Using Dilution Feature of the AutoPlus

Introduction
This technical bulletin describes the use of the dilution feature of the Hanson AutoPlus
Maximizer autosampler. This feature allows the user to combine a volume of sample
with a volume of diluent.
Dilution can be implemented on-line (at the same time that the sample is taken) or off-
line (at a time other than the sample time). A Multifill collector is required for both types
of dilution.

Dilution Feature Limitations


The main limitation with the dilution feature of the Hanson AutoPlus is the fact that the
sample material and diluent are not mixed by the instrument other than the action of
injecting one volume into the other. Therefore, this method should be used only where
a secondary mixing step is added by the user or when it can be demonstrated that
secondary mixing is not required.

On-Line Dilution
To use On-Line dilution, the user must have an understanding of scripting protocols with
the Hanson AutoPlus (refer to Technical Bulleting 1014). To implement the feature,
create a protocol and program the unit to perform either the Collect Only or Collect &
Detect function. Enter the script edit mode and insert a dilution function immediately
after the Collect Only or Collect & Detect function above. When the script is
implemented at sample time, after the sample is collected, it is then diluted using the
parameters programmed below.
The parameters that can be programmed for on-line dilution are:
 Rinse Volume (0-99 ml): the instrument will rinse the fluid lines to waste by this
amount using diluent from the source selected below before starting the dilution
process.
 Dilute Volume (0-99 ml): the instrument will dilute the sample just collected by the
amount programmed in this parameter.
 Source (A|B|C): is the port connected to the source diluent solution.
 Occurrence: determines when the dilution occurs as per the scripting rules.

Off-Line Dilution
Off-line dilution can be performed whether the AutoPlus is in the basic or the advanced
mode and does not require knowledge of scripting. To implement the feature, press the
syringe control key. Press key “4” to select the off-line dilution. If this option is not
available, press the “9” key to view the automated functions. Program the parameters
listed below and then press the “run” key to dilute the samples on the rack.
The parameters that can be programmed for off-line dilution are:
 Rinse Volume (0-99 ml): the instrument will rinse the fluid lines to waste by this
amount using diluent from the source selected below before starting the dilution

 Dilute Volume (0-99 ml): ): the instrument will dilute the sample already in the rack by
the amount programmed in this parameter.
 Source (A|B|C): is the port connected to the diluent solution.
 Total Rows (1-x): the total number of rows that will be diluted, starting with the
current position of the needle block of the Multifill. The user can dilute rows 5-10 by
moving the Multifill needles to row 5 and selecting 5 total rows before starting the
dilution process. The total number varies with the capacity of the rack on the Multifill.
