SEMINAL FLUID ANALYSIS

1. Upon receipt of the specimen, note the time of ejaculation and the time the specimen has been
received.

2. Note the volume of the fluid. Normal volume of the fluid is 3-7 ml per ejaculation.

3. Note the color. Normal color: pearly white, opaque gray to yellow due to high flavin content.

4. Note the odor, which has distinct musty, chlorox-like or acrid odor.

5. Note the reaction of the specimen, which is mildly alkaline with a pH of about 7.7. Alkalinity favors
fertilization.

6. Note for the viscosity.

7. Note for coagulation and liquefaction time.

8. Proceed to the microscopic examination of the specimen. Note for morphology and motility of the
spermatozoa.

Chilled Distilled Water
NSS
9.2 Method: In a WBC pipette, draw the specimen to 0.5 mark and chilled dist. water to 11 mark to
obtain
a 1:20 dilution. Let mixture stand until mucus dissolves. Shake thoroughly and fill the
blood counting chamber. Count 1mm area, one large square and multiply result by
200,000 to obtain number of spermatozoa per mm.

10. Official reports are signed by the Medical Technologist and Pathologist before releasing to the
general receiving and releasing area.
