1. What affects the absorbance of a solution?


The amount of light of a certain wavelength that inhibits a substance from going
through is measured as absorbance. There are two main factors that affect the absorbance:
 Concentration of a substance
 Path length

2. What are the UV and Visible wavelength ranges?

3. MATERIALS

 Micropipette
 Micropipette tips
 Test tubes
 Crystal violet (1 M)
 Test tubes
 Spectrophotometer
 Cuvette
 Distilled water (dH 2 O)

4. METHODS

1- 1 M of crystal violet stock solution was given in test tube by the laboratory assistants.
2- 5 different test tubes were prepared by diluting the concentration of solution taken in
each test tube by 1/2.
3- All test tubes were prepared by diluting the concentration and filled into
spectrophotometer cuvettes and placed into spectrophotometer compartment.
4- The solution of unknown concentration was added to the cuvette and put into the
spectrophotometer machine, and the absorbance value and molarity was measured.
5- Absorbance values are measured from cuvettes placed in the spectrophotometer.

Stock tube => 1 M


First test tube => 0,5 M
Second test tube => 0,25 M
Third test tube => 0,125 M
Fourth test tube => 0,0625 M
Fifth test tube => 0,03125
Unknown test tube => 0,616754 M
