Laboratory, Poona. Obtained PhD degree under The guidance

ULTRAVIOLET AND VISIBLE SPECTROSCOPY
Introduction:

➢ The Principle of UV-Visible Spectroscopy is based on the

Absorption of ultra violet light or visible light by chemical

Compounds, which results in the Production of distinct

Spectra.

➢ UV/vis spectroscopy requires electro magnetic radiation Of

high energy.

➢ The Visible Mregion to 800-400 nm and ultraviolet region to

400-200nm.

➢ The uv region extends from 1000-4000Å or 100-400

nanometer.

INFRARED SPECTROSCOPY (IR)
➢ Infrared spectroscopy (IR spectroscopy or vibrational

spectroscopy) Is the measurement of the interaction of infrared

radiation With matter by absorption, emission, or reflection. It

is used to study And identify chemical substances or functional

groups in solid, liquid, or Gaseous forms.

➢ The infrared portion of the electromagnetic spectrum is usually

Divided into three regions; the near-, mid- and far- infrared,

named for Their relation to the visible spectrum. The higher-

energy near-IR, approximately 14,000–4,000 cm−1 (0.7–2.5

μm wavelength) can excite overtone or combination modes of

Molecular vibrations.

➢ The mid-infrared, approximately 4,000–400 cm−1 (2.5–25

μm) is generally used to Study the fundamental vibrations and

associated rotational vibrational structure.

➢ The far-infrared, approximately 400–10 cm−1 (25–1,000 μm)

has low energy and may be used for rotational spectroscopy

and low frequency vibrations.

NMR (Nuclear Magnetic Resonance spectroscopy)

➢ Nuclear magnetic resonance (NMR) spectroscopy is the study

of molecules by recording the interaction of radiofrequency

(Rf) electromagnetic radiations with the nuclei of molecule es

placed in a strong magnetic field.

➢ Nuclear magnetic resonance (NMR) spectroscopy is a crucial

analytical tool for organic chemists. The research in the

organic lab has been significantly improved with the aid of the

NMR. Not only can it provide information on the structure of

the molecule, it can also determine the content and purity of

the sample. Proton (1H) NMR is one of the most widely used

NMR methods by organic chemists. The protons present in

the molecule will behave differently depending on the

surrounding chemical environment, making it possible to

elucidate their structure.

➢ The source of energy in NMR is radio waves which have long

wavelengths, and thus low energy and frequency.

TYPES OF NMR

➢ Two common types of NMR spectroscopy are used to

characterize Organic structure:

➢ 1H NMR:- Used to determine the type and number of H atoms

in a Molecule.

➢ 13C NMR:-Used to determine the type of carbon atoms in the

Molecule.

➢ 1H NMRAND13C NMR

➢ 1H NMR is used to determine the types and number of

Hydrogen atoms present in a molecule whereas 13C NMR is

Used to determine the type and number of carbon atoms in a

Molecule.

MASS SPECTROMETRY
➢ Mass spectrometry is a powerful analytical technique Used to

quantify known materials, to identify unknown Compounds

within a sample, and to elucidate the Structure and chemical

properties of different molecules. The complete process involves

the conversion of the Sample into gaseous ions, with or without

fragmentation, Which are then characterized by their mass to

charge Ratios (m/z) and relative abundances.

Applications of Mass Spectrometry

➢ Environmental monitoring and analysis (soil, water, and air

pollutants, water quality, etc.)

➢ Geochemistry – age determination, soil, and rock composition,

oil and gas surveying

➢ Chemical and Petrochemical industry – Quality control

➢ Identify structures of biomolecules, such as carbohydrates,

nucleic acids
