• Seq A protein is a negative regulator of DNA initiation.
• SeqA selectively binds to origin (oriC) and inhibits
reinitiation of replication from OriC after the replication has
proceeded.
• The oriC contains 11 copies of GATC palindromic
sequences. When the OriC is fired, the daughter strand are
hemi-methylated.
 Over most of the chromosome, full methylation is restored
within a minute or two of replication . This brief period allows
for use of hemi-methylation as a guide for the mismatch
repair system . However, the origin of replication is slow to
regain full methylation; it takes 10 to 15 minutes.
