Aim: To isolate DNA from given Plant material(Papaya/green peas)

Requirements:
Papaya, 100 ml beaker, liquid detergent, salt, glass rod, chilled ethanol, boiling tubes/test tubes,
filter paper, funnel, blender and spool.

Principle:Recombinant DNA technology has allowed breeders to introduce foreign DNA in


other organisms including bacteria, yeast, plants and animals. Such organisms are called
Genetically Modified Organisms (GMOs). Thus rDNA technology involves isolation of DNA
from a variety of sources and formation of new combinations of DNA.

Firstly, make an extracting solution.


1. Take 2 tablespoon Salt and 80ml of plain water in a 100 ml beaker. Dissolve the salt in
the water properly.
2. Now take 10ml of liquid detergent with a measuring cylinder. Add it to the salted water
solution.
3. Add distilled water to the beaker then to make the total amount of liquid to 100ml. The
extracting solution is ready.
Secondly, take a papaya
1. Make a paste of papaya by mixing it in the blender.
2. Take about 10gm of the papaya paste and keep adding the extracting solution till the
100ml of extracting solution finishes. Now, filter the papaya solution.
3. Take 10ml of filtered papaya solution in a large test tube.
4. Then add 10 to 20ml of chilled ethanol in the papaya solution by the wall of the test
tube. Do not pour it in the middle of the test tube. Do not shake it too.
5. A mucus kind of thread can be seen in the test tube. Pull it up through a capillary tube,
and that's the DNA of the papaya.
Observation:
The DNA appears as white precipitates of fine thread on the spool.

Inference:

Thus DNA can be isolated from the plant cell nucleus by this technique.

Precautions:

· All the glassware must be thoroughly cleaned and dried.

· The chemicals used for the experiments must be of standard quality.

· If ordinary ethanol is used, the time duration for obtaining precipitated DNA may extend

further.
