B. Magnification

Eyepiece X Objective = Total Magnification

Scanner 10x
_______ x 4x
________ = 40x
_______________
LPO 10x
_______ x 10x
________ = 100x
_______________
HPO 10x
_______ x 40x
________ = 400x
_______________
Oil immersion 10x
_______ x 100x
________ = 1000x
_______________

C. Draw letter "e"

1. Normal position right side- up

2. Under low-power objective upside-down & backwards,

covering a wider field.

3. Under high-power objective

increase in mag. of lens power means filed
of view gets smaller with the image getting larger.

D. The Ocular Micrometer

1. 1. Compute for the calibration constant of your ocular micrometer. Show

your solution.
100 stage div. x 10 mm = 1000 mm
Under LPO: 1 ocular div. x1000mm = 100 ocular div. x (?) mm
1000 mm/100x = 10 mm

10 stage div. x 10mm = 100 mm

Under HPO: 1 ocular div. x1000 mm = 100 ocular div. x (?) mm
1000 mm/400x = 2.5 mm

1.2. How do these compare with each other?

The calibration of HPO is higher than that of the LPO

2.1. Determine the width (in μm) of the Paramecium (same area). Show your
solution.

Using LPO: 1 div. = 10 mm

19 mm x 10 mm = 190 mm

Using HPO: 1 div. = 2.5 mm

76mm x 2.5 mm = 190 mm
IF. 77 mm x 2.5 mm = 192.5 mm

2. 2. How do these compare with each other?

Even in different magnification of the Paramercium, its width will be approximately be the same.
II. Guide Questions:

1. What is the correct procedure for carrying the microscope?

In order to avoid any mishaps or accidents, it is important to remember to hold the miscroscope with one hand around

the arm or its handle while the other hand is under the base for support. It is the most correct and secure way to hold

walking with said microscope. It is also important to avoid touching the lenses when doing so.

2. Compare the image seen in the compound microscope and dissecting

microscope in terms of magnification, dimensions, inversion of image
and direction of movement. Tabulate your answers.

compund 40x, 60x, 400x 2. dimensional inverted lens could be rotated

microscope 1000x view

Dissecting between 5x & 50x 3. dimensional one lens is unmovable

Microscope not inverted
view and could be zoomed

3. How does one improve contrast when using a dissecting microscope?

One could adjust the field iris diaphragm to improve the contrast and even the resolution of a specimen. Placing suitable

filters or even aperture within the optical path in the illuminating system may also improve its contrast (Sanderson, 2000)
