• Used to mount water-miscible preparations • Used for dehydrated and cleared


• In cases where stain is removed or preparations in Xylene or Toluene
decolorized with alcohol or xylene • Recommended for majority of staining
• Made up of 3 Girl Scouts of the Philippines methods

1. WATER (low Refractice index)


─ Good only for temporary mounting
─ Does not allow tissue to be seen under oil immersion lens (OIO)

2. GLYCERIN (RF: 1.46)


─ Used as preservative;
─ Semi-permanent mounting medium that will keep the section monted for years especially if
sealed on edges with paraffin wax.
─ Ex. Include Glycerin jelly (standard mounting medium)

3. FARRANT’s MEDIUM
─ Take longer time to harden; REQUIRES RINGING

4. APATHY’s MEDIUM
─ Used for methylene blue stained; nerve preparations
─ DOES NOT REQUIRE RINGING

5. BRUN’S FLUID
─ Used for mounting FROZEN sections

1. CANADA ALBASAM (RF: 1.524)


─ Natural resin extracted from canadian tree / abus balsamea
─ Can be made neutral or acid by adding Calcium carbonate or salicylic acid

2. Dp PX (RF: 1.532)
─ Di-butyl – Phthalate – Poly- Styrene – Xylene
─ Used for small tissue sections

3. XAM (RF: 1.52)


─ Synthetic; dries out quickly without retraction

4. CLARITE (RF: 1.544)


─ Synthetic; soluble in xylene, generally preferred than DPX (2)

• RINGING
─ Sealing the margins of the coverslip to prevent escape of fluid
─ Ringing media:
a. KRONIG CEMENT – 2 parts paraffin; 4-9 parts powdered colophonium resin
b. DUROFIX

COMMON ADHESIVES

1. Mayer’s egg albumin - most commonly used, convenient, inexpensive, used in celloidin sections
2. Dried Albumin
3. Gelatin
4. Gelatin- formaledehyde mixture
5. Starch paste
6. Plasma – readily available from outdated blood from blood banks, dispensed at 0.5 mL each
7. Poly-L-Lysine – widely used in immunohistochemistry
8. APES – useful in cytology specifically cytospin preparations

Note: When using ALBUMIN; Should be filtered + addition of THYMOL CRYSTALS to prevent growth of
mold
