What Are Stem Cells ?

∎ Stem cells reproduce themselves (self-renewal).


∎ Stem cells generate progeny destined to differentiate into functional cell types.
∎ Stem cells persist for a long time.
∎ Stem cell behavior is regulated by the microenvironment (niche).

Minimal Criteria of MSCs (International Society for Cellular Therapy (ISCT)

(1) Adherence to plastic in standard culture conditions


(2) Expression of the surface molecules CD73, CD90, and CD105 in the absence of CD34, CD45, HLA-DR, CD14
or CD11b, CD79a, or CD19 surface molecules
(3) A capacity for differentiation into osteoblasts, adipocytes, and chondroblasts in vitro

Sources of Mesenchymal Stem Cells (MSCs)

• MSCs can be isolated from different adult tissues preferably :


-bone marrow (BM)
-peripheral blood (PB)
• Adipose tissues (AT) (such as abdominal fat, infrapatellar fat pad, and
buccal fat pad)
• Neonatal birth-associated tissues, including placenta (PL), umbilical cord
(UC), Wharton jelly (WJ), amniotic fluid (AF), and cord blood (CB), and
then stored for future possible applications.

How to detect stem cell markers in the peripheral blood ?

∎ Hematopoietic stem cells : CD34+

∎ Mesenchymal stem cells : CD73+/CD105+, CD90+/CD105+,CD73+/CD90+

Regulation of differentiation of mesenchymal stem cells (MSCs) and their functions
Functions :
∎ Mesenchymal stem cells can differentiate into mesoderm and nonmesoderm-derived tissues.
∎ Immunomodulatory effects.
∎ Key role in maintaining and replenishing endogenous stem cell niches.

Immunomodulatory effects of MSCs

∎ MSCs suppress proliferation of both T


helper (TH) and cytotoxic T cells (Tc).

∎ Trigger differentiation to TH2 and


regulatory T-cells (Treg), resulting in an anti-
inflammatory environment.

∎ Induce monocytes to M2, an anti-


inflammatory effect.

∎ Suppress B cell proliferation, decreasing the


production of antibodies.

What are the mechanism of actions of MSCs ?
Mesenchymal stem cells also contain extracellular vesicles (EV)

• Exosomes : diameter 40-150 nm

• Microvesicles : diameter 150-1000 nm

• Apoptotic bodies : diameter 50-2000 nm

Conclusions

• Stem cells are capable of renewing themselves and differentiation into


different kinds of tissues.

• Mesenchymal stem cells have the immunomodulatory and anti-


inflammatory effects, and can be isolated in culture and peripheral blood.

• Clinical trials have shown the beneficial effects of MSCs in a variety of


diseases such as heart and lung diseases, and osteoarthritis.
