer must
tally with EN60950 stipulation of the <<security of information technology
equipment>>.connecting the three electrical source line to the power supply socket,
the socket must connect to the earth safely.

Attention: every cable connecting to instrument must be safe、 fastness, and


good contract; especially communication interface must contract without error and
reinforce bolt .you can use the instrument after confirming the connection is without
mistake.
This instrument belong to CLASS I instrument, so the socket must connect to
the earth safely

Chapter 5 Software

5.1 Introduce
Controlling the QQ-96(MLD-2000A) to work by using this software which is
resident in the computer,

5.2Installation of software
It divides into two cases: one is new installation and other is update installation. New
installation means first install this software; whereas during use the software, there is
a updated software is released, which is called update installation.
5.2.1 New installation
Step 1: Switch on the computer and insert the installation CD of Chemiluminescence
Immunoassay analyzer in CD-ROM, then press mouse with two clicks on CLIA
SETUP. EXE to install software. However to better protect CD-ROM and prolong its
lifespan, we recommend to copy file of CLIA SETUP. EXE to D disk of computer or
to lower-level file, then press left part of mouse with two clicks to process the
installation procedures of CLIA SETUP. EXE, after that it starts to install software
and several seconds later screen shows:

Step 2: Press “Next” in this interface to continue install the software, then it
shows:

Should any problems occurred during installation, please press “Cancel” to exit(the
same below).
Step 3: Please make a decision after carefully read the License Protocol. If there is
certain clause you can not accept or any contents exits confusion, please select “ I do
not agree upon the clause in License Protocol(D)” and press “Cancel” to finish the
installation procedures; otherwise choose “I agree upon the clause in License
Protocol(A)” and press “Next” to continue installation, then system notes user to
input user’s information.

During installation, if user has to perform certain modification on inputted
information at previous menu, one can press “Previous” and modify it.
Step 4: Input user’s name and name of institution: “MS User” is defaulted by system,
one may modify it according to user’s actual situation.
Option of “User of this application program”: “All persons can operate this
instrument A(all users)” is defaulted by system; one can also select “Only for oneself
(M)(User)”, so other persons can not perform this software. Then press “Next” to
continue installation and one may select installation type as follows:

Step 5: It is highly recommended to choose “Complete installation”, press “Next” to
continue installation

Superior user may select “Self-define” and press “Next” to process self-defined
installation.

In this window user can check the installing path and needful room of hard disk.
Generally speaking, it defaults installing path as C:\Program Files\KPSON\CLIA\,
or user can modify that by press “Change…” with click mouse one time, then it
shows “Change current file destination” interface as follows:

Back to previous interface by press , or establish a new file with .

Modified installing path is displayed in the column of File Name. Press “Ok” to
save the modified information, otherwise click “Cancel” to get back the menu of
5-2-7.
It may take up about 31MB disk space for complete installation. User may observe
the total size of disk, free space and minimum space required by software installation
with press “Space” button.

Return to the menu of 5-2-6 by press “Ok”.
While low disk space is displayed, one can clear disk space or press arrow in

to reduce some functions in order to decrease minimum

Step 7: Press “Finish(F)” to complete the installing procedures and the software is

installed successfully, at the same time one can see the logo on the table.
Software can be initiated by click this logo two times.

Step 8: At the first time of using this software, it shows “Please Input Unlock Code”

Unlock cord is provided by distributor or service center authorized by our company.

Step 9: It shows “Doctor Log In” , and password should be inputted:

The system defaults password is 111111 and user can reset it at the menu of Doctor

Management, then press “Ok”, it enters the main menu.

Then press , it shows system setting:

Input hospital’s name and registered code that is provided by our company, then

press “Ok” to complete the registration of software so as to ensure normal operation.
Each instrument has different registered code, which is provided by distributor.
5.2.2 Update installation
Step 1: When there is a new software is released, it can process new version
software either by press download list or with the file of CLIA SETUP. EXE in CD
supplied by us. It shows:

By the way, when current software would not be used any more, one can cancel it

Main Menu is composed of Function buttons, display of Plate information area,
Toolbar, Curve display area, Well information and reagent list areas. In the next
paragraphs all parts would be explained in greater details respectively.
At the top of Main Menu is Function area, which includes eight menus of Doctor,
Operation, Case, Reagent, Plate, System, Chart, Help and ten function buttons of
Doctor Log In, Establish Plate, Clear Plate, Save Case, Case Inquiry, Save & Print,
Doctor Management, Reagent Browse, System Management, System Help. These
could perform the setting of log in, system, user’s information, Plate information and
reagent information. At the lower part of Main Menu is plate information area, which
shows 96 wells with six rows and twelve ranks corresponding Elisa Plate and its
corresponding well type and reagent information that at the current position of plate
coordinate. Toolbar, at the lower right of Plate information area, is consists of
Standard, Q.C., Compound, Patient, Negative, Blank, Extract, Assign, Concentration
and Measuring. Lower left of Plate information area is Curve display area, which
shows reagent’s luminescent value(or concentrated curve) measured according to
reagent standard, furthermore it displays tips and measuring status in the process of
measuring. Under the toolbar is display area of Qualitative reagent, Well information
and Quantitative reagent. Well information, at the position of cursor pointed,
includes well number, well type, luminescent value and unit time; name and judged
formula of Qualitative reagent are displayed at the column of Qualitative reagent
sheet; Quantitative reagent is valid information of item’s curve extracted from
experimental reagent standard saved before.

Save case: Save case for future reference.

Case Inquiry: Inquire about saved patient’s case.

By press the arrow beside of Inquiry Item, it includes Name, Case No., Outpatient
No., Measuring Items, Operator, Policy No., Sender, Sender’s Sector, Sender’s No.,
Bed No., Sample No.. According to different requirements, operator can choose
suitable inquiry item, and input known inquiry condition, then press inquire button.
Or if necessary one can perform inquiry by input limited period, but before that one

has to put a tick on , otherwise inputted limited period is invalid.


Furthermore, one can input “%” in the column of Inquiry condition to process
obscure inquiry, and browse all patient’s cases.

Save & Print: the function of this button is similar with Case

Inquiry. The difference between these two buttons is directly select out

frame of Case Inquiry; whereas is first save current patient’s case, then
select out frame of Case Inquiry.
④ Reagent

Extract reagent: after complete measuring well position of awaiting sample


in Plate information display area shows luminescent value of awaiting sample. One
can select reagent’s standard well positions (Generally are six) which are complete

test, then press it shows the frame of “Add new reagent”

Input new reagent’s information one by one and select suitable fitting algorithm,
then press “Ok” to save the reagent. Save measured luminescent value of standard
reagent so as to use again in homogeneous experiments(within reagent warrantee
period).
Select optimum fitting algorithm is based on:
a. Fitting curve is of monotonicity. Of course, concentration and luminescent value
of several standard samples must have monotonicity and non-reproducibility,
otherwise it cannot fitting curve ;
b. Fitting curve should be as smooth as possible;
c. Measured several points should be fallen on the fitting curve, or equally scattered
at two sides of curve.

Assign: respectively select patient sample and Q.C. standard sample well
position of required fitting data after extract reagent, and choose corresponding

reagent’s name from reagent sheet, then press . Or press arrow to assign
certain range if necessary, after the assignment procedures are completed it starts to
calculate concentration.
If the range has not be added, one has to do it in the menu of Add Reference Range

Input Range Name, Reference Lower Limit, Reference Upper Limit, then press
“Ok”.

Concentration Calculation: after press this key, it shows concentration of


patient’s sample on the lower part of patient’s sample well position through
calculation handling. If it shows

It gives a notice that is “some well(s) haven’t been assigned or their RLU is zero,
please check green well position”. At same time it indicates unselected well with
green frame. Then close this warning frame of “Cannot fitting”, and perform
corresponding operation according to tips.
Edit Reagent: Edit or modify information of current reagent at the position of cursor
pointed.

One can modify Reagent Name, Abbreviation, Concentration Unit, Valid Days,
Single-well Time, Remark, etc. in above menu; the function of “Add Range” here is
the same as add range in the menu of Assign reagent. To expired or wrong reagent
range one can select it with mouse, then delete it by press “Cancel Range”.
Finally, the function of “Fill In Plate” is when add given reagent standard, fill well
information of several standard in current plate in order to process contrast.
Cancel Current Reagent: delete current reagent standard at the position of cursor
pointed. It shows

Press “Yes” to delete, otherwise press “No”.


⑤ Plate

Establish Plate: after press this key, it establish a blank plate, in addition add
the number of this plate in the menu of Plate. For instance, the number of newly
established blank plate has been ticked off, see following Figure

Among them, there is a tick before 2006-01-06 0004, which means it is the current
plate file.

Clear Plate: clear current plate by press this key, it shows

Clear current plate by press “Yes”. Before clear out it please confirm the useful
information in the plate is saved, since these information would be lost for good after
clear out, otherwise press “No” to delete current procedure.
Notice: Although the operation of “Establish Plate” and “Clear Plate” turns
current plate into blank one, “Establish Plate” would not change the original
plate and establish a new plate file; however “Clear Plate” is based on original
plate to clear out original data and turns it into blank plate. The difference of
two buttons is number of current plate while operation is completed.
Copy patient’s information from other plate: it can copy patient’s information from
saved plate file to the same position of current plate, and original data which place
has been changed would be replaced. Therefore please carefully read the warning
before make a decision.

Press “No” to delete current operation, or press “Yes” to continue.

Open plate file: it can open saved plate files, and system automatically defaults the
path as C:\Program Files\KPSON\CLIA

Notice: Although the operation of “Copy patient’s information from other
plate” and “Open plate file” can transfer saved information of plate file to
current plate, however, the operation of “Copy patient’s information from other
plate” is copy saved plate information to current plate; and “Open plate file” is
directly open saved plate file. The difference of two buttons is number of
current plate while operation is completed.
Automat luminescent value: Put luminescent value of current plate into Excel.
It defaults C:\Program Files\KPSON\CLIA as save path, which could be assigned by
user, then input file’s name in the column of file name and press “Save”.
2005-12-13 0006: it is the number of plate file. Once a new plate is established, it
will automatic produce a corresponding number, from which front part is the product
date and latter part is different plate number produced in the same day.

Once establish or open a plate, its number appears in the menu of Plate. User can
change different plates by press plate number with mouse. Number with tick
indicates this is the current plate.
⑥ System
System management: specialized in registration of this software. At the first time of
using this software, one need to input institution’s name and registration code.

Confirm the name and code is correct before press “Yes”, then it shows

It means the software is registered successfully and could be use; if it shows

It indicates wrong registration code, user has to register again.
Hospital name should be corresponding with registration code, and registration code
is automatic produced from software registration system of our company. System
could not work normally if software registered failure, and user has to contact us to
get the registration code.

Compound well position is generally used on condition that the measuring results is
strictly required. Take mean value from several well positions with same reagent type
in order to make measuring results much precise. Detailed steps are as follows: pull
certain well position with mouse to a well position that has been set as reagent
standard or patient’s sample or Q.C. standard, then this compound well position
establishes relations with the target sample, several compound well positions can
establish relations with certain target sample simultaneously. After complete test, it can
obtain mean value of measuring results of one or several well positions.

6.2 Gist of sampling experiment


Carefully read the reagent instructions before operation. Strictly abide by operating
steps and requirements during experiment; at the same time one must observe safety
regulations in laboratory. It is place emphasis on frequent questions as follows:
① Sample collect
User should collect sample with clean disposable blood collection tube, and must
do individual protection during blood collection, such as wear glove and gauze
mask etc.. Any waste must be destroyed according to relative law strictly. Only
use vacuum blood collection tube contains resolving gel, otherwise immediately
separate serum with centrifuge. Be sure to test collected sample as quickly as
possible, and it can be preserved at 2~8℃ if storage time is less than 24 hours;
frozen it under -20℃ for long-term storage and avoid frequent frozenness and
reconstitution. However certain hormone, tumor marker and medicine etc.
combined with serum protein may result in changes to measuring results before
and after sample freezing storage.
Severe hemolytic sample, chyle blood sample and the sample has been polluted by
microbe can not be used for measuring.
② Prepare reagent kit
Reagent kit should be preserved in refrigerator with proper temperature according
to reagent instructions and used up before expiry. Take it from refrigerator before
use and keep it for a while under room temperature( see details in reagent
instructions). Dissolve or dilute concentrated solution with distilled water in
accordance with requirement before use; To certain frozen standard or Q.C. in
reagent kit should strictly add distilled water in accordance with required volume,
after light mix up and keep it motionless for 15~20 minutes, one can use as it
completely reconstitution; Make sure that the luminescent substrate solution and
enzyme marker is not subjected to direct strong light.

Do not use different batches of reagent in one quantitative experiment. To
guarantee reliable results, simultaneous sampling and measuring among standard,
Q.C. and patient’s sample in experiment, it is better to perform complete standard
curve at every test. If it is use the curve established and saved by user before, one
has to perform curve calibration, which method can be found in reagent
instructions. Generally speaking, standard value of using calibration point should
be as close as possible to normal value; at the same time, reagent kit with
double-side normal value, such as most part of hormone and medicine etc. must
select two-point calibration; whereas the reagent kit with single side, such as part
of tumor marker can choose single point calibration.
Take reagent kit back to refrigerator as quickly as possible after use. Do not keep
it under room temperature for long period. Unused micro well strip that is
unlocked should be sealed again and preserve it at 2~8℃ along with other parts
of reagent kit.
③ Sampling
All clinical samples should be treated as infectious samples, so one must do
individual protection such as wear glove and gauze mask etc.. Any waste must be
destroyed according to relative law strictly.
Any sampling volume must strictly abide by requirements written down in reagent
instructions. Add reagent against the wall of well in plate so as to avoid splash and
make air bubbles; and use new aspirating head each time to avoid carryover.
For ensure identity of each sample well on reaction time, it should shorten
sampling time during add sample and reagent. It is recommended to add
marker(enzyme compound) or capture antibody first, at this moment every well is
blank and added reagent is same, so it is possible to use same aspirating head to
test every well, which both economize consumables and save the trouble of
changing aspirating head, therefore save time also. Under this condition we
suggest you use continuous sampling unit or sampling unit in rows in order to
raise working efficiency. Then add standard, Q.C. and patient’s sample in
corresponding well, when add patient’s sample user has to replace new aspirating
head after adding one sample so as to avoid carryover; However for adding
standard and Q.C., if it starts sampling from lower content, that is in A, B, C
sequence, then it is unnecessary to change aspirating head every time.
To avoid evaporation or overflowing in the process of vibration and incubation,
and drop out impurity or polluted each other, one should seal up microplate with
cover membrane after sampling, then fast fix the microplate on oscillator to start
incubation. If there is not incubation device, one should put microplate on
platform to vibrate, or mix up with hand before put into incubation.
④ Incubation
Make sure microplate always keep horizontal status during incubation, and
incubation time must strictly abide be the requirements in reagent instructions. If
use micro oscillator, it must ensure its proper oscillatory amplitude, which both
makes solution in microplate fully mix up and not splash solution on cover
membrane. If adopts thermotank, it must ensure microplate is even heated, at the

same time its temperature fluctuation would be controlled within ±1℃. If use
constant-temperature water bath box, one should use distilled water as medium
and replace water frequently in order to avoid turns rotten and turbid, meanwhile
it is better to keep the level of solution under half place of microplate flank wall.
Do not submerge the microplate.
⑤ Washing
Washing type and times should strictly abide by reagent instructions, and use the
cleaning solution provided in reagent kit. Make sure all components fully
dissolved when produce cleaning solution. It would be best to produce cleaning
solution when it is used, if use the cleaning solution that is produced before, which
should be ensure within storage shelf-life and no turbid phenomenon. Make sure
fill in every well position with cleaning solution during washing. If wash it by
hand, one must ensure to exert same strength on every washing and spin-dry; if
use automatic washer, one should check every wash well and tubing at regular
period so as to avoid blockage or getting fungus.
⑥ Add luminescent substrate solution
Firstly pat the microplate to dry with clean water absorbed paper after washing,
then take an appropriate amount of luminescent substrate solution and add it in
corresponding well position of microplate. Sampling volume must strictly
according to reagent instructions. Use new aspirating head to avoid influence by
polluted impurity. Add solution against the wall of well in plate so as to avoid
splash and make air bubbles. For ensure identity of each sample well on reaction
time, it should shorten sampling time during add luminescent solution. We suggest
you use continuous sampling unit or sampling unit in rows in order to raise
working efficiency. Microplate should be put in measuring room immediately
after add luminescent substrate solution to test its luminescent value, however its
optimum measuring time is within 10~20 minutes after add luminescent solution.
Measuring time of every well must strictly abide by reagent instructions.

6.3 Gist of software operation


Carefully read the reagent instructions before operation. Strictly abide by operating
steps and requirements during experiment; at the same time one must observe safety
regulations in laboratory. It is place emphasis on frequent questions as follows:
① Software setting
Open software on table by press shortcut key, then perform corresponding setting on
property of every well position according to plate layout during sampling. Quantitative
experiment includes standard setting, individual and its corresponding concentration,
Q.C. standard setting, individual Q.C. range that is Q.C. maximum and minimum limit,
position of patient’s sample, individual and patient’s information etc.. For obtain
precise measuring results, one has to correctly input concentration of every well and
its relative information.
② Luminescent value measuring
Theoretically, Microplate should be put in measuring room immediately after add
luminescent substrate solution to test its luminescent value, however its optimum

measuring time should be reference to reagent instructions. When measuring several
microplate at the same time, one should make sure the identity of time from add
luminescent substrate solution to measure luminescent value in every plate. User
should switch on the power of analyzer before measuring, and stabilize microplate that
is complete handling of sampling on tray of analyzer. Close the lid of door of analyzer
host well. Do not perform other operation on software during measuring. Do not
execute other work or process other program with the computer of analyzer in process
of measuring. Room temperature should be controlled within 20 ~ 30℃ when
measuring luminescent value. Keep it away from strong light, strong magnetic field or
other radioactive rays. Operation and maintenance of analyzer should strictly abide by
user’s manual. Please consult with specialized person in time if it occurs fault. It is
forbidden to operate the analyzer with fault.
③ Data analysis and standard extraction
After measuring, one has to perform preliminary analysis on measuring data. At
beginning check several standards’ relationship between luminescent value and
concentration. There are two requirements, firstly, luminescent value changes with
monotonic increasing or decreasing according with concentration’s monotonic
change; secondly, if two well positions with different concentration, their
luminescent value is variant. To luminescent value of Q.C. standard, which should be
within the range of Q.C.. There are two probabilities if measured value is beyond
Q.C. range, firstly, it may make a mistake on preceding sampling procedure or
software setting; secondly, reagent may get invalid or polluted, if so user has to
replace new reagent. It temporarily not analyze patient’s well position.
④ Reagent standard extraction
When extract reagent standard, one select and only select valid standard well position
that need to be extracted, and press “Extract”, then choose to add new reagent or
replace current reagent in reagent warehouse according to tip, and complete fitting
curve in accordance with further tip. If use curve that performed and saved a few days
ago as current experimental curve instead of reagent’s curve that is measured with all
standard taken from reagent kit, one must makes calibration with single or double
point to this curve. To ensure measured results with clinical sense, standard value of
calibration should be as close as possible to normal value of reagent kit. In principle,
reagent kit with two sides normal value, such as most part of hormone, medicine etc.,
must use two point calibration; whereas reagent kit with single side normal value, such
as part of tumor marker, can use single point calibration.
⑤ Assign reagent range
When assign the reagent, to reagent only possesses single reference range of normal
value, one can press “Assign”; whereas to reference range of normal value that is
divided into several ranges according to different period or other conditions, one
should t o press arrow beside of “Assign”, then select detailed reference range of
normal value, which should be added according to regent instructions. One thing need
to be pointed out is the assigned reference range of normal value is displayed in
patient’s case report; to well position of patient’s sample that detailed range is
selected by press arrow beside “Assign”, name of detailed range and reference

range of patient are displayed in patient’s case report; if well position of patient’s
sample is assigned by press “Assign”, then all names of reagent range and its
corresponding ranges of reagent are shown in patient’s case report.
⑥ Concentration calculation and report formation
It is complete the concentration calculation of patient’s sample by press “Calculate
Concentration”, then one can check the measuring results of patient in print report or
case inquiry. Doctor refers to formed report and reach a diagnostic conclusion
according to patient’s detailed clinical behavior and corresponding symptom.
To further raise clinical sense of measured results, we suggest that based on reagent
kits provided by manufacturer all hospitals establish reference system of normal value
that is more precise, practical and beneficial to clinical diagnosis by themselves.

Chapter 7 Maintenance

Daily Maintenance
1. keep the cover clean, use soft calico to wipe it , and dip some water if
necessary.
2. please take the reaction plate in the tray out after measuring, the reaction
plate must not leave in the instrument beyond 45 minutes
3. If any water liquid in the analyzer, please blot it up by bibulous paper.
4. operation temperature within -10~40℃, relative humidity is less than
85%
Monthly Maintenance
It is no need to Monthly maintenance, if the analyzer is under routine use; if not ,it
need once per month(the way as daily maintenance)
Quarterly maintenance
To check the excursion of the analyzer (once per quarter), the way is to measure
white vacant plate, the signals of every well do not exceed 2000 RLU means that it is in
working order.
Annually maintenance
Do as procedures list above, no need annually maintenance.
How to replace fuses on rear cover.
Changing fuses
Unscrew the cover of fuse holder at the rear side of the device . 2. Remove the faulty
fuse. The fuses are identified on the fuse body and adjacent to fuse holder. 3. Insert a
new fuse. Always use approved fuses. 4. Screw the fuse holder cover. 5. Perform a
functional check as the above 4.3 section.

Chapter 8 Package, storage and transportation


Package: Chemiluminescent Immunoassay instrument is packed in plastic bag,
which is fixed in the paper box.
Storage: packed instrument should be stored within -10~40℃, relative humidity
is less than 85%, and the room is without corrosive gas and with good ventilation.
Transportation: pay close attention to damp-proof, shock-proof, gradient should less

Chapter9 Common faults and troubleshooting
For user to use the QQ-96(MLD-2000A) Chemiluminescence Immunoassay
Analyzer produced by our company well, we will list the common malfunctions、
analysis and handing .Through the illustration, the user can learn the
inner structure and working principle ulteriorly.
Phenomenon 1: the indicator is on after switch the analyzer on, but it can not works
well.
Analysis& Handling:
1. The power supplied without voltage or the socket is loose. To check the voltage of
socket and the connection between plug and socket, and insert the plug into the socket
again.
2. Fuse is burned out. There are two fuses at the back of analyzer, have a check of
them, if it is burned out, then replace fuse.
Phenomenon 2
Indication of error appeared after opening the software to measure

Analysis& Handling:
1. It is probable that the power of analyzer is off , please turn it on
2. Communication cable is not connected or loose, Reconnect the communication
cable ,then screw on two fixed screws of connector of communication cable.
3. Software is not registered or registered information is error, one has to register
again.
4. The main board does not work, please connect the manufacturer to change main
board.

Phenomenon 3 All well positions’ luminescent value are as low as local noise
during measuring.
1. No plate or the plate in the improper place. Please put into the plate accurately.
2. The reagents is out of date or some reagents missed during experiment. Please
use effective reagents to experiment.
3. The move part of analyzer does not work well, please connect the manufacturer
to change circuit board.
4. The photon measuring unit does not work, please connect the manufacturer to
change photon measuring unit.
Phenomenon 4: menu is fixed as follows, and the analyzer can not work

Analysis: one may press mouse on current menu when software is processed here,
which initiate the function of menu lock.
Handling: press mouse again on current menu.
Phenomenon 5: some or all well positions’ luminescent value are rather high during
measuring.
Analysis: the lid of door is not completely closed during measuring; external light
continuously irradiate on the cover of instrument or the instrument neighbors upon
strong electric field, strong magnetic field or may influenced continuous disturbance
from strong electromagnetic wave.
Handling: firstly, be sure the lid of door is closed well, then observe there are
equipments or apparatus that could irradiate strong light, electric field, magnetic
filed and electromagnetic wave exist or not around Chemilumeniscence
Immunoassay analyzer, if so please switch them off, or at least turn them off during
measuring . Then initiate system again and perform measuring.
Phenomenon 6: luminescent value of certain well position is rather high during
measuring, and these positions have not any distributed regularity on ELISA plate.
Analysis: voltage is unstable, or other apparatus that is frequently turn on and off
share same socket with the instrument; or disturbance from strong light, electric field,
magnetic filed and electromagnetic wave around the instrument.
Handling: firstly, check the voltage that should be meet the demand of about
220-240V/50Hz, we highly recommended to equip with a stabilizer for this

instrument. Do not share same socket with other equipments.
Phenomenon 7: After initiate measuring, it sounds abnormal snapping or something
crashing in reagent door.
Analysis: Place ELISA disk improperly, or accident is occurred during
transportation.
Handling: Turn off power and check the ELISA disk is stable or not, then initiate
system again to measure, if it still can not work, please turn power off and stop
operating and contact with our technicians.
