1. Peptone- water sugar reactions


- A series of peptone- water sugars
Glucose—mannitol—lactose—sucrose—dulcite—urea
- Can be used for the biochemical differentiation of E. coli
- Acid production red
- Gas positive bubbles

2. Voges-prokauer (VP) test


Some Enterobacteriaceae ferment glucose with the production of acethyl methylcarbunol
oxidized and reacts with α-naphthol red color

3. Indole tube test


Indole positive bacteria break down amino acid trypthopan to indole reacts to Kova’s
reagent red color

4. Nitrate reduction test


Organism is incubated in a broth containing nitrate and being observed after 4h red color (+)
E. coli positive nitrate production

5. API
These kits use dried reagent in cupules to which a suspension of the test organisms is added.
The strips are incubated overnight and the reactions determined. This allows the rapid
processing of a large number of isolates.

6. Methyl Red Test

E. coli is one of the bacteria that


produce acids, causing the pH to drop
below 4.4. when the pH indicator
methyl red is added cherry red
(positive MR test)

7. Citrate test

Simmon’s media contains bromthymol blue, a


pH indicator yellow at acidic (around 6) and
gradually changes to blue at alkaline pH
(around 7,6)

Culture on MSA (mannitol salt agar), showing yellow colonies. This is a selective medium for
recovering S. aureus when screening for carriage in infection control investigations.

Culture on nutrient agar, produce a greenish


pyocyanin pigment, non-lactose fermenter.
It is oxidative positive.

Microscopic: gram – bacilli, typical enterobacteria most have a similar morphology and can’t be
distinguish by gram staining
