Methodology

(Anatomical)

1. 10 species fresh sample was collected.


2. Plant was left in fixing solution (FAA) and was left for at least 48 hours or 2 days
before used.
3. Specimens was cut into shorter length which was 2cm length for each stem part.
4. Sectioning process using microtome for certain species.
5. Sectioned part of specimens was immersed in different types of solution.
6. Specimens undergo dehydration process with different solution of alcohol and xylene.
7. The specimens was transfered to a glass slide by using paint brush.
8. The specimens was mount by using Canada Balsam or Euparal and covered with a
cover slip.
9. The prepared specimen’s slides was left dried thoroughly on a slide warmer.

(Morphological)

1. 6 species fresh samples was collected.


2. The mounted specimens were sprayed with alcohol spray all over the whole plant
evenly.
3. The specimens were spread out between the folds of old newspapers or blotting sheets
avoiding overlapping of parts. The blotting sheets with plant specimen should be
placed in the plant press for drying. After 24 to 48 hrs the press is opened.
4. The dried specimens were mounted on herbarium sheets of standard size. Mounting is
done with die help of glue, adhesive or cello-tape.
5. A label was pasted or printed on the lower right hand corner. The label should
indicate the information about the locality, altitude, habit, date and lime of collection,
name of collector, common name, complete scientific name.
