Safety
Safety Practices and Hazards

PICO series pH, Conductivity meter has been carefully designed in accordance with the safety

standards. It is in the interest of the purchaser that he is aware of the resultant dangers of misuse

and or incorrect handling of these products.

Information on safety practices is provided with your instrument and operation manuals. Before

using your instrument & accessories, you must thoroughly read these safety practices.

The following general safety precaution must be observed during all phases of operation, service

and repair of this instrument. Failure to comply with these precautions or with specific WARNIGS

elsewhere in this manual may impair the protection provided by the equipment. In addition it

violates safety standards of design, manufacture, and intended use of the instrument.

Electrical Hazards:-

PICO pH, Conductivity meter contains electrical circuit, devices and components operate at very

low. Hence it is very safe to handle the instrument.

Enclosure covers that are retain by fasteners, which require the use of the tool for removal may be

opened by Labindia trained, Labindia qualified or Labindia authorized service engineers. Personnel

without such training or persons currently undergoing a course of training require through

instructions. This operator’s manual should form the basis of these instructions.

Application of the wrong supply voltage connection of the instrument to an incorrectly wired supply

outlet or improper electrical grounding can create a fired hazards or a potentially serious shock

hazard and could seriously damage the instrument and any attached accessory.

 DC Adapter – The meter uses detachable DC Adapter with earth connection. Failure to

ensure adequate earth grounding by not using this adapter may cause reading instability. If

the adapter becomes damaged, replace immediately with same rating and same

specifications.

Use Of Instrument:-

The unit is design and built for use in laboratories. It is intended to be use to
determine the pH or Conductivity or Ionic Concentration of the liquids under
measurement and analysis. Use appropriate safety as per standards for handling of
chemicals during measurement.

Any use that does not confirm with technical data of the instrument is being to be a case of
misuse. The customer shall bear sole responsibility of any damage due to such misuse.

Following Applications / Uses in particular are inadmissible:-


 Use of Epoxy body Sensor with organic solvents.

General Hazards:-

General Hazards arise from:-


 Samples mixtures, Unknown chemical composition.
 Combustible Gases or solvent vapours close to instrument.

 Only Labindia Authorised, Labindia Trained personnel is allowed to service


the instrument.
 Do not use this instrument if it has unauthorised modifications. Unauthorised
modification may result in fire, electric shock or other hazards.

Chapter 2

Introduction
LABINDIA herewith offers you the state-of-the-art micro-controller based pH / Conductivity / Ion
measurement system with user friendly dialog based easy to use operation, making error free and
precise readings. This bench-top pH/Conductivity/Ion meter is an ideal help in every laboratory for
routine measurement as well as R&D work.

By selecting one of the pH/Conductivity/Ion Measurement Mode, it simultaneously measures pH,


mV and temperature in pH mode or Conductivity and temperature in Conductivity mode or Ion
concentration in Ion measurement mode. Optional modes available are pH data logging, TDS &
Salinity measurement. A single MODE Key for Measurement / Function Selection within the
selected measurement makes system more versatile with Password protection to stored data. 5-
point pH calibration is available in Linear Regression or Segmentation (point to point lines).

The meter has Individual non volatile data memory storage block with memory scanning facility on
display, such as 40 pH Values, 40 mV Values, pH Data logging value and 40 Conductivity, TDS &
Salinity values and 40 Ion concentration values. With data logging function in pH, user can study
Kinetics of chemical reaction. The pH values recorded during Data Logging can be printed in the
graphical form with the limit marks.

The GLP Approach for Calibration and data presentation gives more authentic Reporting with its
print out on plain inexpensive paper. It has two Data report presentations –
(a) Standard : 40 data readings or (b) GLP : 40 data readings with alphanumeric Sample names
and ID numbers.

pH Measurement Theory:

pH measurement is one of the most common requirement of today’s laboratories. Laboratories


involved with Environmental measurements, Water and waste water, soil analysis, Industrial
boiler/cooler feed water, Pharmaceutical & Bulk drug manufacturing, Specialty and Agro Chemical
manufacturing, Electroplating, Food processing requires to do routine pH measurements for
various aspects such as Pollution control, Quality control, Process monitoring & control etc.
Generally pH is measured in the scale of 0 to 14 pH units. But with the development in technology
and growing needs of the various Industries, pH measurement range is extended from – 2.00 pH
to 20.00 pH.

Measurement of pH of any Aqueous Sample solution gives the degree of acidity or alkalinity of the
solution. Mathematically pH is defined as the negative logarithm of the Hydrogen Ion
concentration. pH Electrode measures the pH of a Sample solution potentiometrically. When the
pH electrode is immersed in the sample solution the electrical signal gets developed across the
sensing membrane of indicator electrode and reference electrode. Output of the electrode varies
with change in pH value with a linear relation of 59.16mV / pH unit value. The electrode slope
changes with change in temperature, hence temperature compensation is required. With the PICO
pH meters this slope change can be compensated manually or automatically. For precise pH
measurement automatic temperature compensation (ATC) with separate temperature sensor is
needed.

value @ 25°C. It also Stores, Protects, Scans & Prints the data with GLP concept.

Conductivity Measurement Theory

Conductivity is the measurement of a materials ability to conduct electric current. The basic unit of
conductance "G" is the "siemens" (formerly called the Mho). Conductance is the reciprocal of
resistance (measured in ohms).

Measurement of Conductivity of Sample solution gives the degree of electrical conductance in a


defined volume of the solution. Electrical Conductivity of the solution is proportional to the number
of ions available in it. And hence conductivity measurement will give a direct reading of the
solutions concentration.
The principle for measurement of conductivity is simply by applying potential across two platinum
plates which are placed in a sample (normally AC voltage) and then the current is measured.

Conductivity can be determined from the voltage and current values. Because conductivity is
affected by cell geometry, specific conductivity should be used. The specific conductivity can be
calculated by multiplying the conductivity by the electrode’s cell constant. This constant is
determined by the formula, length / area. The unit S/cm is too large and usually the measurements
are done in the units mS/cm or uS/cm. The conductivity of the sample solution changes with
change in temperature, the relation is given as below,
Gt = Gtcal {1 + a(t - tcal)}
Where: Gt = Conductivity at any temperature in °C
Gtcal = Conductivity at calibration temperature in °C
a = Temperature coefficient of solution at tcal
tcal = Calibration temperature.
With the PICO pH/Conductivity meter’s this change can be compensated manually or
automatically by considering the Temperature Coefficients of the Sample. For precise Conductivity
measurement automatic temperature compensation (ATC) with separate temperature sensor is
needed and Sample and Standard should be at same temperature, preferably at the reference
calibration temperature 25°C

The Temperature coefficient mostly used is 2% per °C, since this approximates water and NaCl.
Conductivity is useful for measuring total dissolved solids (TDS) and salinity.

INSTALLATION
1. REQUISITES

 Leveled Platform/Table with nearby 230Vac, 50Hz/ 5 Amp. Power socket. (Ensure the Input
Power supply is within +/- 10% tolerance).

 Any interruption to earth wire is prohibited. Risks caused by possible internal


defects are thus prevented.

 Make arrangements for liberal amount of fresh distilled water and any five standard traceable
buffers (difference between consecutive buffer should not be greater than 4 pH unit) and
1~2gms. KCL or 0.1N HCL for pH Electrode activation.
Refer pH electrode maintenance section, Page no. 62.
 Conductivity calibration solution 1.413 mS (0.01M KCL).
 Clean & dry glass beakers for all sample measurements.
 Tissue paper roll and Suitable container for Waste disposal.

2. SETTING UP THE METER

 Carefully remove the meter wrapping and the accessories from the packaging.
 Place the meter on the table where it will be installed for measurement purpose.
 Align the unit for complete view of LCD Display by opening the Tilt Legs.
 For safety reasons and to ensure sufficient cooling in the electronic
compartment, the side walls of the instrument must be kept at least 20 cm
away from the walls or other objects. No containers, chemicals or other
appliances should be placed behind a instrument.

3. ASSEMBLING THE ELECTRODE STAND

 Get the Flat Base round plate & holder arm supplied with the unit.
 The Electrode Stand is made by inserting the Holder arm bottom adapter onto the round base
plate pin firmly and tightens the wing nut on it for sturdy assembly.
 Adjust the Electrode holder and stand angle as required and tighten the wing nut on the center
joint to lock the setting.
 Carefully insert the Electrode from top and temperature probes in the holder.
 Place the assembly on flat horizontal plane besides the meter.

4. PRINTER INTERFACE CONNECTIONS


A) Parallel Printer
 Place the printer on the suitable side of the meter.
 Connect the printer data cable to the 25 pin D-shell connector provided on the back panel.
 Adjust the printer dip switch settings as shown in. Appendix - A.
 Ensure that printer is loaded with paper.

B) Serial Printer
 Place the printer on the suitable side of the meter.
 Connect the printer serial data cable to the RS232-2 port in 9 pin D-shell connector
provided on the back panel.
 Ensure that printer is loaded with paper.

5. PC CONNECTIVITY
 Connect the supplied serial cable to the 9 pin D Male connector at the rear side of the
instrument and other terminal to com port of the PC. A Hyper Terminal Program is used to
receive the data from the instrument.
 Refer PC connectivity for more details.

6. SWITCHING ‘ON’ THE pH/CONDUCTIVITY METER

 Ensure proper Mains fuse has been installed in the instrument as per the
input voltage mentioned on the instrument label.

 Connect the pH, Conductivity Electrode and Temperature Sensor to appropriate socket /
Connector to back panel of the meter.
 Carefully remove the cap from end of the Electrode (This cap is used in shipping to protect the
electrode bulb and also to keep the electrode in moist condition until put into use)
 Check the electrodes internal reference filling solution level, if necessary level it with
recommended solution (3M KCl).
 Place the pH electrode into electrode storage solution, pH 4 buffer or saturated KCL solution.
 Place the conductivity electrode into D. M. Water/ Distilled Water to remove any impurities
adhering to the probe body and the platinum pins.
 Connect the DC adapter to a suitable 230 VAC/50 Hz stabilized power supply with proper earth
connection.
 Switch ‘ON’ the meter. The 20 character, 2 line display will read as follows.

Splash waterproof polyester keyboard ( 20 keys ).

Key functions:
L.H.S. Keys: Alphanumeric Dual Function ( A~Z , 1~9 ), Zero (0) with Dash (-), Dot (.),Slash(/),
decimal point ( . ) and Clear Entry( CE ) Key.

R.H.S. Keys: Functional Keys and RESET key.

Description of Functional Keys:

MEM : To Clear the Stored Readings / Calibration data of the selected


CLEAR Mode from meter Memory.

RESET : To Reset the meter.

CAL : To perform CALIBRATION within the selected Operating Mode.

FWD : This key has dual function, (a) To check the reading in scan mode. (b) To
TEMPCO select Auto / Manual temperature compensation.

SCAN : To SCAN / read the stored readings with the help of FWD/TEMCO key.

PRINT : To Print the Stored Readings / Calibration data of the selected


Mode from meter Memory with GLP format.

MODE : To Select Meter Operating Mode.

ENTER : To acknowledge the Entered value.

CE : To clear the wrong entry & to go backward for previous display screen.
BACK

 : To Select the GLP or Standard Mode between sample measurement & storage.
GLP GLP mode :- In this selection, the Sample related detail such as
Sample Name and ID. NO. can be entered after each Measurement.
Standard mode:- In this selection, Direct Measurement can be done
with-out entering the Sample related details.

Operation

USING THE INSTRUMENT :

After the Meter is powered ON only two keys are active, i.e. RESET and MODE.

As described in the Section 2 – Panel details page 17,

 RESET key is used to initialize the Meter. By pressing the RESET key Meter gives Beep and

displayed and later acknowledged by pressing ENTER key.

Please note the Second Line of the display shows the Active Keys. The very first Mode Displayed

II. Calibration of Instrument and pH Electrode: CAL

The PICO+ pH meter is available with two interpolation method for calibration.
1. Segmentation (default) or 2. Linear Regression (Optional) or Both.
In this manual both the calibration methods are explained for user information purpose.

Data Logging of pH v/s Time is an important facility for the study of Chemical Kinetics Reaction or
Enzymatic Kinetic Reaction. This can be also used for process monitoring in laboratory pilot plant
runs.

 The instrument records the pH reading w.r.t the User entered Data Log. Intv. (1 ~ 60 min.) and
No. of Intervals ( no. of readings – 24 max). The pH Data can be logged upto 24hrs. duration
for Slow Kinetic reaction with the Log. Intv of 60 minutes each.
 With pH Limit 1 & Limit 2 values for marks, the Graphical representation of the reaction trend
gives more meaning to the recorded values.

III. Taking Conductivity reading of Sample and Storing :- ENTER

 Remove the Conductivity electrode and Temperature sensor from the solution and rinse them
with distilled water 2 ~ 3 times, soak the excess water with tissue paper.

 Again rinse the Cell with the Sample solution, then insert the Cell and Temperature sensor
(RTD) in the another beaker containing same Sample.

 Stir the Sample Solution keeping the Conductivity Cell and RTD immersed.

 Incase of Standard Mode selected, pressing the ENTER key the readings are Stored in the
memory without Sample Name & ID.NO., else if GLP operating mode is selected at start
readings gets Stored in the memory with the Sample Name & ID. NO.

Calibration Reminder is another helpful feature in PICO Conductivity meter to assist the user for
complying with GLP / ISO regulatory needs.

The Calibration Reminder can be enabled by setting the next Probable DATE for performing
Calibration. PICO conductivity Meter’s internal Real Time Clock keeps the watch on it when
powered ON and the measurement function is selected on that date, the Buzzer will generate
BEEP sound and the Display prompts for re-calibration.

Ensure the DATE and TIME of internal Real Time Clock is SET correctly. Refer page no. 58.

Chapter 5

Operation Guideline
Useful Hints for pH & mV measurement:-
 New electrodes (or electrodes that have been in storage) should be conditioned in one inch
(25mm) of pH 7.0 or 4.0 buffer for several hours prior to their use. Make sure that the wetting
cap has been removed from the tip of the electrode.

 Always remove the wetting cap and fill hole plug during calibration and measurements.
Replace the fill hole plug when done.

 Calibrate the electrode daily. Though a 1-point calibration may be suitable for some
applications, we recommended that a 2-point calibration (that brackets the expected pH range)
be performed.

 Be aware that if you are measuring hot or cold samples without the use of an ATC probe, the
values displayed are not accurate. You should be calibrating and measuring at room
temperature.

 When transferring from one sample to another, always rinse the electrode with distilled water
and bolt dry. Handle the electrode carefully and do not use it as a stirring rod.

 Do not use calibration buffers after the expiration date printed on the package. Keep the
bottles tightly capped and stored according to the manufacturer’s instructions. Never put used
buffer back into the bottle of new buffer.

 For small sample volumes, make sure that both the pH bulb and the reference junction are in

 Response time is a function of both the electrode and the solution. Some solutions have very
fast response while others, particularly those with low ionic strength, may take several minutes.

 Samples must be in solution (water). You cannot measure the pH of a dry sample.

Useful Hints for Conductivity & TDS

 When measuring, make sure the solution is above the cell chamber slot.

 Make sure the cell chamber is free of bubbles when measuring. To reduce air bubbles, stir
centrally and then leave to stabilize.

 Allow sufficient time for the sensor to stabilize when measuring samples of different
temperatures.

 Rinse sensor with next measurement sample between measurements, and ensure sensor is
dried when re-zeroing the instrument.

 For greater accuracy, calibrate using a standard close in value to the sample.

Instrument Maintenance:-
The Labindia pH & Conductivity Meter needs no maintenance except for an occasional wipe with a
damp cloth. The front panel is made of ABS Resin and is not affected by the solvents. ABS Resin
is known to be affected by some organic solvents, do not use solvents like Chloroform, toluene,
xylene Acetone and methyl-ethyl-ketone. It is good laboratory practice to wipe away any spills as
soon as they occur.

Regular maintenance schedule and proper storage of pH electrode will maximize performances
and help to extend the life of electrode.

Weekly :-

1) Inspect electrode for scratches, cracks salt crystals built up on membrane / liquid junction
deposits.
2) The reference chamber of refillable electrode should be drained, flush with fresh filling solution
and refilled.
3) Use a dedicated pH electrode storage solution to keep electrode in proper working order.
4) Constant use of this solution will keep the electrode ready to use and provide fast stable
response.

Short term storage :- Between measurement / up to one week.
Soak the electrode in pH storage solution.

Long term storage :- More than one week.


The reference chamber should be filled and the fill hole securely covered. Cover sensing element
and reference junction with its protective cap containing few drops storage solution.

pH ELECTRODE MAINTENANCE

Solutions used to clean electrodes must be handled with the care according to toxic
or corrosive substances. Please refer to relevant MSDS.

 Do not allow the reference chamber of the electrode to dry out. Always keep it filled with the
proper fill solution.
 Formation of KCl salt at the tip and side of the electrode is normal and should be rinsed off
with warm water.
 Leave the tip of the electrode in an inch (25mm) of pH 7.0 or 4.0 buffer when it is not being
used. Do not leave it in distilled water. For long term storage follow the electrode
manufacturer’s instructions.
 If the electrode slope is off or the electrode response has become sluggish or inaccurate,
change the fill solution. If that does not improve the response then the pH sending glass and
the junction have probably become coated with some of the samples being tested.
 Test the junction for flow – rinse the tip of the electrode, blot dry and let the electrode hang in
the air for 15 minutes. A proper flowing junction should have KCl salt crystals forming on it. If
none appear, review the following suggestions for cleaning.
 Protein contamination – Soak the tip of the electrode for 1 – 2 hours in a solution of
10% Pepsin and water with enough Hydrochloric acid added to bring the pH of the
solution to 1.0. Rinse the electrode and soak in pH 7.0 buffer until stable.

 Oil contamination – Carefully clean the tip of the electrode using a cotton swab soaked
with alcohol or acetone. The tip of glass body electrodes can be put directly into organic
cleaners but do not put plastic or epoxy body electrodes into organic solvents. Rinse
the electrode and soak in pH 7.0 buffer until stable.

 All other – Soak tip of the electrode in 0.1 M

 Hydrochloric acid for one hour. Rinse the electrode and soak in pH 7.0 buffer until
stable.

pH Electrode cleaning

 General
 Soak the electrode in O.I.M HCL for 30 minutes
 Removal of deposits
 Protein :- Soak the electrode in 1% pepsin in 0.1 M HCL for 15 minutes.
Inorganic :- Soak the electrode in 0.1 M tetrasodium EDTA solution for 15
minutes
Grease & Oil :- Rinse the electrode with mild detergent of methanolic Solution.

Conductivity electrode :- Maintenance, Cleaning and Storage

Maintenance of conductivity equipment includes periodic checks

 Protect the conductivity system from dust and excessive heat and cold.

 Keep all cable connector dry and free of dirt and extraneous matter.

 Protect connector ends in a clean plastic bag when not in use.

Sensor cleaning

 Conductivity sensors must be clean to produce accurate results; residue from previous
samples can coat surfaces of sensor and cause erroneous reading.

 Clean sensors thoroughly with deionized water before and after making a measurement.

 Remove oil residue or other chemical resistors (salts) with a detergent solution. Sensors
can soak in detergent solution for many hours without damage.

 Never rub or use a brush on platinum coated sensors.

APPENDIX – C

Interpolation Methods used for pH meter calibration

 Linear Regression:

A single Best fit line is drawn for all the data points. Because a single line is
drawn to best fit all pH data points, the line may not go through all (any) of the
calibration points if there are more than two points.

The Slope and offset for this type of calibration are referred as best fit Slope
and best fit Offset. This type of calibration may be performed with
temperature compensation.

To perform Calibration using this mode, it is recommended to use the


Accurate/Traceable buffer solutions. Several time rinsing of pH electrode with
distilled water and then buffer prior to calibration.

 Segmentation (Point to Point) :

The calibration is divided in Sets or “Segments” of two calibration points and


lines are drawn between each segment. A slope and Offset are calculated for
each pair of calibration point.

The Average of the Slopes and Offsets of the segments are reported in the
calibration report. This type of calibration may be performed with temperature
compensation. There must be at least three calibration points to apply this
method.

PICO+ series – Data Downloading Capability to Personal Computer

Installation/setup Instructions:
The instrument software has been incorporated with RS232C protocol interface for transferring the
printable data to PC having standard data terminal software Windows - Hyper terminal.

Features of PC data transfer,

1. Data transfer to : Printer or Personal computer.


2. All reports - a) Readings Report, b) Calibration Report and c) Individual reading report and Data
logging Report (without Graph) which are available for printing can be transfer to PC on-line/off-
line (needs Hyper terminal active in the PC).
3. The data is captured as ASCII character via Hyper terminal software and then can be saved by
the user as MS word .doc or .text file by Note Pad with suitable name by using Cut Paste utility.
Since the communication is based in Terminal emulation mode for supporting Table borders – the
Font should be selected as “Terminal” only.

COM Port Configuration/Settings:-

TERMS OF WARRANTY

LABINDIA ANALYTICAL INSTRUMENTS PVT LTD., hereinafter known as Manufacturer, warrants all their
products manufactured and sold by them or through their authorized distributors, to be free of any defect in
material and workmanship, subject to Manufacturer’s inspection by their authorized personnel within the
following terms and conditions: -

1) Period of Warranty: Free Warranty period is applicable only for those cases where full payment is
received as follows:
(1) 18 months if the full payment is received against supply and presentation of documents.
(2) 15 months if the full payment is received within 30 days of supply irrespective of installation.
(3) 12 months from the date of installation for the cases where full payment is not received till installation.
However, in this case, period between the date of installation and date of receipt of full payment will be
deducted from 12 months and reduced period of Warranty will only be applicable.

2) Scope: (a) Any part found defective by reason of faulty workmanship or defective material
(fair wear & tear expected) will be repaired or replaced free of charge.
(b) All pH/Ion/Conductivity instruments are to be returned freight to pay basis to us on following
address*. We after repair will send them to the customer freight paid. Since these are low
priced items, offering free service at buyer’s place by deputing our Engineer is not feasible.
You are therefore requested to co-operate with this request.

In case you need repair at your place we will depute our Engineer provided you agree to

pay 1 or 2 A/C return railway fare, hotel & incidental expenses, etc. of our Engineer. In
this case please note that we will not charge you for our Engineer’s time, etc. This payment
will be in advance and visit duration will be only for one day. On your request we will be
happy to furnish you our quotation for this repair.

Other instruments will be serviced by our factory-trained engineers at buyer’s place.

(c) Following items are not covered by warranty: -


(1) All Electrical Displays (LCD, LED etc.)
(2) All glass parts like pH Electrodes, Bowls, Slides, Cover Slips, Beakers or Capillaries, etc.
(3) Parts made out of any type of glass such as Lenses, Prisms, Reaction Tubes, Glass
heating elements, etc.

3) Limitations: The Warranty shall not apply if: -


(a) the instrument is subjected to abnormal use or not used, stored or not maintained in
accordance with the instructions given in the user’s manual or by our engineer during
installation, or any damage caused by accident, collision, or fire.
(b) the instrument has been modified in any way.
(c) the claims are made without proof of purchase (i.e. invoice, bill, cash memo, etc.)
(d) if it is noticed that attempts were made to repair the instrument by outsider or customer’s
untrained instrument engineer, etc.
