II. HIGH PERFORMANCE CHROMATOGRAPHY- HPLC


1) Introduction, characteristic features of HPLC, comparison of HPLC and GLC.
2) Principle of HPLC, instrumentation, components, types of detectors.
3) Quantitative analysis, application of HPLC.
4) Super critical fluid chromatography, characteristics of super critical fluids.
5) Instrumentation, components of SFC, comparison of SFC with HPLC and GLC.
6) Application and recent developments.

III. THERMAL MEHTODS OF ANALYSIS – TGA & DSC


1) Introduction, thermogravimetric analysis, thermogravimetric curve.
2) Instrumentation, principle of working, derivative thermogravimetric analysis (DTGA).
3) Factors affecting TGA, simultaneous DTA and TGA curves.
4) Application of TGA and numerical.
5) Factor affecting DTA, application of DTA.
6) Differential scanning calorimetry (DSC).
7) Techniques of DSC.
8) Application and numerical.
9) Thermometric titration.
10) Application and numerical of thermometric titrations.

IV. INFRARED SPECTROSCOPY – FTIR


1) Introduction, range of infra red radiation, origin of infra red spectra.
2) Selection rules, vibration modes of atoms molecules vibration, factor influencing vibration frequencies.
3) Calculation of stretching frequency, instrumentation, components null, pressed pellet and attenuated total
reflection (ATR) technique.
4) Working of double beam spectrophotometer, advances, interpretation of spectra, examination of IR
spectrum.
5) Characteristic group techniques of organic molecules- typical functional groups

V. ULTRA VIOLET SPECTROSCOPY – UV-Vis
1) Introduction, terminology associated with absorption measurements, theory, measurement of the
spectrum, color of the compounds, types of transitions, types of absorption bands.
2) Selection rules for electronic transitions, chromophore concept, Woodward – Fieser rules, Fiesher – Kuhn
rules .
3) Charge transfer spectral absorption, the effect of steric hindrace to coplanarity.

5) Examples- water, food, organic molecules.

VI. ANALYSIS OF CHEMICAL MOLECULES


Organic and bioorganic can be analysed using several equipments. They can be indentified by specific
equipment, they further have to be characterized using other equipment. Water, food molecules,
biochemical, biological, molecular biology materials can be analysed to their specifications and
acceptable limit for routine usage. Any manufacturing unit have to establish their need and comply to
the regulation of the law of the land. For import/export purpose, for regular consumption they need to
be characterized and certified to obtain a test report. So specialized equipment for each functional
requirement are available. Each of them need to have an introduction, construction of the equipment,
operation, maintenance specialized sample preparation requirement, analysis using specific criteria to
meet the specifications. So skill has to be developed by educated personnel in their area of interest and
technical completely has to established for the proper interpretation of the results obtained. Any
number obtained using the equipment has to be interpreted correctly, properly with emphasis as the
measurement accuracy and error considerations.
Topic will be covered with suitable examples and practical demonstration will be given on each
equipment.
