Biochemistry is a platform of creation and significant contributions to the
fields of biology, physiology, immunology, microbiology, pharmacology, and
toxicology, as well as the fields of inflammation, cell injury, and cancer. These
close relationships emphasize that life, as we know it, depends on biochemical
reactions and processes. In the first experiment, we study what is protein
quantification and why we need this method in our life.
Protein quantification is necessary to understand the total protein content in
a sample or in a formulated product. Accurate protein quantification is important as
a range of other critical assays require precise total protein content results in order
to generate data. Careful selection of the protein quantification method is required
to ensure accurate data.
In this practice, the Hartree-Lowry assay is used for the quantification of
proteins. A standard protein of known concentration is used for the construction of
a calibration curve. Folin-Ciocalteu reagent is added to protein solutions to develop
a color whose intensity is colorimetrically measured. The Albumin solution is
chosen as the appropriate standard. Various known concentration of albumin
solution is mixed well with reagent to enhance color development. Nowadays,
people apply protein quantification in heatlth test. Protein in the blood plays a very
important role, constitutes cells and participates directly in the physiological
processes of the body. The test of total protein in the blood is significant in
measuring serum albumin and globulin content. The concentration of protein in the
blood helps to reflect abnormalities of the liver, kidneys, gastrointestinal diseases,
nutritional status, ... of the body.
Apart from Hartree-Lowry assay, there are some methods to estimate the
protein concentation like BCA, absorbance at 280, Bradford or Biuret assay,.. To
me, total protein quantification applying Biuret assay is the most interesting
application and I really want to detect it in the future. Total protein quantification
is to check the nutritional status of the patient, detect some diseases such as
multiple myeloma, protein disorders, infections, autoimmune diseases, and protein
loss diseases. The total protein in the blood of the patient is determined by the
colorimetric method based on the Biuret reaction principle:
+In an alkaline environment, molecules with 2 or more peptide bonds will complex
with Cu2+ions
+In Serum proteins react with Cu2+ions
In the alkaline environment, the complexes are blue purple. The color
density is proportional to the protein concentration of the patient sample. If patient
have high protein concentration, they may have these health condition:Cancer,
multiple myeloma, dehydration, Chronic kidney disease, liver failure. And if
patient with low concentration they may get malnutrition, congestive heart failure,
kidneys disease,.. Protein quantification also use in other field, for example in
serum products, animals breeding or food idustry,..
Learning Biochemistry helps us understand more about the diversity of
diseases, genetic, evolution and DNA and apply to our life. From biochemistry
experiment, people enable to create safe drugs, medicines, and vaccines to avoid
the virus, repel the pathogen and make agriculture and food development or use
testing method to check for the protein in our body, our food to have a healthy life
and improve the biochemistry knowledge.
