A. Hydrolysis of Carbohydrates

Solutions Observations Reaction

After the heating process the prepared solution, the


1% sucrose color of the solution remained clear.
After Fehling’s A and B solution was added and the
1% starch solution was heated for few minutes, a light blue
colored liquid was formed at the surface of the upper
layer and in the middle has a foggy green color and at
the bottom is clea.
Next is the conc, HCl it was added to the solution.
Then the solution place at the water bath and boiling
for 30 minutes, a mixture of bluish and greenish
colored solution was formed, and at the bottom of the
solution a clear liquid was formed.

The solution was place on a boiling water bath for
1% glucose five minutes, the lower color of the solution was
formed light yellow and the upper was formed dark
blue and in the middle part was formed dark green.
The lower portion of the solution was formed dark
1% fructose yellow and the middle of the solution was formed
dark green same as the bottom.

After heating the solution for 5 minutes the solution
1% sucrose remained color blue and there’s no red color was
formed at the bottom.
1% starch After heating the solution for 5 minutes the solution
remained color blue and there’s no red color was
formed at the bottom.

C. General Test for Carbohydrates

Molisch Test

1% glucose After the heating process a light purple ring at the junction of the two liquids was
observed.
After the heating process a light purple ring was observed during the conduct of the test.
1% fructose

After the heating process a dark violet ring was observed at the junction of the two
1% sucrose liquids.

1% lactose

Compared to 1 % glucose the solution become clear and it was visible that can be
1% starch identified.

Iodine Test for Polysaccharides

When 3 drops of 1 % of starch solution place in concave of the spot plate and 1 drop of
iodine solution is added the color of the mixed solution change and it turn into greenish-
1% starch
yellow.

QUALITATIVE TEST FOR CARBOHYDRATES

Benedict’s Test

When we added 10 drops of 1 % glucose together with 2ml Benedict’s reagent, and
boiled into the water bath for 3 minutes the color of the chemical changes from blue to
1% glucose
brick red color. Formation of brick-red precipitation was observed within 3 minutes.
Therefore reducing sugars present.

When we added 10 drops of 1% of fructose added with 2ml Benedict’s reagent, and
boiled into a water bath for 3 minutes the color of the chemical changes from blue to
1% fructose
orange.

When we added 10 drops of 1% sucrose together with 2ml of Benedict’s reagent, and
1% sucrose
boiled into the water bath for 3 minutes the color changes from blue to green. During the
boiling session the chemical reacts to it and it changes the color into green.

When we added 5 drops of starch hydrolysate sample together with 2mL of Benedict’s

reagent is added and boiled into the water bath for 3 minutes, the final color changed into

When we added 10 drops of 1% glucose together with 2ml Barfoed’s reagent , and boiled
1% glucose into the water bath for 3 minutes the chemical’s color remains unchanged to blue.
Therefore it is negative for Barfoed’s test

1% fructose When we added 10 drops of 1% glucose together with 2ml Barfoed’s reagent , and boiled
into the water bath for 3 minutes, brick red forms at the bottom of the side of the tube.
No reaction changes.

Seliwanoff’s Test

1% glucose There were no signs of changing the color during and after the boiling session for 3
minutes.
The color changed while being boiled of 3 minutes , from clear turned into light orange.
1% fructose Therefore it shows that fructose is positive for seliwanoff’s test

Bial’s Test

When 2ml Bial’s reagent was added and boiled into the water bath for 3 minutes the
1% glucose color changed into bright yellow.

When 2ml Bial’s reagent was added and boiled into the water bath for 3 minutes the
1% fructose
color changed into dark yellow - brown .

1% ribose When 2ml Bial’s reagent was added and boiled into the water bath for 3 minutes the
color changed into blue - green color.
