The document discusses preparing a standard solution of phosphorus by diluting a stock solution into 5 standard solutions with increasing concentrations ranging from 0.2 to 1.0 mg/L. The absorbance of each standard solution was measured using a UV-Vis spectrophotometer. A linear calibration curve was constructed using the least squares method to relate concentration and absorbance. An unknown sample was also measured and its concentration determined using the linear equation.

In conclusion , The standard solution of phosphorus can be prepared by dilution of
stock solution. The stock solution was diluted into 5 standard solution. The concentration of standard 1 is 0.2 mg/L , following by 0.4 mg/L for standard 2 , 0.6 mg/L for standard 3 , 0.8 mg/L for standard 4 and 1.0 mg/L for standard 5. Then, allow 30 minutes for all the solution for colour development. Next , the average absorbance for 5 standard solutions is 0.105 for standard 1 , 0.207 for standard 3 , 0.390 for standard 3 , 0.493 for standard 4 and 0.547 for standard 5. The standard deviation for 5 standard solutions is 0.004 for standard 1 , 0.006 for standard 2 , 0.002 for standard 3 , 0.003 for standard 4 and 0.015 for standard 5. The absorbance for all the solutions calculated by UV-Vis spectrophotometer. To find the
gradient of the equation , we used m=
( Σ x i y i− n ) Σ xi × Σ y i
. The value Σ x i is 3.00mg L- and
2 2 ( Σ xi ) Σ xi − n 2 the value Σ y i is 1.742. The value of Σ x i and Σ x i y i is 2.2 and 1.2792. From all the value , we can get the value of gradient ,m. The linear equation for y=0.5831x-0.00155 by using least square method. From the graph , it can be seen that , the higher the concentration of solution , the higher the absorbance. Beside that , an unknown sample also has been prepared by the lab instructor. The absorbance and the concentration of unknown sample are calculated from the linear equation. After the calculation , the absorbance of the unknown is 0.410 and the corresponding concentration is 0. 7058mg L-1
