2.1 Unpacking

The analyzer is packed in three boxes: one box each for the main unit, the
sample loader and the accessories. The sample loader is available in two types:
the 90SL and 290SL. The accessory kit for the sample loader is packaged with
the sample loader.

Two or more people should unpack the unit. Open the boxes in an area with
plenty of free room and take great care not to hit the boxes against benches, etc.

First, check that the outside of the box is not damaged (e.g., dents). Next, open
each box and make sure that the main unit and sample loader are not damaged
in any way.
Also, make sure that none of the accessories are missing by matching the items
against the list provided in the next section.

2.2 Accessory Inspection

The analyzer components are packaged separately and consist of: the main unit,
accessories, and the sample loader. Two sample loaders are available, 90SL
with 9 racks and 290SL with 29 racks.
The packaged system consists of three boxes containing the main unit,
accessories, and the sample loader. Each component comes with the
accessories indicated below. Verify that all accessories are present.

Installation Environment

Install the unit on an even table top without direct sunlight, air currents,
poisonous gases, dust or vibration.
Operate the unit under the conditions indicated below.

Environmental conditions

Temperature: 15 °C ~ 30 °C
Humidity: 40 % ~ 80 % R.H. (without condensation)
Dust: About the quality in an office
Maximum Altitude: 2,000 m

Caution Do not use in an environment with drastic temperature


differences. Such an environment can cause condensation
to form, resulting in short circuits or improper functioning.

Environment of Transportation and Storage

Transport and store under the following conditions.

Temperature: 5 °C ~ 50 °C
Humidity: 80 % R.H. or less (without condensation)
Other: Keep dry and store indoors

The analyzer should only be moved by two or more people using both hands to
grasp the bottom section of the analyzer (Fig. 2-1).

Required Installation Space

Refer to the figures below and be sure to secure sufficient space around the
main unit to prevent blocking the fan. Also, secure a height of about 880mm,
equal to 400 mm + the height of the main unit (480). In addition, avoid direct
ventilation from other instruments.

Installation of the Main Unit

The main unit has to be carried by two people standing on opposite sides of the
box. The destination bench must be flat and have enough strength to hold the
weight of the main unit and the sample loader. Connect the waste tubing in
accordance with the procedure described on the following page.

Next, open the blue door on the left side of the main unit and the door of the
column oven located below the screen. Use a screwdriver (+) to loosen the three
screws holding the needle cover (the transparent cover of the sampling unit) and
remove the cover by pulling it forward. Reattach the needle cover after the
sample loader has been connected and the needle positioning adjustment has
been completed. The sampling unit is fixed in place for transportation with a red
screw. (See Fig. 2-4 below.) Use a screwdriver (+) to remove the red screw.
Retain this red screw for later transportation purposes. Manually push the
sampling unit back a little and then push it in as far as it will go.

2.5 Connections

Waste Tube

Insert the waste tube firmly into the waste port located on the bottom of the main
unit. (Refer to Fig. 2-5) Securely tighten the waste tube with the bandage
provided in the accessory box.
Insert the other end of the tube to the waste bottle. (Refer to Fig. 2-6)
Note if the waste tube is bent, the waste eluent may not drain out smoothly.
Adjust (cut) the tube length to keep the tube end above the waste eluent level.

When changing the analyzer location, make sure that the tube is not loose,
broken or bent and that the waste eluent drains out smoothly.
The sample loader must also be temporarily disconnected when changing the
analyzer location.

1. If the waste tube is bent and waste eluent cannot smoothly drain, the
sample dilution may not be accurately executed during the assay.
Do not try to pick up or move the tube.
2. Keep the waste tube end above the top of the waste eluent.

Sample Loader Connection

1) For the 90SL

Take the sample loader out of the box and remove the cushioning and cover.
Take off the front SUS panel. Take care not to drop the screws into the unit
when doing so.
Temporarily place the main unit and the sample loader next to each other.
Remove the lower left panel of the main unit to facilitate cable connection
(two locations).
Connect the P1 (GND cable connector), which can be seen through the
panel hole in the panel on the lower left side; also, connect the R1, which is
a connector coming from the sample loader.
* The GND cable is the green and yellow striped cable.
Connect the 60P flat cable sticking out of the panel hole of the main unit to
the relay board (CB1) of the sample loader. (This cable is connected to two
terminals. One is to the left (the CN4 of the ASM-board) and the other is to
the right (the CN3 of the DRV2-board), both of which can be seen through
the panel hole.) Press the connectors solidly in as far as they will go.

Next, connect the sample loader to the main unit; do not catch the cable.
On the rear panel of the sample loader, there are a total of four screw holes
(two right and two left screws) for connecting the sample loader to the main
unit. At least two of the top screws must be screwed in (use the M5
accessory screws). If the system must be installed on a weak or flexible
bench, make sure to use all four screws.

Put the screws through the rear panel of the sample loader from the front
and tighten so that no looseness remains. At this time, if the screw holes are
out of alignment, use the adjustable feet on the main unit and/or sample
loader to adjust the height. Verify that the center of the screw holes align
between the main unit and sample loader and fix the tightening screws in
place.
Attach the lower left panel of the main unit, if removed.
The SUS front panel should be attached after adjustment of the rack transfer
or bar code reader.

2) For the 290SL

Use two people to remove the sample loader slowly from its box. Hold the
left and right sides and remove the cushioning and cover. Put the sample
loader on a bench and verify that the loader is level and stable. If necessary,
use the adjustable feet on the sample loader base to adjust.

Take off the front panel (SUS) of the sample loader. Take care not to drop
the screws into the unit when doing so.
Temporarily place the main unit and the sample loader next to each other.
If necessary, use the adjustable feet to adjust the height so that the screw
holes align.

Next, remove the lower left panel of the main unit.

Disconnect the 24 V power source cable fixed by the clamp folded in front of
the ASM-board and remove the connector housing from connector R4. Store
the removed connector in a toolbox or something similar to avoid losing it.
Fix the clamp back in its original position.
Put the connector R4 through the panel hole in the lower left panel of the
main unit and connect it to the P4 of the loader. Connect the GND cable
connector P1 from the main unit with the connector R1 from the sample
loader. Connect the 34P flat cable of the sample loader to the CN5 (DRV3)
of the ASM-board in the main unit (left side to the front). Make sure that the
lock is firmly connected.
(* Make sure to press this lock when removing these flat cables. Do not
attempt to remove the cables forcibly, as this could damage them.)
Connect the 60P flat cable sticking out of the panel hole of the main unit to
the relay board (CB1) of the sample loader. (This cable is connected to two
terminals. One is to the left (the CN4 of the ASM-board) and the other is to
the right (the CN3 of the DRV2-board), both of which can be seen through

the panel hole.)

Press the connectors solidly in as far as they will go.

Next, line up the main unit and sample loader; do not catch the cable.

In the same way as with the 90SL, connect the sample loader rear panel
and the main unit using the four screws (two each for the right and left sides).
Refer to the procedure for the 90SL.

Reconnect the lower left side panel of the main unit.


The SUS front panel should be attached after adjustment of the rack transfer
or bar code reader.

Elution Buffer Tube (Suction Tube)

Match the color marks of the Elution Buffer tube with the same color on Elution
Buffer pack No. 1, 2, and 3, and then insert the tube into the aluminum pack and
seal by closing the bottle cap.

The suction filter is attached to the top of each Elution Buffer tube. If the tube is
bent, straighten it out before connecting it and make sure it reaches the bottom
of the aluminum pack.

For aluminum pack reagents, squeeze the pack manually to remove as


much air space as possible, and then firmly close the bottle cap. If the
cap is loose, the elution buffer may deteriorate. It will also become
difficult to check the remaining volume visually.

Hemolysis & Wash Solution Tube

Open the Hemolysis & Wash (H/W) solution cap and insert into it the Hemolysis
& Wash solution tube (with anchor and bottle cap) sticking out of the H/W
Solution port on the left side of the main unit, then tighten the bottle cap. Make
sure that the anchor has reached the bottom of the bottle.
If you are using a 4-L bottle of Hemolysis & Wash solution (LL), use the
specialized bottle cap for LL provided as a standard accessory.

Power Source

Securely connect the power cord to the AC inlet of the main unit.
Make sure that the unit’s main power is off (O) before inserting the plug into the
socket.
The socket's power capacity must be 10 A and above. Be sure to connect the
unit to three-pin power socket.? ? ? 2P3 P ? ? ? ? ? ? ? ? ? ? ? ? ? ? ? ?

Caution 1. Do not use the same power source as that used for high
capacity equipment such as a refrigerator compressor.
2. Do not touch the power source with wet hands. This may
cause electrical shocks.
3. Be sure to ground the unit.
4. To allow the power to be easily switched off during an
emergency, do not place anything in front of the switch.
5. Leave enough space to allow the power cord connector to
be unplugged from the AC inlet.
6. Never insert too many power cords into the same socket.
Never use with an extension cord.

2.6 Power on

Turning Main Power On

The main power switch of the analyzer is located at the back of the left side, just
above the AC inlet.
The side marked “I” indicates power on, and the side marked “O” indicates power
off.

The main power switch also acts as a breaker. If the main power switch is turned
off immediately after the power is turned on, the analyzer may short-circuit. If this
should occur, be careful not to touch any metal parts of the analyzer. Immediately
turn the main power off, unplug the power cord from the power socket and
contact a service representative.

Caution Do not touch the power source, sheet key, or screen with wet
hands. You could receive an electric shock.

The memory is cleared when the analyzer is shipped. When you start the
analyzer for the first time, insert the system card in advance in order to
read the system program.
If the analyzer has been previously started, verify that there is no card in
the smart media socket or no other card than results stored card is
inserted in the socket before startup.

Procedure (New Device)

< For initial installation >

1) Insert the attached system card into the smart media socket on the
analyzer.
2) Turn on the main power switch. You will hear a startup beep.
After the display data is automatically checked (check screen) and the
G8 logo screen displays (Screen 2-1), the BOOT screen (System Loader
- Screen 2-2) displays and a check of the device’s internal circuits is
automatically run.

(Normal Operation)

3) If the internal circuit check terminates with no problems discovered, the


backlight will temporarily dim and the device will enter a stand-by mode,
waiting for the POWER key to be pressed.
If there are any problems (NG) on the BOOT screen, the ERROR LED
will illuminate and a “Check the hardware!” message will be displayed.
You will not be able to continue. The instrument requires inspection.
4) Press the POWER key located at the top of the sheet keys on the right of
the screen (Fig. 2-17).
The green POWER LED on the left side of the screen will light up when
this key is pressed.

5) The system and AS (auto sampler) programs are automatically read from
the system card. The operational status displays on the screen.
AS installation (Screen 2-3) takes about 5 minutes, and system
installation (Screen 2-4) takes about 30 seconds. A total of
approximately 6 minutes is required for all operations.

6) Next, the Language Display Selection screen (pop-up) will be displayed.

With the “>” marker on the default (English), press the INSTALL key.

7) Once the system parameters are set, “Checking Data ……Initialized” is


displayed on the last line.
8) In addition, the Assay Mode Selection screen (pop-up) that was initially
started up is displayed (Screen 2-6).

9) Leave the “>” marker on A1c STD MODE and press the SET key.
If there are no program problems, “Loading Data ……...Complete” is
displayed on the last line, the analyzer automatically starts up, and the
Main Screen (First Screen) as indicated in Screen 2-7 is displayed.

10) The startup sequence continues. The sampling mechanism is checked,


the pump is washed, the Elution Buffer is primed, the sampling and
dilution tubing are initially washed, and WARMING UP starts.
When installing the unit, cancel these operations by pressing the STOP
key three times in succession.

Procedure (Restarting)

< For devices that have already been started up (demo version, etc.) >

1) Confirm that there is no card or any other card than the result-storage
card in the smart media socket. If the system card or a
parameter-storage card is in the socket, this information will
automatically be read during startup and the device’s internal memory
will be overwritten.
(The parameters in the internal memory will be overwritten when the
parameters are saved to the card, only when the parameter “CLR” was
input. In other instances, the parameters are not overwritten.)
2) Turn on the main power switch. You will hear a startup beep.
After the display is automatically self-checked (check screen) and the
G8 logo screen (Screen 2-1) is displayed, the BOOT screen (System
Loader - Screen 2-2) is displayed, and then a check of the device’s
internal circuits is automatically run.
3) If the internal circuit check terminates with no problems discovered, the
backlight will temporarily dim and the system will enter a stand-by mode,
waiting for the POWER key to be pressed.
4) Press the POWER key located at the top of the sheet keys on the right
side of the screen. (Fig. 2-17)
The green POWER LED on the left side of the screen will illuminate
when the key is pressed.

5) The system program, AS (auto sampler) program, and backup


parameters will all be checked automatically. (Screen 2-8)

(Normal operation when the system card is not being read)

6) If there are no problems with the backup program or other parameters,


the analyzer automatically starts up, and the Main Screen (First Screen)
as indicated in Screen 2-7 is displayed.

7) The startup sequence continues. The sampling mechanism is checked,

dilution tubing are initially washed, and WARMING UP starts.
When installing the unit, cancel these operations by pressing the STOP

2.7 Adjustment of Sample Loader

Verifying the needle tip placement

Set the sample conatiner (primary tube or vial) on the rack and place it
on the loader.
Move the sample needle with MAINTE screen and verify whether the
needle move down to an appropriate height by eyes from the top of the
needle and side of the rack.
Refer to this manual (4.6 Parameter Settings for Engineers, 4.7
Maintenance Screens for Engineers) for the operation procedure in
MAINTE screen and details of parameters for service person.

If the needle placement is out of the center of the sample tube or its
height is not appropriate, it may cause the needle broken or trouble.

First of all, make sure of the installation for the main unit and the sample
loader and also confirm no cable catch between the main unit and the
loader, the height of the main unit which can be adjusted by adjustable
feet to be appropriate.

If the installation has no problem, as the fine adjustment of move length


(the number of pulse) is required, change it to an appropriate value
through the parameter screen for service person.

The tip of the needle is best placed above


the bottom of the container by 2 ~ 2.5 mm.

STAT port

The adapter for the φ13 tube is attached on shipment to STAT port. It
will be suitable to the 75 mm standard primary tube (φ12-13) or sample
vial. If try to use the φ14, change the adapter for the φ14. The adapter is
not needed if try to use the φ15.

An adapter can be replaced or removed easily by detaching two screws


mounting the plate of STAT port, which can be seen from the front. In
this case, do not drop off screw and parts for the adapter inside of the
sample loader.
When try to use sample vial, it requires an adapter regardless of size.
Without adapter, no appropriate placement is expected in the port.

Bar Code Confirmation

A bar code reader comes standard on all G8 sample loaders. If you are not
using bar codes, or do not want it to illuminate them, turn the #1 SW2 (8P
DIP-SW) on the ASM-board to OFF before turning on the main power.
If the SUS front panel is removed, it is easiest to adjust the bar code reader
position from the front, regardless of the sample loader.
In addition, the selection of the bar code reader specifications and the
detailed settings can be executed through BCR screen.
Refer to this manual “6.21.6 Bar code Reader Adjustment” for further
information about the installation and location adjustment.

< Preparation >


Prepare a bar code label that will actually be used, a sample container
(empty primary tube), or a vial adapter. Be careful of the label angle and
location during placement. Be sure the bar code label is oriented towards
the analyzer (that is, towards the slit of the sample rack - refer to the
operator’s manual about how to set the sample).
Place the rack on the sample loader and move the sample to the position
where the bar code can be read by moving the rack (adjust the position to
the right by two tubes from the sampling position). In this process, adjust the
rack guide (on the bottom of the rack) so that it is placed exactly at the
sampling position fixed by the latch holder. (You can move the rack by hand,

< Bar code reading check >

A bar code reading check is executed with the key on the MAIN –
MENU – UTILITY - BCR screen. (Screen 2-9) When this key is pressed,
the bar code reader executes a single flash (~300 msec) and executes a
reading operation. Once the information has been read, you will hear a beep
and the light will turn off. The read results are displayed on the last line of
the screen. Compare the results with the printed label to make sure the bar
code was correctly read. It is also recommended that you prepare multiple
labels and sample containers to check operations at multiple positions on
the rack.

- If the bar code reader continues to illuminate without a flash of light, the
mode of the bar code reader must be set.
Set MODE SET on the BCR screen to “1” and press the key. The
settings are applied to the bar code reader.

- Bar code reader flashes but does not read.


The bar code settings or specifications may not be correct.
Make sure that the bar code specifications agree with the setting on the
BCR screen and reset if necessary. Press the key.
The G8 uses the following four types of codes as bar code specifications:

CODE39, ITF, NW-7 (Codabar), CODE128

In addition, the following three codes can be set. G8 can set a maximum of
four codes at the same time of the seven codes total.

JAN (UPC/EAN), Industrial 2of5, COOP 2of5

- When you are only able to read from time to time (low reading ratio).
A low reading ratio could be caused by poor label printing quality, improper
bar code placement, or insufficient upper and lower margins (blank space).
See the operator’s manual and “Chapter 3.4 Samples" in this manual to
verify the reason.

2.8 Priming

Take the following procedure for priming reagents.


Display MAINTE followed by REAGENT CHANGE.
Highlight all of #1,#2 ,#3 and H/W key. By pressing CHANGE key, each
reagent priming and resetting of remaining volume of each reagent will
automatically start.

2.9 Drain Flush

Open the blue door of the main unit and execute the drain flush.
Press DRAIN FLUSH key on the above REAGENT CHANGE screen.
The comment to open / close the drain valve appears on the screen, follow
the direction to open / close the drain valve and OK key input.

2.10 Filter Insert

The filter attached to the analyzer is used for the QC inspection in the factory.
Therefore, replace with new filter at the beginning.
With the procedure, refer to the operator ’s manual.

2.11 Column Connection

Connect the column along with the operator ’s manual.


Run the pump with the second screen in Main screen and make sure of no
leakage around the filter, column connection and others during delivering
reagent.

2.12 Parameter and Date Setting

- Flow rate setting


Input the FLOW FACTOR to set the flow rate on the PARAMETER screen.
The FLOW FACTOR is a unique value for each analyzer. If you open the
right door on the main unit (CO, line filter section), a label with this
information is affixed on the lower right of the line filter holder (a label with
this information is also affixed on the inside right side of the pump housing).
To make fine adjustments, refer to “2.14 Flow Rate Fine Adjustment” in this
manual.
- Setting the primary tube push-down
The primary tube push-down length (100 mm TUBE) is set on the
PARAMETER screen. If the length of the primary tube that is actually being
used is 75 mm, set to “0”. If the length is 100 mm, set to “1”. If there are two
different lengths, set to “1”.
- Date setting
Verify the current year/month/date setting on the TIMER screen. If you find
any discrepancy, reset it. In addition, the automatic start-up time and date
can be set on this screen.
- Setting a comment
Comments can be input on the COMMENT screen and printed out on the
report. Such comments normally include the facility name and instrument
serial number.
- Automatic save setting
You can set automatic saves to the smart media card, and you can set
whether to save the list data on the Parameter (RAW AUTO SAVE) screen.
- Communication condition setting
When connecting with a host, set the communication settings on the
RS-232C screen. You can also set whether or not to query here. See “The

Host Connection Specifications” for further details.


- Setting the initial lamp intensity of the detector
Input the lamp intensity initial value of the detector into the parameter
#LAMP INIT REF for use by service personnel.
For further details, see “4.6 Parameter Settings for Engineers” and “5.3.2
Setting Initial Lamp Intensity of Detector” in this manual.
- Adjusting the touch panel
The factory default settings are normally used.
If you want to adjust the screen contrast or touch switch, refer to “7.8 LCD”
in this manual.

2.13 Assay Operations

Use a calibrator or controller to verify assay results and the chromatogram.


Refer to the operator ’s manual for further instructions.

2.14 Flow Rate Fine Adjustment

To finely adjust the flow rate, change the FLOW FACTOR to fulfill the
conditions indicated below. (in STD mode)
- Assay Sample : Control Level 1
- SA1C elution time : 0.59 min ~ 0.60 min

Once the flow rate has been finely adjusted, verify the conditions below.
- A1B elution time : 0.28 min ~ 0.32 min
- A0 elution time : 0.77 min ~ 0.81 min
If the conditions are not met, leaks or poor fluid pumping may result.
Verify the flow line carefully.

2.15 Calibration

Verify the chromatogram. If it is normal, calibrate the machine.
