Test Method for Sodium Ascorbate

1. Identification

1.1 Specification: Positive

1.2 Method

1.2.1 The sample solution (20mg/ml) slowly reduce alkaline cupric tartrate TS at 25℃,

and does so more readily upon heat.

1.2.2 Acidify 2ml of the sample solution (20mg/ml) with 0.5ml of 0.1N HCl, add 4 drops

of methylene blue and warm to 40℃, a deep blue color forms and disappears almost

completely with in 3 minutes.

1.2.3 Dissolve 15 mg sample in 15 ml of 5% trichloroacetic acid water solution, add

about 200 mg of activated carbon, shake for 1min. and filter through a small fluted

filter, returning the filtrate, if necessary, until clear. Add 1 drop of pyrrole to 5 ml of the

filtrate and agitate until dissolved, and then heat in a water bath at 50 ℃, a blue color

appears.

1.2.4 Sodium identification test pass.

2. Specific optical rotation

2.1 Specification: +103°~ +108°

2.2 Method: Weigh accurately 5.0g of substance being examined, Dissolve the

sample with 50.0ml of carbon dioxide-free water, and test it with Polarimeter.

3. PH

3.1 Specification: 7.0 ~ 8.0

3.2 Method: Take 2.0g of the sample, dissolved and diluted to 20 ml with carbon

dioxide-free water, then tests it with pH meter.

4. Loss on drying

4.1 Specification: ≤0.25%

4.2 Method: Dry it in a suitable vacuum drying tube, phosphorus pentoxide being used

as the desiccant, at about 60℃ for 4 hours.

5. Assay

5.1 Specification: 99.0 ~ 101.0%

5.2 Method: Dissolve about 400 mg of Sodium Ascorbate, accurately weighed, in a

mixture of 100 ml of carbon dioxide-free water and 25 ml of 1mol/L sulfuric acid.

Titrate immediately with 0.05mol/L iodine VS, adding 3 ml of starch TS as the

endpoint is approached. Each ml of 0.05mol/L iodine is equivalent to 9.905 mg of

C6H7NaO6.

6. Heavy metals

6.1 Specification: ≤0.002%

6.2 Method: Method II, USP <231>

7. Pb

7.1 Specification: ≤2ppm

7.2 Method: Take 5.0 g of the sample add 0.1 mol/L nitric acid solution and dissolved

to 25.0 ml, then test it using Atomic Absorption Spectrometer.

8. Appearance

White to yellowish crystal or crystalline powder.
