1. Scope & Applications:


 Autoclaves Use to prevent internal crowding; the sterilizing temperature up to121°C).
 Autoclaves (stem sterilizer)
2. Definitions:
 No Definitions.
3. Precautions:
 Do not Autoclave Wet (if electric component get wet , it may be broken ,get a shock .
 Wear heat resistant gloves, eye protection, and lab coat.
 To prevent steam burns, make sure that the autoclave pressure is near zero before open the door.
 Allow steam to escape gradually by slowly cracking open the autoclave door. Allow load to cool for
10 minutes before removing.
 Do not autoclaves sealed containers or full bottles with narrow necks as they may explode.
 Do not autoclave materials containing solvents, volatile or corrosive chemicals (such as phenol,
chloroform, bleach, etc.), or radioactive materials.

4. Procedure
4.1 operation
 Check if power cable is connected and press power button.
 Close drain valve by moving lever to anticlockwise.
 Open the lid and Pour water ( about 1.5 l or more of water)
 Load object in stainless wire basket then set in chamber slowly.
 close lid and slide the lid lock
 set operation mode , value of temperature and time
 Press starts key to begin operation.
 Press stop key after operation complete an release the lid lock lever.
 Unload object safely from chamber (below 800c in chamber).
 Turn off power switch by press power button.
4.2 Calibration
Sterilizing efficiency measurement can be tested via two methods:
4.2.1 Biochemical method:
 By getting some bits of paper or cotton saturated of thermophilic microbes as "Bacillus
stearothermophilus in a paper envelopes and putting them in the autoclave.
 After sterilizing we put these bits of paper on surface of suitable media and incubate it at 55 oC. If a
growth took place ………………….sterilizing failure.
 Note, it is well known that theses microbes dead at 121oC for 12 minutes.
4.2.2 Chemical method:
 By putting a sublimated substance as sulfur which sublimate at 120 oC in capillary tubes and hanged
inside the autoclave within sterilizing, if the sulfur get sublimated …..This indicates the Sterilizing
efficiency.

5.1.1 Check of water level in chamber. By transposing of water.


 Turn on power switch.
 Open lid, and empty the chamber.
 Drain with lever of drain valve which is located in lower right of exterior.
 After draining whole water in chamber, close the valve with lifting lever.
 Put on base plate bottom of chamber
 Pour fresh water into chamber up to level of base plate.

5.1.2 Check of pressure gauge.


 Check indicator of pressure gauge points 0Mpa when opening lid. If not, please contact our local
distributor or us.
5.2 Weekly:
5.2.1 Cleaning of chamber and exterior.
 Turn on power switch.
 Open lid, and empty the chamber.
 Drain with lever of drain valve which is located in lower right of exterior.
 Clear impurities in chamber, and rinse inside of chamber in water.
 Wipe exterior with a cloth dipped into ethyl alcohol infiltirating.
5.2.2 Maintenance of lid gasket.
 Turn on power switch.
 Open lid.
 Wipe the gasket with a soft cloth dipped in ethyl alcohol infiltrating.
 Wipe opening mouth of chamber with a soft dipped in ethyl alcohol infiltrating.
