UNIT 1.

PREPARATION OF PIPETTE, REAGENTS

AIMS

 Calibration of a pipette
 Prepare a standard solution, 0.10 N NaOH
 Calculate the concentration of an unknown acid solution by using prepared 0.10 N NaOH
solution

LAB PRACTICES

1. Experiment 1: Calibration of a pipette

1.1 Materials and tools

 Analytical balance reading to 0.0001 g (0.1 mg).

 V mL glass (bulb) pipette
 100 mL beaker.
 Thermometer 0 to 100 oC, reading to 0.1 oC.
 Distilled or deionised water (about 200 mL) in a conical flask
 Paper towel or tissue

1.2 Protocols

 Step 1: Weight the empty beaker

o check beaker: it should be clean and dry,

o put the beaker on the scale pan of the analytical balance, close
windows of the balance and weight the empty beaker
o Read the the weight m1 (g). Write down m1(g) in your Lab note

 Step 2: Weight the beaker containing water

o Measure temperature of water using thermometer

o Using a V mL pipette that are needed to check. Check the pipette:
dry and clean
o Suck V mL of water in the pipette then transfer water to the beaker
o Close windows of the balance
o Read the weight of the beaker containing VmL water
o Write down m2 (g) in your Lab note

 Step 3: Check the specific weight of water at the measured temperature, d (g/mL), write down in
your lab notebook.

Repeat the step 1-2-3 three times.

 Step 4: Calculate the true volume of the pipette.
