0 INTRODUCTION

Filtration is basically to separate solid-liquid mixture by using a membrane filtration.


There are two type of filtration which are conventional and cross flow filtration. Tangential
flow filtration (TFF) is one type of cross flow filtration. The different between these two
filtration is the flow of feed through the membrane filter. Membrane filtration is a separation
technique that widely used depending on membrane porosity, it can be classified as a
microfiltration or ultrafiltration process. In this lab, ultrafiltration is used in the TFF.

Tangential flow filtration (TFF) is a rapid and efficient method for separation and
purification of biomolecules. It can be used to fractionate large from small biomolecules,
harvest cell suspensions, and clarify fermentation broths and cell lysates [ CITATION Lar18 \l
17417 ]. TFF is use as fractionation of large from small biomolecules, clarification and
removal of cells as well as cellular debris from fermentation or cell culture broths [ CITATION
Lar18 \l 17417 ]

The basic flat sheet tangential flow unit consists of a shallow channel with rectangular
flat sheet membranes on one or both sides (Raja Ghosh, 2006). The membranes are supported
on grooved plates which allow easy collection of permeate. The feed is pumped into the
channel and permeate which crosses the membrane is collected by the grooved plates. The
advantages of the flat sheet tangential flow membrane module is the ease of cleaning and
replacement of defective membranes. To add to that, it have ability to handle viscous feeds
and feed with high levels of suspended solids. TFF membrane also can be used for
ultrafiltration, microfiltration or nanofiltration.

In this experiment, yeast suspension is use in the TFF. In the yeast suspension
contains high solid particles. Thus, using TFF it can filtrate the pure yeast liquid and can use
for further experimental usage.
OBJECTIVES

1. Study on Tangential Flow Filtration and perform the procedure.


2. Identify the components of TFF membrane filters
3. Explain the principle and operation of TFF membrane filters
4. Understand correlation of concentration polarization between flow rate and trans
membrane pressure.
