analyzer for service engineers of Erba Lachema and their partners.
All information contained here is the intellectual property of Erba Lachema and should not be
used or reproduced without prior agreement of Erba Lachema, the manufacturer.
This manual was written with the intention to give the most precise and up-to-date, detailed
description of operation and use of the analyzer for laboratory purposes.
Descriptions contained here are relevant to ECL 105, software version 1.0

INTRODUCTION
To be well up in the instruments, please read this manual carefully to have the knowledge
for servicing the instruments perfectly and avoid extra costs and wasting precious time.
In this manual, we are using the following conventions:
reader, analyzer – stands for ECL 105
This ECL 105 Service Manual contains the functional descriptions of all analyzers, operation
of the mechanics systems, adjustments and settings, and very important information for the
Service Personnel about the service operations and possible problems.

1.1 Name and serial number


Name: ECL 105 Coagulation Analyzer
Serial No.: Every instrument has its own serial number, which is printed on the rear panel
label.

1.2 Integrated software and available information


The integrated software controls the instrument operations, displays, stores, recalls data, and
allows the user to perform QC procedures and modify the user settings. The software version
number can be read out from the from the Self test information during starting.

Every software version is upgradeable (using an special software and USB pen drive) by the
latest program developed by Erba Lachema, and it can be downloaded from:

2 FOR YOUR SAFETY

2.1 Who Should Use This Manual


This user manual is intended for Erba certified Service Engineers providing product support
for the Erba ECL 105 coagulation analyzer. The manual includes information about the
operation, maintenance and servicing of the ECL 105 analyzer. It also contains steps
necessary to perform the setup procedures to tailor the operation of the analyzer to the
requirements of your laboratory.This manual also maintenance requirements to keep the
analyzer functioning properly.

2.2 Special Symbols Used In This Manual

materials, like controls and calibrators, as potentially
infectious. Observe appropriate precautions when

2.3 General Precautions


Make sure to retain the original packaging material for safe transportation and
storage in the future.
To ensure reliable operation and reliable results:
 Only human samples should be analyzed
 Required maintenance (user and service level) should be performed as
recommended in this manual
 Only Erba certified service personnel should perform service actions
 Only genuine Erba service materials and spare parts should be used
Only Erba certified service personnel that have successfully completed the
‘ECL 105 Service Training‘ program are qualified to service the analyzer.
The IVD equipment complies with the emission and immunity requirements
described in relevant part of the IEC 61326 series.
Electromagnetic environment should be evaluated prior to operation of the
device.
This analyzer contains electronic components. Please handle
electronic waste adhering to local or federal regulations.

2.4 Environmental Factors


Operate the ECL 105 analyzer within the ambient temperature range of 17-32°C° and a relative
humidity range of 10% - 80%. The optimum operating temperature is 25°C (~77°F).
Avoid exposing the ECL 105 analyzer to direct sunlight or to extreme high or low temperatures.
If the ECL 105 analyzer was subjected to extreme temperatures during shipment or storage,
the analyzer must be placed for at least one hour in a room whose temperature is within the
operational range before installation or use.

2.5 Electrical Requirements


The analyzer should only be operated from a wall outlet meeting these power input
requirements:
 100-240 VAC and 50/60 Hz
 Power Consumption: 45 Watts
 In-Rush Power : 150 VA or less
Please ensure that the wall outlet is also capable of supplying the power consumption of any
additional devices (such as a printer).
Only the adaptor supplied with the instrument should be used. If the power network is not
reliable, contact your representative for options such as the installation of an external UPS
module.

The instrument should not be placed near potentially interfering devices capable of emitting
radio frequencies (e.g. radio or television transmitters/receivers, radars, centrifuges, X-ray
devices, fans, etc.).
This analyzer is designed to be safe for transient voltages to INSTALLATION CATEGORY II
and POLLUTION DEGREE 2.

2.6 Waste Disposal


Used diagnostics cuvettes contains human plasma and should be considered to be a potential
infection and biohazard threat. Safe laboratory practices must be followed including the use of
personal protective when operating the ECL 105 and handling samples and used cuvettes.

2.7 Emergency Situations


Always follow all applicable laws and regulations with regard to emergency situations.
If the ECL 105 needs to be powered off due to an emergency situation (like fire, thunderstorm
etc.), follow the procedures in user manual.

In case of fire, do not use water to extinguish the fire unless the ECL 105 is
disconnected from the electrical network!

Optical scheme and principle

3 OPTICAL SCHEME AND PRINCIPLE

3.1.1 Determination of Clotting times


The detection of clotting time is done optically, by measuring the scattered light generated by
the formation of the fibrin clot in the mixture when sample and necessary reagent(s) are
combined respecting the necessary incubation times. The light emitted at 640nm by a LED is
directed towards the reaction cuvette and the scattered light intensity is measured at a 90°
angle by a photo detector. It is weak at the beginning of the reaction which starts immediately
at the addition of the starter (the last reagent to be added), it then increases gradually as the
clot formation takes place and reaches a steady value once the clot is fully formed. The
algorithm that monitors the scattered light intensity in real time stops the measurement when
the measurement becomes stable. Different algorithms can be applied to best fit the type of
reaction seen with the different parameters. The clotting time is then determined from the
measured reaction curve.

3.1.2 Immuno-turbidimetric measurements


The turbidimetric measurements are done using a LED emitting at 800nm. A photo detector
positioned on the other side of the cuvette measures the intensity of the light after crossing the
reaction mixture. The intensity of the light will vary as the reaction evolves and the Optical
Density (OD) is calculated. The Delta OD (dOD) is calculated from the beginning of the reaction
to the end. Using a calibration curve with known concentrations, the patient sample is
calculated in the calibrators’ unit. This measurement method is intended for immunologic

reactions using latex based reagents that are compatible with 800nm wavelength, typically D-
Dimer. During these reactions, an immune complex antigen-antibody will form and create
optical turbidity which will decrease the light intensity captured by the photo detector. This
change in light intensity transformed in delta OD is proportional to the concentration of the
tested analyte present in the analysed sample.

1) Install the measuring block onto the incubator with socket head metric screws (2pcs.
M3x25) and spring washers (2pcs. Ø3).

2) Position the detectors and LEDs to the measuring block according to the electronic
BOM list and component placement drawing.

3) Install the measuring block – incubator complex, the detector and the LEDs together
onto the motherboard with pan head cross recessed metric screws (5pcs. M3x8).
After tightening screws, solder the electronic components.

4) Start up the instrument, do the calibration and change series resistors if it is


necessary.

5) Fasten the TO220 housing components and the bimetals to the incubator with pan
head metric screws (3pcs. M3x8), spring washers (3pcs. Ø3) and with the flake
(1pc.). Positioning has to follow the electronic BOM list and component placement
drawing. Heat transfer paste has to be used between the incubator and TO220
housing components and bimetals. After tightening screws, solder the electronic
components.

6) Solder ferrites onto the motherboard according to the electronic BOM list and
component placement drawing.

2) Connect the foil cable of the printer to its socket on motherboard.

3) Connect the foil cable of the display to its socket on motherboard, and then place the
display on its standoffs.

4) Assemble the upper case and the printer cover.

5) Place the upper case on the base and fasten them together with pan head cross
recessed PT screws (5pcs. KA 35x10).

Algorithm

6 ALGORITHM
 50%: The device takes a measurement every 40 ms (Chyba! Nenalezen zdroj
odkazů.). The measurement holds up till the second read time plus one second. The
result will be where the first and the second read time’s average AD value is equal to
the measurements AD value.
 BestFit: The device takes a measurement every 40 ms (Chyba! Nenalezen zdroj
odkazů.). The measurement stops automatically when the signal AD value changes
less than 16 value in 4 sec and the signal changed minimum by 10 %. The device
continuously loads a histogram. The histogram’s resolution is 8 AD value. The
histogram will have two maximum, the start condition and the clotted condition (1.
fig.). The search function make a one step function with the two maximum’s AD value
and the width is 4 sec. . Than the routine push it through the measurement’s curve. It
searches that point where the area bounded by the two functions is minimal.
(2. fig. and 3. fig.). That point will be the coagulation point.
 BestFit APTT: Same algorithm like the BestFit. The difference is the stop condition. It
will be stop when the signal changed by 80%.
 BestFit Fine: Same algorithm like the BestFit. Chyba! Nenalezen zdroj odkazů.).
The measurement stops automatically when the signal AD value changes less than
16 value in 12 sec and the signal changed minimum by 10 %.
 Fibrinogen: Same algorithm like the BestFit. The difference is the histogram stores
every AD value.
 dOD: Do a measurement at the first and the second read time. The result will
calculated with the dOD = log10((AD1/AD2)) formula.

8 SERVICE MENU DESCRIPTION

8.1 Passwords
On the System service menu you can log in with differential passwords.
1. User password: 4895
2. Service password: 1270
3. Admin password: 6231

1) User level:
a) You can remove all QC data.
b) You can remove all result data.
c) You can set the device to the factory defaults. The device will preserve the stored
quantities.
2) Service level:
a) You can use all of the user level methods.
b) You can edit the device properties and test the peripheries.
3) Admin privilege
If you type the password in, you will have admin privilege, so you can:
• start measurements even if there is no cuvettes, reagents added to the device or
reagents are expired;
• start measurements even if the blocks aren’t incubated yet to the target temperature;
• modify factory methods.

8.3.1 MEAS

8.3.1.1 Measurement test

In this tab you can test the device’s measurement periphery.


 The table displays the measured AD values and the programmable amplifier’s gain
state.
 Whit the LED change you can turn on the usable LEDs.

Red LEDs are turned on.


Use the 830 Sysmex reference cuvette.
Required AD range: 280 – 470.

8.3.1.2 Incubator test

 In this tab you can test the device’s incubator block.


 There is real time data in the table.
 The table displays the PWM’s duty cycle, the controller’s P, I, D parameters, measured
AD value and the temperature in °C.
 You can set the target temperatures under the table, but these will be saved only until
the device is turned on.
 You can calibrate the temperature, if you write your measured (with a thermometer)
results in measured edit field, then click on set button

8.3.2 Periphery

8.3.2.1 USB

In this tab you can test the USB’s communication. If you connect a mass storage to the device,
it will detect the USB key and the test will be available in the software.

 Mass storage test: The device will try to create a simple text file on the mass storage
and try to write it. If it’s failed, the circle next to the button will be red, otherwise it will
be green.
 If you connect an USB keyboard, the device will detect it and the test will be available
in the software.
 Keyboard test: If you type something on the keyboard, it will be displayed in the text
box.
 USB B test: Connect the device to a PC on a USB B connector the device will emulate
a virtual COM port. First use you must have the ST-VCP driver. USB B test is running
an echo client. Please use a simple terminal client (ex. Realterm). If you send a string
to the device, it wills response “Response: + received string” answer.

8.3.2.2 Ethernet

If you connect an Ethernet cable, the device detects it and displays the IP addres.

8.3.2.3 Printer
In this tab you can test the thermal printer.
 Print blank line: The printer will print a blank line.
 Print test page: The printer will print a test page.
 Paper sens. status: If it’s red, the printer doesn’t sense paper. If it’s green the printer
senses the paper.
 Door sens. status: If it’s red, the printer senses that the printer cover is open. If it’s red,
the printer senses that the printer cover is closed.
 Printer temp: Displays the printer head’s temperature.

8.3.2.4RFID

If you put an RFID tag next to the RFID reader, the device detects it, tries to write on it, read it
back, and then compare the R/W results. Each step is indicated with green/red dot, depending
on if the result is good or not.

8.3.2.5 Serial

Connect the device to a PC on a serial connector. RS232 test is running an echo client. Please
use a simple terminal client (ex. Realterm). If you send a string to the device, it wills response
“Response: + received string” answer.

In this tab you can see the real time clock.


 Battery voltage: displays the RTC’s battery voltage. The voltage must be in 3.1V – 3.4V
range.
 Time: Displays the actual time on the device.

8.3.3 System

 Remove QC results button


All stored QC results will be deleted.
 Remove QC results button
All stored sample results will be deleted.
 Factory defaults button
The device will return to the factory default parameters.

9 TROUBLESHOOTING
9.1 No backlight
 Check the cable connection.
 Measure the supply power on the C18 terminates. The supply range should 15 – 22V.

 If it’s 7.5V the supply IC doesn’t work, it should send back the mainboard. If the range
is good replace the LCD, the LCD assembly you found on page 10.

9.2 Blank LDC or faulty display


Check the connection of the LCD. If its good try to replace it. The LCD assembly you can found
in 5.5

9.3 No touch response or wrong touch response


• Check the display gasket(a008) and the display position. Check the connection of the
LCD touch cable.

9.4 Printer doesn’t print some characters


• Check the cable connects of the printer module. If it seems good replace the printer
module.

9.5 Time and date lost after turning off


• Replace the battery with CR2032.

9.6 RFID doesn’t read


• Check the connection of the module. Log in the service menu. Approximate an RFID
tag to the reader.

9.7 LED or detector doesn’t work properly (Wrong sample results)


• Check the working of the LEDs and detectors. Do the measurement test.
Guide of test, you can see 8.3.1.1.
• You can check light of red LED by visually.

9.7.1 Replace the defected LED:


 Bend out the LED and check the light by visually.

 Replace the LED, if it necessary - Pay attention the polarity!

9.7.2 Set the current of LED with series resistor


(according to the wrong test result):
Notice:
 Use ESD protected equipment for repair.
 Shut down the power supply, before repair actions.

 Solder out the resistor and choose another, nearby value.


 Try a new resistor value (Put it on the pad with tweezers) and do the measurement
test.

 If the measurement test is good, than you can solder new resistor.
