Stages of plant tissue culture

■ Media preparation
■ Collection of explant
■ Sterilization of explant
■ Initiation
■ Multiplication
■ Shooting
■ Rooting
■ Hardening
Explants:
■ Explants – are part of plants which has the ability to become the whole plant rapidly.
■ Examples:
Bamboo- nodes
Banana- meristematic region of bud
Pomegranate- nodes
Ginger- rhizome

Node (Explant)
Composition of the MS (Murashige and skoog) media:
Collection of explant, Sterilization and initiation
■ Cut the plant at the required regions
■ suspended the explants in 100 ml of sterile water containing 0.1 % streptomycin
(antibacterial agent) and carbendazim (antifungal agent)
■ Swirl the solution for about 20 min.
■ wash with fresh sterile water trice.
■ Add 2 drops of TW media (Tween 20) in 100 ml of sterile water inside laminar air flow
and add the explants in the solution and shack vigorously for 15 minutes.
■ Then wash the ex-plants with sterile water until the foam is removed.
■ Then implant the ex-plants inside the initiation media.
■ Then wrap the bottles and seal then store at controlled light and temperature for 4
weeks.
Multiplication:

■ Separate cluster of stems into individual plantlets.


■ Then inoculate into multiplication media.
■ Store it in controlled condition up to 4 weeks.
■ Repeat the same step for 8 times.
Shooting:
■ Remove the parts of shoots like leaves and cut in the middle of the plantlet.
■ And remove dead parts as well.
■ And inoculate in shooting media.

Rooting:
■ Remove the dead cells.
■ Then cut the roots without removing the basement.

Hardening - Primary Hardening
■ plantlets are taken out of the tissue culture bottle and planted in small containers containing
coco-peat.
■ These plants are placed in tunnel like shelters.

■ They need only 25% sunlight.


■ They are watered in the morning before 10 am and in the evening after 3 pm.
■ After one and half months these plants will be ready for secondary hardening.
Hardening - secondary hardening
■ plants are grown in plastic bags filled with coir pith (red soil: sand: coir in 1:1:1 ratio).

■ Coir pith acts as a manure (or compost) giving nutrients, while sand increases the percolation so that
water drains off quickly and also allows air to circulate.
■ This process is carried out in the green house.
■ After secondary hardening the plants are ready to be sold.
