This device was examined and left the factory in perfect technical condition. To preserve this and
protect safe and faultless operation, the user must follow the orders and remarks of this service
manual.
To preserve this condition and to guarantee a safe working, informations and working advices,
which are included in this working instructions, have to be heeded by the applier.
The hard- and software is subject to a program of continuos evaluation and improvement and,
therefore, may be changed in the future. This also concerns service requirements.
All samples and reagents should be treated with caution accorded to those known to contain
pathogenic organisms. Similarly, the cleaning of component part of Photometer 5010 should be
done with respect to human health.
This instrument has been constructed and checked in accordance with Standards IEC 1010. When
the instrument leaves our factory it is in perfect condition from the point of view of work safety. To
keep it that way and to ensure safe operation the user must follow the instructions and warnings
given in the operation instruction manual.
The electrical protection of the apparatus corresponds to Class I (it has a protective earth).
The power plug must only be inserted into a socket that has a protective contact. The protection
must not be abolished by using an extension lead that does not have a protective earth wire.
Warning:
Any break in the earth wire inside or outside the apparatus and any loose connection of the earth
wire can make the operation of the apparatus dangerous. A deliberate break or interruption is not
allowed.
When the housing is opened or the components are being removed (except when this can be done
by hand), live parts may be exposed. Connection may also be live.
Therefore, if the carrying out of an adjustment, service or repair on the open apparatus in the live
state is unavoidable, this must be done by an expert who is familiar with the dangers involved.
Make sure you only use fuses of the specified type and rated amperage when replacing the old
ones. Repaired fuses must not be used and the fuse holder must not be bypassed.
If you have any reason to believe that the instrument can no longer operate safely, take it out of
use and make sure no one can use it accidentally.
It must be assumed that safe operation is no longer possible when the instrument:
• fails to operate
• has been stored under unfavourable conditions for a fairly long time
On repairable modules the quality and function is always provided by the manufacturer according
to the latest technology. This keeps Photometer 5010 always on the highest quality level.
The exchange price for modules will be kept on a low level to guarantee repairs, on an economical
basis.
2.2 Conditions
Warranty period The warranty period for instruments is 16 months starting with the date of
for instruments ... shipment ex works Berlin/Federal Republic of Germany or 12 months
starting with the date of the first installation, whichever period is shorter.
... and spare parts For spare parts a warranty of 6 months from installation date of the part
applies. Prerequisite for this is, that the warranty period for spare parts shall
not fall short of the warranty period of the instrument where a spare part has
been installed. A further prerequisite is, that this period shall not exceed a
period of 24 months starting with the delivery of the spare part ex works
Berlin/Federal Republic of Germany.
Hint:
In case the instrument has a remaining warranty period of more than
6 months, the parts remain under warranty until the warranty period of the
instrument expires.
Important information:
− Only parts marked with "A" in the price list are generally accepted under
warranty.
− Only return those parts marked with "R" in the spare part list.
− All defective parts ( non-"R" and "A" parts ) should be kept on stock for
a period of 7 months. In case the manufacturer needs the part for
investigation it will be requested from Berlin.
− All parts returned to Berlin and not requested by Berlin will be sent back
at the expense of the dealer/customer.
Handling of As a general rule, all instrument repairs should be carried out by authorized
repairs and trained personel only.
Repair of instruments
Complete instruments are not accepted for replacement or repair unless
this has been agreed with ROBERT RIELE GmbH & Co KG.
Before replacement or repair can take place, the validity of the request must
be examined and the question of costs must be settled in a written
agreement with ROBERT RIELE GmbH & Co KG.
Terms of delivery All shipping conditions for spare parts to the dealer/customer are
ex. work Berlin.
Emergency In case of emergency, a copy of the complete RA form should be sent by fax
shipment (++49 / 30 / 404 05 29) or electronic mail (e-mail: info@riele.de) to
ROBERT RIELE GmbH & Co KG, Berlin
If fax was used for Emergency RA the original RA form has to be mailed or
sent by electronic mail to Berlin.
Handling of costs Replacements for R-parts are shipped at a repair price. In case the parts are
no longer repairable, the dealer/customer will be charged lateron with the
difference of the new price.
Replacements for warranty requests are shipped free of charge.
In case the manufacturer does not accept the warranty request, the
dealer/customer will be charged lateron with the R-price for R-parts and the
new price for non R-parts.
In case of
instrument out of − Installation date of spare part when part was installed within the last
warranty 6 months.
MODE FUNCTIONS
The mode functions serve for changing definite basic settings during a course of method. It is not necessary to
exit the method and to change to the level of UTILITY PROGRAMS (OM chapter 8). Also temporary parameters
remain unchanged. Corresponding to these features this group of functions is available only within the repetition
sequence of a course of method (chapter 5.5 – DESCRIPTION OF METHOD COURSES).
MODE FUNCTIONS Selection level of the mode functions: Selecting the desired mode function by
direct selection i.e. input of the number key in question (see table) or by scrolling
through all mode functions with [+] ... [+]. Confirm with [ENTER].
Exit the selection level of the mode functions and change back to the course of
method with [BREAK].
After working off a mode function the return is carried out to the course of method.

In the service program level there are functions influencing special basic parameters of the instrument.
Because they are allowed to be changed divergent from the work configuration only in exceptions and only by
trained staff, their unintentional selection is protected by a password.
Service tools selection: Direct selection of desired service tool, i.e. input of
INPUT 1-n/+ concerning item no. (see table) and confirming with [ENTER], or by scrolling
SERVICE TOOLS through all service tools with [+] ... [+].
Abort of service tool selection and change back to program selection with
[BREAK].
A service tool can be broken off with [BREAK] at an any time. Then a change is
carried out back to the selection of service tools.
After working off a service tool the return is carried out to the selection of service
tools.
S1 System initialization
With this function the default parameters will be set.
Start menu by [MODE] [LF] [1]:
Confirm with [ENTER].
Following settings are then typed over in the battery powered RAM. They remain
SYSTEM INIT. S1 active, even after switching the instrument off/on, until they are specifically
SERVICE TOOLS changed. This action ends with a long beep and a print-out of the program
release.

S3 Parameter initialization
Attention! The extended memory for the parameters of all nonlinear methods is deleted by this function without
call-back and the corresponding user defined methods become useless.
Start menu by [MODE] [LF] [3]:

S4 Serial number
Display of the current serial number. A new number can be entered with max. five digits.
Start menu by [MODE] [LF] [4]:
Confirm with [ENTER].
S5 Boost mode
To improve the low light gain at small wavelengths, the measurement time is extended when operating below
a definite wavelength. This definite wavelength will be displayed for approx. 2 sec. when starting this service
program. The default value is 390 nm.
Start menu by [MODE] [LF] [5]:
Confirm with [ENTER].
Enter [0] = BOOST OFF or a value between 300 and 900 as definite wavelength
BOOST ON/OFF S5 in nanometers, below which the boost mode is activated. Finally confirm the input
SERVICE TOOLS with [ENTER].
S6 Filter select
The assignment of filter positions can be made.
Start menu by [MODE] [LF] [6]:
Confirm with [ENTER].
Select filter position by numeric key. Confirm the input with [ENTER].
FILTERSELECT S6 Enter new wavelength by numeric key. Confirm the input with [ENTER].
SERVICE TOOLS Finish the operation with [BREAK].
S7 Automatic Result
With this service program a continuous measuring mode based on calculation procedure 1 (endpoint with
factor) is possible. After the input of wavelength, factor and delay-time and after measuring blank the con-
tinuous measuring mode is started.
Start menu by [MODE] [LF] [7]:
Confirm with [ENTER].
This mode is active until [BREAK] is pressed (and held for a while).

S8 Temperature functions
A sub menu with eight items for temperature functions is called.
Start menu by [MODE] [LF] [8]:
Confirm with [ENTER].
Direct selection of desired service tool, i.e. input of concerning item no. and
TEMP. FUNCT. S8 confirming with [ENTER], or by scrolling through all temperature functions
SERVICE TOOLS with [+] ... [+].
The number of the concerning method must be entered and confirmed with
BCC FROM METH.S9 [ENTER]. Then the BCC is displayed in the decimal form.
SERVICE TOOLS Press [ENTER] and select another number of a method.
Press [BREAK] for exit.
By input of a correction value between 0 and 255 and confirming with [ENTER]
ADC CORRECT.S10 measurements seeming too low can be increased a little bit. This correction
SERVICE TOOLS recognizably affects only at high results (~ 2 Absorbance).
S11 Voltages
Display of the actual voltages on the pcb.
Start menu by [MODE] [LF] [11]:
Confirm with [ENTER].
In the second line the display shows the version of PCB layout, e.g. the value „c“.
Change the value by [+] in case of a deviation and confirm the input by [ENTER].
Leave the window by [BREAK] in case of no change.

Open the Photometer 5010. Separate the top case from the bottom case.
The filter wheel is turned by hand so that the corresponding hole is at the cut-out of the motor fastening plate
(see page 4.10.2).
With the Tool kit filter exchange ( RIELE ID-No. 5010-001 ) a filter exchange can be easily carried out. The
Tool kit filter exchange consists of the tools A and B.

Tool A
With the tool A (left illustration) and a pipe wrench the metal slice can be pushed from the hole in the filter
wheel. The cheeks of the pipe wrench grasp at the tool and at the filter wheel. With a screwdriver the Seeger
ring and the metal slice can be taken.
Set the filter glass into the corresponding hole of the filter wheel. If the filter has only one reflecting surface,
this reflecting surface must be judged to the light source. The Seeger ring can loosely be inserted into the hole
in the filter wheel.
With the tool B (left illustration) and a pipe wrench the filter glass and the Seeger ring can be fixed.

Replacement
The Photometer 5010 has to be opened. The top case has to be separated from the bottom case.
Remove the lamp shield. The lamp shield is fixed with two screws at the bottom case.
Loosen the two screws for mounting of halogen lamp.
Remove the halogen lamp.
Installation
Insert the halogen lamp. Do not touch the glass of the halogen lamp with fingers.
Use the plastic foil within the original packing of the spare part.
The socket of the halogen lamp should be mounted horizontally with direct contact to the lamp holder.
Tighten the two screws for mounting of halogen lamp.
Install the lamp shield.
Press [MODE], [MODE], [ 9 ], [ ↵ ], [ ↵ ], [ ↵ ] for control of ADC-counter value in corresponding filter position
(OM chapter 8.2.9 / AD-Transducer measurands).
Check one after the used positions of the filter wheel.
Select corresponding hole ( e.g. 2 ) of the filter wheel with [MODE], [ 2 ].
The ADC-counter value should be within a range of 3.000 and 600.000.

The halogen lamp has to be adjusted horizontally by the adjustment screw to maximum value of ADC.

The photodiode is situated at the optical unit. The Plug P8 ist connected with the plug ST8 on the mother
board RR-115.
Replacement
The Photometer 5010 has to be opened. The top case has to be separated from the bottom case.
Unplug the photodiode from the mother board RR-115.
Remove the two screws (B).
Remove the adjustment piece (C) of the holder.

Sipping lever:
Remove pin from sipping lever.
Loosen 2 screws of bearing holding the metal tube inlet and remove both.
For installation into new top case proceed in reverse order.
Sliding lid:
Loosen guide rail at the top case and remove it together with sliding lid.
Remove spring bearing.
For installation into new top case proceed in reverse order.
Important: Be informed spring tension is factory adjusted to a certain force.
Special tool and adjustment gauge is necessary.

Remove top cover for printer, paper and ribbon.
Disconnect 18-pin plug CN16 from printer-controller-pcb.
Loosen 2 front screws in the hollows and remove the other 2 screws.
Remove printer mechanism.
Installation
Solder printer connection cable (ID 1707655001) to printer mechanism (ID 1223003001). Take care not to
solder 3 wires which are not used (see drawing). Also care for blue mark.

The basis plate has to be separated from the bottom case.
Disconnect all plugs from the pcb. The plugs are labled with ST1 up to ST11.
Remove five screws fixing the pcb with basis plate.
Installation

Use heat conductor paste for getting cooling effect.

Continue with installation instruction at the end of this chapter in case of changing a board.
Potentiometer P1
An voltage / frequency converter IC13 (AD654) is connected to the output of the amplifier.
The microcontroller can set the amplification of the amplifier stage by the multiplexer IC12 (ADG428). The
eight different settings of amplification are called V0 up to V7. The setting V0 is equivalent to the lowest
amplification.
The micocontroller converts the frequency signal of the voltage / frequency converter to a digital result in a
range between 0 and ~ 600.000. This result is called value of the ADC.
The value of the ADC in the case of photocurrent = 0 (darkness) is called offset. The offset is

maximum. Adjust the AD-transducer value to 500.000 (± 50.000) counts with the potentiometer P4 on
the mother board. In case of not reaching this value, turn the potentiometer P4 clockwise (increasing) to
the maximum. Then reduce the amplification with turns counter-clockwise (decreasing) until AD-
transducer value is changing.

Microcontroller System
The temperature control circuit is a PID-controller, a software module in the microcontroller system. The PID-
controller works in the background all the time. Dependent on the cuvette adaptor the PID-controller uses
different parameters.
The required value of temperature is ascertained by the program of the microprocessor system. The
temperature is controlled according to the set temperature in the selected method.
The PID-controller gives information to the higher program about the given temperature tolerances and
therefore about the readiness for measuring.
Heating
The control variable of the controller has influence on the heating. A switched Darlington transistor in source of
power-switching is used as heating element. The PID-controller produces a voltage at the emitter of the
Darlington transistor by a pulse-width-modulation. This voltage controls the current through the transistor.
Involved components are the microcontroller system IC3 (8031PLCC), the OP-booster IC1A (LM324) and the
MOS-Fet T3 (BS170).
Cooling
For cooling or counter cooling during heating the microcontroller system switches on a Peltier element. There
are four possible levels with following terms and meaning:
0 = Cooling out
1 = Cooling (low level)
2 = Cooling (middle level)
3 = Cooling (high level)
In case of temperature changing by method selection cooling level 0 is selected if desired temperature is
higher as selected one.

operating. The temperature value is board-characteristic and cuvette
adaptor specific. The values for the temperature levels of 37 °C, 30 °C
or 25 °C are ascertained in factory and noted during the check of the
board. They are stored within the battery powered RAM.
Dependent on the used cuvette adaptor different service programs can
show and edit the values stored in battery powered RAM. The system
can be adjusted to lower or higher temperature values. A change of
about 25 counts means a temperature change of 0.1 °C.

Replacement of EPROM
1. Before start of replacement empty tube system absolutely by [WASH] and the wash lever.
2. Switch off the device and disconnect the mains cord.
3. Turn device on its top side.
4. Dismantle lid of EPROM.

6. Replace the EPROM by using an IC extractor. Notice that electrostatic discharge can destroy the
EPROM and the motherboard of Photometer 5010. Avoid to produce shortcuts on the pcb. Some

FLASH Memory
The FLASH memory IC14 (28F512) is a non-volatile memory which may be erased and programmed
electrically. This feature allows to replace the software of Photometer 5010 consisting of:
• operating system
• method data
• text data
The process is done by a serial interface utility DOWNLOAD ( see OM chapter 5.8) and a software modul
running on a personal computer. The devices Photometer 5010 and the personal computer have to be
connected by a serial interface cable.
1. Connect the devices Photometer 5010 and the personal computer by a serial interface cable.
2. Switch on both devices.
3. Install in a new directory of the personal computer the files DL_5010.EXE, D_LOAD.HEX and
P_100.HEX.
4. Photometer 5010: Start the serial interface utility DOWNLOAD ( see OM chapter 5.8) by [MODE]
[SERIAL] [5] [ENTER].
5. Personal computer: Change to DOS-input level. Select the directory with the installed three files.
Start the program file DL_5010.EXE.
DL_5010.EXE is a stand alone executable file for IBM/DOS systems.

Manual chapter 6.2). Notice that electrostatic discharge can destroy the FLASH MEMORY or

The photometer-cuvette temperature is measured in case of using the single cuvette adaptor in an
aluminium-dummy cuvette or in case of using the flow-through cuvette adaptor in the original flow-through
cuvette.
The resistance of the thermistor is measured by a 4½ digit digital multimeter (DMM). The requirements of
the DMM are given in chapter 7. The corresponding temperature is listed in chapter 8.
3. Operating conditions
The calibration should be performed under normal operating conditions, i.e.
• Room temperature: 15 °C to 30 °C
• Relative humidity: 40 % to 80 %
• No ventilation or draft
Note that the variation of the room temperature should not exceed ± 2 °C during the calibration process.
5. Test procedure
a. In case of using the single cuvette adaptor insert the dummy-cuvette instead of a square cell.
In case of using the flow-through adaptor remove the joint inlet tube cuvette / aspirate tubing and
the outlet tube cuvette.
b. Connect the thermistor lead to the two-wire resistance input of the DMM.
c. Insert the thermistor-probe into the hole of the photometer dummy-cuvette or the hole of the outlet
tube cuvette of the flow-through cuvette.
Caution: Handle the delicate probe with care! Pay attention to the correct position of the probe.
Avoid bubbles within the aperture of the flow-through cuvette.
d. Switch on the DMM and Photometer 5010 and wait 15 minutes for warming up.
e. Select the two-wire resistance function and appropriate range on the DMM. The resistance of the
thermistor is in the range of 6 kOhm up to 10 kOhm for the temperature range of 25 °C up to
37 °C. The table of resistance of thermistor ID 090-063 shows the assigned resistance for a given
temperature.
i. Select the temperature 25 °C by using service program item 1 – S8.1 - Temperature regulation
on / off. Activate the temperature control by selecting ON and 25 °C.
j. Check the temperature control by using service program item 5 – S8.5 - Temperature required /
actual.
As long as temperature is not stabilized, the character 16 of the first line is alternately showing a
“T” or a black rectangle. It needs minimum five minutes until a stabilized temperature level is
reached. Wait 10 minutes furthermore.
k. Read the displayed measurement of DMM in Ohm. Look up in the table of resistance the
appropriate set of values. Calculate by performing a linear interpolation the assigned temperature
value (see chapter 9).
l. Make an automatic temperature calibration by using service program item 4 – S8.4 - Temperature
calibration.
m. Check the temperature control by using service program item 5 – S8.5 - Temperature required /
actual.
As long as temperature is not stabilized, the character 16 of the first line is alternately showing a
“T” or a black rectangle. It needs minimum five minutes until a stabilized temperature level is
reached. Wait 5 minutes furthermore.
n. Read the displayed measurement of DMM in Ohm. Look up in the table of resistance the
appropriate set of values. The temperature value should show the desired value.
o. Read the required value (first value in the second line) and subtract the value 3000. The result is
called temperature value. Note this value in calibration protocol combined with adaptor type and
temperature level.

p. Repeat above mentioned steps with the temperature levels 25 °C, 30 °C and 37 °C for both types
of adaptors.
6. Documentation
For documentation use a form containing following table:
If the 24 months resistance-accuracy fulfils the above accuracy requirements, the latest
Calibration Certificate can be ≤ 24 months old.
If the resistance R is found in the table, get the appropriate temperature value and finish this chapter.
If the actual temperature of the thermistor is in the range of the required temperature ± 1.5 °C, two values
of resistance can be read from table with the relation:

The resistances R0 and R1 form an interval. They are a set of data points next to the resistance R.
The assigned temperature values of the resistances R0 and R1 are called T0 and T1.
The assigned temperature T can be calculated by the formula:
