Performance Summary - Inclusivity and Exclusivity
The 3M™ Petrifilm™ Salmonella Express System is used for the rapid qualitative detection and biochemical confirmation of
Salmonella in enriched food and food process environmental samples. The 3M Petrifilm Salmonella Express System consists
of the 3M™ Petrifilm™ Salmonella Express Plate, 3M™ Petrifilm™ Salmonella Express Confirmation Disk, 3M™ Salmonella
Enrichment Base, and the 3M™ Salmonella Enrichment Supplement. For high microbial bioburden samples only, a
secondary enrichment of Rappaport-Vassiliadis R10 (R-V R10) is used.

The 3M Petrifilm Salmonella Express Plate is a sample ready-to-use chromogenic culture medium system that contains a
cold-water-soluble gelling agent and is selective and differential for Salmonella, providing a presumptive result. The 3M
Petrifilm Salmonella Express Confirmation Disk contains a biochemical substrate that facilitates the biochemical confirmation
of Salmonella organisms. The 3M Salmonella Enrichment Broth is used with the 3M Salmonella Enrichment Supplement for
the selective enrichment of Salmonella species in foods and food process environmental samples in conjunction with the 3M
Petrifilm Salmonella Express Plate and 3M Petrifilm Salmonella Express Confirmation Disk.

Method Selectivity – Inclusivity and Exclusivity Testing

Method selectivity testing was performed by 3M Food Safety.

Study Design:

Pure bacterial cultures were derived from purchased lyophilized preparations or from frozen stock cultures.

There were two procedures performed for the Inclusivity and Exclusivity Study.

1. Procedure A –Low Microbial Bioburden Method:

To represent low microbial bioburden sample testing, Salmonella strains were diluted and inoculated into prepared 3M
Salmonella Enrichment Base with 3M Salmonella Enrichment Supplement, and incubated at 41.5°C for 18 hours.

After enrichment incubation, samples were individually streaked onto hydrated 3M Petrifilm Salmonella Express Plates.
Plates were incubated at 41.5°C for 22-26 hours.

After plate incubation, presumptive positive colonies were circled and 3M Petrifilm Salmonella Express Confirmation Disks
were applied. Disked plates were incubated at 41.5°C for 4-5 hours.

2. Procedure B – High Microbial Bioburden Method:

The Low Microbial Bioburden Method (Procedure A) was followed as above with the following exception: after the primary
enrichment, 0.1 mL of each primary enrichment sample was transferred into 10 mL of Rappaport-Vassiliadis R10 (R-V R10)
for secondary enrichment, and incubated at 41.5°C for 8 hours.

After secondary enrichment incubation, R-V R10 samples were individually streaked onto hydrated 3M Petrifilm Salmonella
Express Plates and plates were incubated at 41.5°C for 22-26 hours. After plate incubation, the presumptive positive colonies
were circled and 3M Petrifilm Salmonella Express Confirmation Disks were applied. Disked plates were incubated at 41.5°C
for 4-5 hours.

Results:

Inclusivity - false negative rate


Inclusivity is defined as the ability of a method to detect the target analyte from a wide range of strains. One hundred and five
(105) Salmonella isolates (Appendix 1) were tested. A false negative rate of < 1% (>99% inclusivity) was determined. Only
one strain of one serotype, Salmonella enterica enterica Gallinarum, was shown to be negative by the 3M method.

Exclusivity - false positive rate


Exclusivity is defined as the lack of interference from a relevant range of non- target strains. Thirty two (32) non - Salmonella
isolates (Appendix 2) were tested. A false positive rate of 0% (100% exclusivity) was determined.

The following two tables summarize the results of the Inclusivity and Exclusivity Studies as related to false negative and false
positive rates.
