Vibrio Fischeri Is A Bacterium That Served As The Model...
Vibrio fischeri is a Gram negative bacterium that served as the model organism in this
experiment. We isolated only the luxAB portion of the entire lux operon and inserted it
into the pGEM vector plasmid to transcribe the luciferase enzyme required for
bioluminescence. Within the lux operon, luxAB is responsible for producing subunits
that form luciferase, which oxidizes the aldehyde made by luxCDE into the reduced
flavin mononucleotide FMNH2 and results in the production of light known as
bioluminescence[6]. The luxI genethat is also present on the operon produces the
autoinducer which interacts with a regulator, LuxR, [and] activates transcription of the
lux operon at high cell density to induce bioluminescence[4]. This interaction between the
products of luxI and luxR is known as quorum sensing and serves as an evolutionary
purpose in the symbiotic relationship with Euprymna scolopes, a type of squid that
contains the light organ where the bacteriaresides[6].
In the first portion of the experiment, Vibrio fischeri chromosomal DNA was isolated by
lysing the bacterial membranes and removing the proteins and lipids. Addition of a
lysozyme solution breaks down the peptidoglycan layer in Gram negative bacteria and
proteinase K/SDS degrades the proteins and disrupt the membranes. Liquid liquid
extraction using Tris buffered phenol permits separation of proteins and lipids from the
DNA into the organic layer. It is also essential that the isolated DNA is pure and free of
Essay on Servants in Victorian England
Servants in Victorian England Servants were imperative to the functioning of middle
and upper class homes in Victorian England. Without the veritable army of servants for
the upper and upper middle classes, women would not be able to live the leisured lives
they had grown accustomed, and would certainly not have the time to flaunt their status
with neighbor calling and the numerous balls and social activities. Even most lower
middle and middle middle classes employed at least one servant, as assistance was
almost a necessity in maintaining the home (Roberts 205). For the most part, these
servants had an appreciation for their work, with the opportunity to live in an upper class
home and have job security, as the alternative was... Show more content on

Women were dependent on their maids, as performing a task as menial as dressing
themselves would not be viewed as proper for a lady of status. The lowest servants
would be the footman and housemaids, whose duties involved mostly cleaning,
scrubbing, and polishing silver. The size of the home determined how many footmen
and housemaids would be employed. In the beginning of the century, laundry maids
were common, which is believable that with all the garments worn, especially by the
lady of the house, that a person could be employed just to wash the family s clothes.
By the end of the century laundry maids were less common with the advent of
professional laundries. If a family owned a stable, a staff of servants including a
coachman, a groom, and a stable boy were required (Roberts 206). Other outdoor
servants may include game keepers and huntsmen, who were responsible for hunting food
for the family (Margetson 167). Although frequent in Victorian literature, governesses
were not as common in reality, representing a small portion of the servant population.
The education required by governesses, and the fact that they were usually selected from
upper classes, allowed them to be viewed as slightly higher status than the low servants
(Roberts 206). For the maids of the household, their duties consumed nearly all of their
time. Their day would typically begin at 6am and may not have ended until after midnight
I Sat Down For A Discussion With One Of The Attorneys At...
In August 2016, I sat down for a discussion with one of the attorneys at Hotze Runkle,
Justin Dunlap. Mr. Dunlap had just joined the firm a month earlier as part of the firm
s expansion into pharmaceutical litigation, an area of practice that he has had years of
experience prior to joining the firm. He graciously took the time one afternoon to sit
down and allow me to conduct an interview with him. First, I asked him to discuss his
educational background and how he began to practice law. Mr. Dunlap, a native of the
Dallas area, completed his undergraduate degree at the University of Kansas where he
studied history. He said that while he thoroughly enjoyed his degree program s content,
he did not see a future career for him with history and given that many of his older
family members, including his mother, were practicing lawyers, law schoolseemed to
him to be a natural next step for him. He returned to Dallas to get his law degree from
Southern Methodist University (SMU). He said that, he felt extremely lucky that his
parents were able to help finance his legal education, so finances were not a major factor
in his law school decision. When I asked him how he came to choose SMU, he
indicated that the two most prominent factors were the location in the state he wished to
practice and the school s estimable reputation in Texas and the surrounding states. Mr.
Dunlap told me that once he got to law school, he was sure that he wanted to work for
the JAG Corps once he was
The Flaws In The Movie Kingdom Of Heaven
The movie Kingdom of Heaven is a very good movie. It is biased off of the crusades
more specifically the second crusade. However, no matter how good any movie is
there is always flaws, this one is no different. The amount of flaws I am able to choose
from is tremendous. I have categorized them into three main topics. First I will be
talking about the false things that have to do with Balian himself. Next will be the
weapons them self s and how they fire/are used and outcomes of them. Last I will state
many of the other false tings in the movie and back up why they are false. In the
movie there are many incorrect elements about Balian and surrounding Balian. One of
the larger issues I can see is that he is not from France. As Helena P. Schrader says
Balian was born in the crusader Kingdom of Jerusalem. The movie started out with a
scene of Balian burying his dead wife. Several men ride by on horses and one says She
was a suicide cut her head off... but she really never committed suicide. However,... Show

First up is the longbow, a great invention but sadly it was not yet of this time. Believe
it or not the long bow wasn t on the Europe main land until many years after it was
invented. According to Max Perham the longbow was invented by the Celts in Wales
around 1180, but was not really used by the English military until the 1300 s. Also
according to Virginia Ream the army s attacking would not just bombard the castle
walls. There were people that would dig under the castle s walls to weaken them then
they would fire at that area to bring them down. I found more information on this mining
strategy on medieval castles.net. The large battle scenes were almost funny on how the
catapults and trebuchets they had fired. It looked more like a cannon firing the
projectiles. Nevertheless, these machines fire more like a rainbow. You can find more

An Interview For The Univision Media Network
After Zorro, people spoke Spanish to me for ages. I m Welsh but that movie instantly
gave me a new ethnicity. This was a quote by the actress, Catherine Zeta Jones, when
asked to speak Spanish in an interview for the Univision media network. This is one of
the many results that come from of a complex reality of identity. Individuality is
controlled by a social structure that labels a person based on similarities to a particular
ethnic group. Society has the need to shape the ethnic identity of a person. They are
categorized into a homogenized group defined by resemblances of their nationality
and culture. This results in discrimination and oppression. The fact of a person s
identity has never been neutral. The way a person is treated by society, according to
how they are perceived, is the cause of identifying and shaping an identity with social
factors that provide a negative outlook on cultural tolerance. The complex reality of
identity is a factor that is influenced by the culture a person was raised in and handed
down to by their families. This causes society to take that factor and turn it into a way
of establishing an identity upon people. Due to a resemblance to an ethnic background,
people are compared according to their social, financial, and behavioral status. Society
will always attempt to identify a person to their best knowledge to feel a sense of
belonging and being at a minimal level of thinking with ethnic groups to be comfortable
with racial differences.
Goodbye To My Father
It was June 1917 when I had to say goodbye to my dad. We knew that the war was
going on in Europe, but we didn t know the extent of it until just a few months ago.
When I had to say goodbye to my dad, I didn t know if it was going to be the last time I
saw him or not. I didn t know how life was going to work, I had so many questions.
How were we going to make enough money to survive? Who was going to take care of
the rest of the kids? Will I have to take care of them because I m the oldest? All of
these questions were going through my mind and they were questions that not even
my parents knew the answer to. I didn t know if anybody knew the answers, so I began
to answer them myself. I said well I ll stay home and play the role of my mom while
Mom goes out to work. I ll get my brothers and sisters up for schoolbecause Mom would
already be off to work. Then I would send the rest of the kids off to school while I
stayed back and did the things my motherwould do. The first year or so that dad was
gone things were far from what I would say was the norm , for my siblings and me at
least. We didn t eat as much as we did when my dad was at home, but we still had
enough for three kids everyday. My brother, sister, and I s lives did not change too
drastically, but my mom s sure did. My mom had to start working long hours and she
would still have to do everything around the house that she did before my dad left. I
decided that I would start to help mom with the dishes and get
