9 MEASUREMENT PRINCIPLE
During measurement of samples, electric current passes between the electrode(+) inside and
the electrode(-) outside in Chamber through Orifice. A sample in Chamber is suctioned by
regular negative pressure. Volume of resistance changes depending on volume of blood cells.
Blood cells are sorted out depending on size and volume. By diluting a sample properly, RBC
and PLT are differentiated as volume of RBC is different with that of PLT.
WBC can not be counted properly just by dilution because the number of WBC is much smaller
than other parameters such as RBC and PLT. So Lyzing Solution must be put into a sample to
clear RBC before WBC count.
WBC can be classified into some subordinate cells such as Lymphocyte and Granulocyte.
Lyzing Solution also affects WBC itself, and it makes WBC contracted. By this action of Lyzing
solution, these WBC subordinate cells can be discriminated by the volume of each cell with
the method of Electric Resistance.
