OPENING

Good afternoon everybody! Im really glad to meet all of you again and I hope we’re still in a
good health my name is I Gusti Ngurah Trisna Meyana Putra with student number seventy
nine from farmasi students math and science faculty, Udayana University. In this good
oppurtunity I would like to present my projects about “Lansoprazole”.

CLASS OF DRUGS

Lansoprazole is the second member of the substituted benzimidazole class of antisecretory

agents approved for use in the United States. These drugs decrease parietal cell acid secretion
by inhibiting H+, K(+)-adenosine triphosphatase, the final step in the secretion of acid.
Lansoprazole is in a class of medications called proton pump inhibitors. It works by
decreasing the amount of acid made in the stomach.

DRUG INDICATIONS

Lansoprazole is used to reduce gastric acid secretion and is approved for short term treatment
of active gastric ulcers, active duodenal ulcers, erosive reflux oesophagitis, symptomatic
gastroesophageal reflux disease, and non-steroidal anti-inflammatory drug (NSAID) (EN-ES-
EI-AI-DI) induced gastric and duodenal ulcers. It may be used in the maintenance and
healing of several gastric conditions including duodenal ulcers, NSAID(EN-ES-EI-AI-DI)
related gastric ulcers, and erosive esophagitis. Lansoprazole prevents recurrence of gastric
ulcers in patients who have a documented history of gastric ulcers who also use NSAIDs
(EN-ES-EI-AI-DI) chronically. Predictably, it is also useful in the management of
hypersecretory conditions including Zollinger-Ellison syndrome. Lansoprazole is effective at
eradicating H. pylori when used in conjunction with amoxicillin and clarithromycin (triple
therapy) or with amoxicillin alone (dual therapy).

THERAPEUTIC DOSES

Duodenal Ulcer

Active: 15 mg PO qDay for 4 weeks (Fiveteen miligrem PI-O kyuDay for Four weeks)

Maintenance: 15 mg PO qDay (Fiveteen miligrem PI-O kyuDay)

Gastric Ulcer

30 mg PO qDay for 8 weeks (Thirty miligrem PI-O kyuDay for Eight weeks)
NSAID-associated GU (EN-ES-EI-AI-DI-associated JI-YU)

Treatment: 30 mg PO qDay for 8 weeks (Thirty miligrem PI-O kyuDay for Eight weeks)

Prevention: 15 mg PO qDay for 12 weeks (Fiveteen miligrem PI-O kyuDay for twelve
weeks)

Gastroesophageal Reflux Disease

15 mg PO qDay for 8 weeks (Fiveteen miligrem PI-O kyuDay for eight weeks)

Erosive Esophagitis

30 mg PO qDay for 8-16 weeks (Thirty miligrem PI-O kyuDay for Eight till sixteen weeks)

Maintenance: 15 mg PO qDay (Fiveteen miligrem PI-O kyuDay)

Hypersecretory Condition (eg, Zollinger-Ellison Syndrome)

60 mg PO qDay initially; up to 180 mg q12hr used (Sixty miligrem PI-O kyuDay initially, up
to one hundred eighty miligrem kyu twelve hour used)

If dose >120 mg/day PO, administer in divided doses q12hr (if dose less than one hundred
twenty miligrem per day)

Helicobacter Pylori Infection

Triple therapy: Lansoprazole 30 mg + amoxicillin 1 g + clarithromycin 500 mg PO q12hr for

10-14 days (thirty miligrem plus amoxicillin one gram plus clarithromycin five hundred PI-O
kyu twelve hour for ten till fourteen days)

Dual therapy (clarithromycin resistant): Lansoprazole 30 mg + amoxicillin 1 g PO q8hr for

14 days (thirty miligrem plus amoxicillin one gram PI-O kyu eight hour for fourteen days)

Penicillin allergy: Lansoprazole 30 mg + clarithromycin 500 mg + metronidazole 500 mg

q12hr for 10-14 days (thirty miligrem plus clarithromycin five hundred plus metronidazole
five hundred miligrem kyu twelve hour for ten till fourteen days)

POTENTIAL SIDE EFFECTS

Lansoprazole may cause side effects. Tell your doctor if any of these symptoms are
severe or do not go away:

 constipation
 nausea

 headache

 dizziness

 diarrhea

Some side effects can be serious. If you experience any of the following symptoms, call
your doctor immediately:
 rash

 blistering or peeling skin

 hives

 swelling of the eyes, face, lips, mouth, tongue, or throat

 difficulty breathing or swallowing

 hoarseness

 irregular, fast, or pounding heartbeat

 excessive tiredness

 lightheadedness

 muscle spasms, cramps, or weakness

 jitteriness

 uncontrollable shaking of a part of the body

 seizures

 severe diarrhea with watery stools, stomach pain, or fever that does not go away

 new worsening joint pain

 rash on cheeks or arms that is sensitive to sunlight

 decreased urination

 blood in the urine

People who take proton pump inhibitors such as lansoprazole may be more likely to fracture
their wrists, hips, or spine than people who do not take one of these medications. People who
take proton pump inhibitors may also develop fundic gland polys (a type of growth on the
stomach lining). These risks are highest in people who take high doses of one of these
medications or take them for 1 year or longer. Talk to your doctor about the risk of taking
lansoprazole.
