1. Describe the nuclear structure for E. histolytica


● Have 4 nuclei that characteristically have centrally located karyosomes and fine,
uniformly distributed peripheral chromatin
● Peripheral chromatin is fine, uniform, and evenly distributed. Elongated,
chromatoid bodies with bluntly rounded ends may sometimes be found.

2. State the microscopic characteristics used to differentiate E. histolytica from E.


dispar.

● Entamoeba histolytica and Entamoeba dispar are morphologically identical


species.
● Only when ingested red blood cells are present in trophozoites of E. histolytica
● The use of the polymerase chain reaction (PCR) to amplify amoebic DNA and
identify genetic difference

3. How is E. hartmanni differentiated from E. histolytica


4. List and describe the characteristics used to differentiate cysts of E. histolytica from
E. coli.

E. histolytica
● Spherical
● 10-20 μm;
● (usually 12-15 μm)
● 1-4 nuclei
● Peripheral chromatin: fine, evenly distributed
● Karyosome: small, compact, usually central
E. coli
● Spherical or oval
● 10-35 μm
● Mature cyst: 8 nuclei (occasionally as many as 16)
● Peripheral chromatin: course, granular
● Karyosome: large, eccentric or central

● Uninucleated
● Large glycogen body that stains with iodine

6. Compare and contrast the cause and diagnostic features of PAM and GAE.

Acanthamoebic CNS infection is also known as granulomatous amebic encephalitis


(GAE). Primary Amebic Meningoencephalitis (PAM) is a disorder that manifests as
headaches, seizures, stiff neck, nausea, and vomiting when the ameboid trophozites of
N. fowleri enter the brain and quickly wreak havoc. Initial complaints from patients may
include fever, headache, sore throat, nausea, and vomiting. convulsions and stiff neck
are meningitis follos symptoms. Also, the patient frequently complains of altered smell
and taste, a clogged nose, and Kerni's sign, which is the inability to fully extend the leg
when the hip is flexed at 0 degrees due to hamstring stiffness.

7.Differences between E. Histolytica and B. Coli


● Histological feature of E.Histolytica Flask-shaped ulcer: because there’s a small
defect in the mucosa but a larger area of necrosis in the submucosa and
muscularis layer
● Inverted flask-shaped ulcer: Balantidium coli
● Gradual onset of abdominal pain and diarrhea in contrast with B.Coli which has
a short incubation period
