1. List down the materials, reagents and consumables used for the following Blood
Bank Laboratory setup and indicate the use of each equipment.
a. Manual Method (5 points)
● glass slides/test tubes
○ serve as a reaction media for manual testing
○ used in preparation of RBC suspension (glass tubes)
● Microscope
○ lab device used to observe reaction/results that are not visually
evident
● Centrifuge
○ device used in apply centripetal force to promote reaction and
sedimentation of mixtures
● Water bath
○ device used to incubate mixtures and mimic in vivo temperature
environment
● isotonic (0.9%) saline
○ solution used in RBC suspension preparation
● Pipette
○ primarily used for aspiration, dispensing and mixing of substances
● applicator stick
○ used for mixing substances of a test performed using glass slides
● antisera (anti-a, anti-b, anti-ab, anti-D)
○ used to detect presence or absence of blood group antigens
● reverse typing cells (A and B cells)
○ reagents used to detect presence or absence of antibodies in a
patient’s serum
● antihuman globulin
○ reagents that is used to detect presence or absence of sensitized
RBC
● low ionic strength solution
b. Semi-automated (5 points)
● foil seal
○ serves as column cover and prevents column to dry up and
protect it from contamination
● reaction chamber
○ where the sample and RBC reagent are dispensed
● glass beads
○ acts as filtration system that separates agglutinated and
non-agglutinated RBCs
c. Fully-automated) (5points)
● Biovue cassettes
○ various types of cassettes available for different types of test
loaded inside the analyzer
● reagent red cells
○ Affirmagen
○ Known cells
○ Used for reverse typing
● screening cells
○ Used to test patient’s serum against reagent red blood cell
samples
○ Pooled cell, surgiscreen, selectogen
● Identification cells
○ Resolvigen is a software used as an automated software
approach
○ ORTHO anti-sera
● Red cell diluent
○ diluent used in preparation of 0.8% RBC suspension
○ LISS is incorporated into it
● isotonic saline
○ used in preparation of RBC suspension
○ Usually used in preparation of 3-5% RCS
● dilution trays
○ used by the analyzer in preparation of RBC suspension
● BLISS (buffered LISS)
○ acts as potentiator that decreases zeta potential and enhances
affinity RBC with Ab
● For Maintenance
○ 70% isopropyl alcohol, NaOH and 7% BSA
2. Differentiate the Bead from the Gel Adaptation of the Column Agglutination
Technology. (Indicate the differences in the following criteria: the material, resi
stance to centrifugation, and centrifugation time.) (5 points)
● CAT-Beads have a glass beads column and can withstand high speed
centrifugation and requires a shortened centrifugation time of 5 mins. Cat Gel has
a gel column material but is high speed labile and requires longer centrifugation
time which is 10m
3. Describe the features of the ORTHO Workstation. (5 points)
a. For the centrifuge, what is the capacity of the cassette that can be loaded.
What is the required centrifugation time?
● This is divided into 10 slots or 10 cassettes in one go and the required
centrifugation time is 5 minutes for glass beads and 10 minutes for gel/
b. For the incubator part, what is the capacity of the cassette that can be
loaded. What is the advantage of having two panels for the incubator?
● The capacity of the incubator has 2 panels with 10 slots each with a timer
with each panel. Its advantage can be in instances such as when the first
panel is currently occupied and there is an incoming STAT request, there
will still be space for incubation and also a separate timer
4. What is the purpose of the ORTHO Optix reader? What are the features of this
equipment? Describe the accuracy of this device. (5 points)
● This device can be used as a standalone reader or a partner of Ortho
Workstation to visualize reactions in each column of the cassettes on a
connected computer. It features built-in cameras inside of it that is the same
camera used in fully automated devices. It can also be connected to LIS where it
can accept orders from the computer and can transfer results towards the
computer. It is 99.99% accurate.
5. What are the similarities and differences of the Orthovision Swift Analyzer and
Orthovision Max Analyzer? (5 points)
● Both devices are the same in function and the turnaround time is also the same
but the only difference is that the Max can accommodate more samples which is
recommended for high volume laboratories or blood banks.
6. Describe the following screening cells in the Fully Automated Blood banking
procedures: Pooled Cell, Surgiscreen and Selectogen (5 points)
● Pooled cell is a one-cell panel usually used for donor screening
● Surgiscreen is a three-cell panel which is most commonly used in PH
● Selectogen is a two-cell panel
7. In a simplified flowchart, illustrate the procedures of manual compatibility testing.
(5 points)

patient sample (serum) + donor’s red cells from the blood bag + LISS → place in clean
tube then put in water bath at 37 C and incubate (3-5% RCS = ≤ 10m, 0.8% RCS = ≤
15m) → add AHG → centrifuge for 15-30s → read, verify and interpret with the
microscope.

8. What is the purpose of performing Antibody Screening? Enumerate cases that we

need to utilize this procedure. (5 points)
● This test is performed to detect high incidence alloantibodies which are usually
performed on transfusion therapy, pregnant women, and in cases of transfusion
reactions with a patient.
