Assignment for the 3rd practical activity

The aim of this practical activity is to find out which of the six variants of extraction is the best one.

Step 1. Read the presentation of the Practical Activity No. 2 and 3

Step 2. Look at the Excel document, Sheet 1, that contains calibration curve constructed with the
values of OD740nm for each concentration of the five etalon solutions

Step 3. Open Sheet 2 of the Excel document (Laboratory results) and identify the OD values assigned
to you and construct the calibration curve for the five etalon solutions with the concentration
marked with red colour

Step 3. Apply a linear fit (trendline) and obtain the equation.

Step 4. Using the obtained equation calculate the content of total polyphenols with the OD 740nm from
the table at the bottom. Because the calculated results are for the diluted samples, you have to
multiply them with the dilution factor and to divide by 1000 for expressing the total phenolic
content in mg gallic acid equivalents/ml extract.

Step 5. Compare the results for finding the best extraction method (clustered column chart)

Step 6. Compare the results for finding the best extraction solvent (clustered column chart)

Step 7. Compare the results for finding concentration of the extraction solvent (clustered column
chart)

Step 8. Formulate a conclusion

Step 9. Save the document with your name
