Executive Summary for Growth Study of Microorganism in Shake Flask

The study of bacterial growth curves is one of the most crucial components of the introduction

to microbiology. The goals of the experiment are to develop a growth curve with lag, log,

stationary, and death phases and analyse and study the kinetics of microbial growth in shake

flask studies. The growth kinetics of microorganism is studied using the shake flask

fermentation experiment by plotting the graph of real absorbance optical density versus time.

This experiment is done on 4th January 2023 until 6th January 2023 and conducted at the

biochemical laboratory.

The terrific broth medium is inoculated with the yeast (Saccharomyces Cerevisiae) for

20 minutes at 121 degrees Celsius. The media were then grown for 4 hours at 300 rpm. To

prepare the cell culture, 2mL of yeast solution were taken from the flask and were added to the

sterilized media of 150mL in 1000mL shake flask. At intervals of one hour, the desired sample

volume was transferred from the shake flask to the sampling tube. Sampling is done every one

or two hours using a sampling tube. To determine the optical density and total cell number, 1

mL of the sample were obtained (biomass concentration). The spectrophotometer is used to

measure the absorbance optical density reading.

As yeast became acquainted with a new environment, the net growth rate for the lag

phase increased from t = 0h to t = 8h. The exponential phase occurred between t = 8h and t =

23h as the cell began to grow rapidly. Then, there’s no stationary phase occurred until t = 23h

as the cell remained active. Addition, the maximum growth rate μmax = 0.0938h -1 and doubling

time at 7.39h were discovered. Unfortunately, due to limited sources of data, the yield

coefficient (YX/S) and saturation constant (Ks) cannot be determined.
