Specifications for Surface Plasmon Resonance based

Protein Interaction System

 The system must be 2 channel or better automated, surface plasmon resonance based
protein interaction analysis system in real-time.
 Should provide comprehensive characterisation of bio-molecular interactions in solution.
Must offer real insights into protein functionality and biological mechanisms. Must give
out quality data to reach the right conclusions. This must be shown by a proven legacy
of publications and installations both internationally and in India.
 The system must provide unique and comprehensive data on the interactions between
biomolecules: Yes/No binding, binding selectivity, binding affinity, binding kinetics,
active concentration.
 System should be able to perform Calibration free concentration assays to determine
active concentration of biomolecules without the need of calibrant molecule.
 System should be able to determining kinetic parameters for a reversible molecular
interaction between a ligand immobilized to a solid support surface and a binding
partner to the ligand in solution in a single cycle kinetics assay where there is no
requirement of intermediate regeneration injections.
 Flow-based system preferably with micro fluidics: Less than 100µl per sample
 Sample Process: At least 15 samples automatically and sequentially in a single run.
 Sample positions: At least 16 positions should be there.
 System should have at least two flow channels of which at least one should be used for
on-line reference subtraction in real time.
 The system must include an Inline degasser, Variable assay temperature control,
 Software should have custom assay Wizards/ defined methods for Single cycle kinetics
and classical multi-cycle kinetics assays and analysis; Concentration analysis.
 Software should have methods/wizards for Ligand immobilisation on the sensor surface
using covalent coupling (Amine-, thiol-, aldehyde-, maleimide- coupling) and affinity
based capture of ligand (Streptavidin-biotin, Antibody-based, HIS tag-Ni-NTA,
hydrophobicity based) on surface.
 User defined fitting models for kinetics and affinity analysis for 1:1 binding; 2 state
reactions; bivalent interactions, etc.
 Protocols, chemistry and suitable chips must be available for study of proteins, tagged
proteins, Low Molecular Weight molecules (as low as 200 Daltons), membrane
associated molecules, nucleic acids, carbohydrates, viruses and intact cells.
 Injection volume on the system should be selectable between 5 to 90 µl, with a flow rate
selectable in the range of 1-100 µl/min.
 System should be able to perform assays at various temperatures 5 to 40 oC, as per user
requirement
 Should have a baseline noise level of < 0.1 RU (RMS) and a baseline drift of < 0.3
RU/min.
 System should be able to analyse samples in refractive index range of 1.25 to 1.40.
 The following detection parameters must be met by the system: Association rate
constant (Ka) 103 –107 M–1 s–1 , Dissociation rate constant (Kd) 10-5 – 0.1 s-1, affinity at
equilibrium 104 – 2 x 1010 M–1, Concentration >1x10-10 M for >10kDa analytes and >1x10-9
M for <10kDa analytes.
 A Human Antibody Capture Kit that provides the reagents and protocols required for
convenient capture of human or humanized IgG antibodies onto a sensor chip surface
must be available.
 A Mouse Antibody Capture Kit that provides the reagents and protocols required for
convenient capture of mouse IgG antibodies onto a sensor chip surface must be
available.
 Chemistry must be available to address components in complex sample matrices such
as plasma, serum or cell lysates may bind non-specifically to the sensor chips,
complicating the analysis of specific binding interactions.
 Flexibility to study a wide range of molecular interactions, involving: Proteins, Nucleic
Acids, Carbohydrates, Lipids, Whole cells/Viruses/bacteria.
 The system must rapidly generate usable data that is simple to operate offering versatile
assay formats
 The system must offer built-in guidance affording short learning curve for new users.
 The system must have intuitive, wizard-based software for fast and easy set up, assay
run and evaluation. Must have integrated methodology support – including
recommendations, tips and troubleshooting. The system must offer additional
knowledge directly accessible on the web from the system software.
 System should be quoted with sensor chips pre-immobilised with protein A , Protein G
and Protein L”.
