gravimetry. Describe each procedure comprehensively.
1. Microbiological Water Testing Water microbiological testing is carried out through particulate gravimetry. An adequate sample volume is passed through a sterilized0.45-μm membrane filter for the analysis of coliform bacteria. Then the filter is placed on a sterilized absorbent pad saturated with a culture medium and incubated at 35 ±0.5 ° C for 22-24 hours. The presence of individual bacterial colonies that develop during the incubation period indicates coliform bacteria. 2. Chocolate's crude fat content For the quantitative analysis of food samples, various standard approaches are based on the calculation of the mass of the sample after selective solvent extraction. By extracting with ether for 16 hours in a Soxhlet extractor, the crude fat content can be determined in chocolate. The ether is allowed to evaporate after extraction is complete, and the residue is weighed at 100 ° C after drying. This analysis was also conducted indirectly by measuring a sample with supercritical CO2 before and after extraction.
